Understanding how essential and toxic elements are distributed in cereal grains is a key to improving the nutritional quality of cereal-based products. The main objective of this work was to characterize the distribution of Cd and of nutrients (notably Cu, Fe, Mn, P, S and Zn) in the durum wheat grain. Laser ablation inductively coupled mass spectrometry and synchrotron micro X-ray fluorescence were used for micro-scale mapping of Cd and nutrients. A dissection approach was used to quantitatively assess the distribution of Cd and nutrients among grain tissues. Micro X-ray absorption near-edge spectroscopