Environmental fluoxetine promotes skin cell proliferation and wound healing

This study investigates the effects of environmentally-relevant concentrations of fluoxetine (FLX, commercial name: Prozac) on wound healing. Pollution of water systems with pharmaceutical and personal care products, including antidepressants such as FLX and other selective serotonin reuptake inhibitors, is a growing environmental concern. Environmentally-relevant FLX concentrations are known to impact physiological functions and behaviour of aquatic animals, however, the effects of exposure on humans are currently unknown. Using a combination of human skin biopsies and a human keratinocyte cell line, we show that exposure to environmental FLX promotes wound closure. We show dose-dependent increases in wound closure with FLX concentrations from 125 ng/l. Using several –omics and pharmaceutical approaches, we demonstrate that the mechanisms underlying enhanced wound closure are increased cell proliferation and serotonin signalling. Transcriptomic analysis revealed 350 differentially expressed genes after exposure. Downregulated genes were enriched in pathways related to mitochondrial function and metabolism, while upregulated genes were associated with cell proliferation and tissue morphogenesis. Kinase profiling showed altered phosphorylation of kinases linked to the MAPK pathway. Consistent with this, phosphoproteomic analyses identified 235 differentially phosphorylated proteins after exposure, with enriched GO terms related to cell cycle, division, and protein biosynthesis. Treatment of skin biopsies and keratinocytes with ketanserin, a serotonin receptor antagonist, reversed the increase in wound closure observed upon exposure. These findings collectively show that exposure to environmental FLX promotes wound healing through modulating serotonin signalling, gene expression and protein phosphorylation, leading to enhanced cell proliferation. Our results justify a transition from the study of behavioural effects of environmental FLX in aquatic animals to the investigation of effects of exposure on wound healing in aquatic and terrestrial animals, including direct impacts on human health. | QRB acknowledges a ‘Happy Chemical’ PhD studentship funded by the University of Hull. In the coming years, the use of microalgal biomass as agricultural biofertilizers has shown promising results. The use of wastewater as culture medium has resulted in the reduction of production costs, making microalgae-based fertilizers highly attractive for farmers. However, the occurrence of specific pollutants in wastewater, like pathogens, heavy metals and contaminants of emerging concern (CECs), such as pharmaceuticals and personal care products may pose a risk on human health. This study presents an holistic assessment of the production and use of microalgal biomass grown in municipal wastewater as biofertilizer in agriculture. Results showed that pathogens and heavy metals concentrations in the microalgal biomass were below the threshold established by the European regulation for fertilizing products, except for cadmium. Regarding CECs, 25 out of 29 compounds were found in wastewater. However, only three of them (hydrocinnamic acid, caffeine, and bisphenol A) were found in the microalgae biomass used as biofertilizer. Agronomic tests were performed for lettuce growth in greenhouse. Four treatments were studied, comparing the use of microalgae biofertilizer with a conventional mineral fertilizer, and also a combination of both of them. Results suggested that microalgae can help reducing the mineral nitrogen dose, since similar fresh shoot weights were obtained in the plants grown with the different assessed fertilizers. Lettuce samples revealed the presence of cadmium and CECs in all the treatments including both negative and positive controls, which suggests that their presence was not linked to the microalgae biomass. ABSTRACT: Plastic debris is a significant threat to marine and coastal ecosystems. Previous research found that waves, wind, as well as density, size, and shape of microplastics, drive their transport and dispersion. In this paper, a set of laboratory experiments on the effect of waves and wave-induced currents on the input rate and cross-shore transport and dispersion of different types of plastic debris, including the macro and mesosizes, in addition to microplastics is presented. 15 plastic-debris types characterized by different sizes, shapes, and densities, including facemasks, were analyzed under regular and irregular wave conditions. The results show that input and transport rates of plastics depend on their terminal velocities and wave steepness. Plastics with higher settling velocities under less-steep wave conditions are likely to escape coastal entrapment and end up in the breaking zone. This paper presents a novel method to select the optimal combination of grid resolution and number of Lagrangian elements (LEs) required in numerical modelling of oil concentrations at sea. A sensitivity analysis in terms of grid resolution and the number of LEs, was carried out to understand the uncertainty that these userdependent parameters introduce in the numerical results. A dataset of 211,200 simulations performed under 400 metocean patterns, 6 initial volumes, 11 grid resolutions, and different numbers of LEs (100 to 500,000), was used to analyze the sensitivity of the model along different Thresholds of Concern. Results show the importance of a correct selection of the number of LEs and the grid resolution in Lagrangian modelling of surface oil concentrations. Plastic floating on the ocean surface represents about 1 % of all plastic in the ocean, despite the buoyancy of most plastics. Biofouling can help to sink debris, which could explain this discrepancy. A set of laboratory experiments was conducted to investigate biofilm-induced effects on the buoyancy of different plastic debris. Ten materials of different densities (buoyant/non-buoyant), sizes (micro/meso/macro), and shapes (irregular/spherical/cylindrical/ flat), including facemasks and cotton swabs, were evaluated. Biofilm was incubated in these materials from a few weeks to three months to investigate the effect of different growth levels on their buoyancy. Biofilm levels and rising/settling velocities were measured and compared at seven time-points. The results show a hindered buoyancy for solid materials, while hollow and open materials showed the opposite trend in early biofilm colonization stages. A relationship was established between biofilm-growth and equivalent sphere diameter that can be used to improve predictive modeling of plastic-debris transport. | This work was funded by the European Regional Development Funds (Interreg Maritime IT FR program) under the SPlasH & Co project (contract number D35F21002010001). This study compares the role of salinity regime and chemical pollution in the biodiversity patterns of estuarine benthic communities. A specific field survey allowed us to explain the response of organisms to mixtures of chemicals and the effects of salinity regime and extreme events on the richness and composition of macroinvertebrate assemblages. The patterns obtained provide further evidence that both stress sources are key factors in macroinvertebrate communities' organization, but the type and magnitude of the changes differ. The abundance of opportunistic species increased according to the pollution gradient, while this indicator was less sensitive to salinity descriptors. In contrast, biotic indices responded to the salinity regime but did not show a consistent pattern in response to pollutants. Multivariate analyses reflected both environmental stress gradients. Exposure to environmental airborne manganese (Mn) can lead to neurotoxic disorders and cognitive deficits. The degree of exposure can be assessed by personal sampling of particulate matter (PM) or through biomarkers of exposure. The aim of this work was to characterise the personal exposure to airborne Mn and other trace metals by measuring their bioaccessibility in PM filters taken from personal samplers in an environmentally exposed adult population living in the vicinity of a ferromanganese alloy plant in Santander Bay (northern Spain). Concentrations of bioaccessible and non-bioaccessible Mn and other metals associated with coarse (PM10-2.5) and fine (PM2.5) modes were quantified from 24 h personal samplers in 130 participants divided into two groups according to their Mn exposure: highly (n = 65) and moderately (n = 65) exposed. Gastric fluid and artificial lysosomal fluid (ALF) were used in the bioaccessibility tests as surrogate agents for the body fluids that can come into contact with coarse and fine particles, respectively. The mean air Mn levels in PM10-2.5 and PM2.5 were 127.2 and 126.2 ng/m3, respectively, in the highly exposed group, and 18.6 and 31.7 ng/m3 in the moderately exposed group. The bioaccessibility (%) of Mn in gastric fluid and ALF was also found to be greater in the highly exposed group. The evaluation of the content of metals and metalloids in particulate matter (PM) and in atmospheric deposition in areas impacted by local industries is essential from an environmental and health risk perspective. In this study, the PM10 levels and atmospheric deposition fluxes of potentially toxic metals and metalloids were quantified at three urban sites of the Cantabrian region (northern Spain), located at different distances downwind of a Mn alloy plant. The content of Mn, V, Fe, Ni, Cu, Zn, As, Mo, Cd, Sb and Pb in PM10 and in the water-soluble and insoluble fractions of the deposition was determined by ICP-MS. Among the studied elements, the highest concentrations in PM10 and deposition rates were found for Mn, Fe, Zn and Pb, associated with the Mn alloy industry, and for Cu, related to non-exhaust traffic emissions. The levels of Mn, Fe, Zn and Pb in PM10 were higher in autumn, when the most frequent winds blow from the S-SW, whereas their highest deposition rates were found in winter and autumn, which are characterized by high monthly average precipitations. The water-soluble fraction of the atmospheric deposition of most metals increased with distance from the Mn alloy plant. The highest water-soluble fractions were found for Ni (72%), Zn (62%), Cu (60%) and Mn (49%). Air quality assessment regarding metals and metalloids using experimental measurements is expensive and time consuming due to the cost and time required for the analytical determination of the levels of these pollutants. According to the European Union (EU) Air Quality Framework Directive (Directive 2008/50/EC), other alternatives, such as objective estimation techniques, can be considered for ambient air quality assessment in zones and agglomerations where the level of pollutants is below a certain concentration value known as the lower assessment threshold. These conditions occur in urban areas in Cantabria (northern Spain). This work aims to estimate the levels of As, Cd, Ni and Pb in airborne PM10 at two urban sites in the Cantabria region (Castro Urdiales and Reinosa) using statistical models as objective estimation techniques. These models were developed based on three different approaches: partial least squares regression (PLSR), artificial neural networks (ANNs) and an alternative approach consisting of principal component analysis (PCA) coupled with ANNs (PCA-ANN). Additionally, these models were externally validated using previously unseen data. The results show that the models developed in this work based on PLSR and ANNs fulfil the EU uncertainty requirements for objective estimation techniques and provide an acceptable estimation of the mean values. The input of trace elements via atmospheric deposition towards industrial, urban, traffic, and rural areas is quite different and depends on the intensity of the anthropogenic activity. A comparative study between the element deposition fluxes in four sampling sites (industrial, urban, traffic, and rural) of the Cantabria region (northern Spain) has been performed. Sampling was carried out monthly using a bulk (funnel bottle) sampler. The trace elements, As, Cd, Cr, Cu, Mn, Mo, Ni, Pb, Ti, Zn, and V, were determined in the water soluble and insoluble fractions of bulk deposition samples. The element deposition fluxes at the rural, urban, and traffic sites followed a similar order (Zn>Mn>>Cu=Ti>Pb>V=Cr>Ni>>As=Mo>Cd). The most enriched elements were Cd, Zn, and Cu, while V, Ni, and Cr were less enriched. An extremely high deposition of Mn was found at the industrial site, leading to high enrichment factor values, resulting from the presence of a ferro-manganese/silicomanganese production plant in the vicinity of the sampling site. Important differences were found in the element solubilities in the studied sites; the element solubilities were higher at the traffic and rural sites, and lower at the urban and industrial sites. For all sites, Zn and Cd were the most soluble elements, whereas Cr and Ti were less soluble. The inter-site correlation coefficients for each element were calculated to assess the differences between the sites. The rural and traffic sites showed some similarities in the sources of trace elements; however, the sources of these elements at the industrial and rural sites were quite different.
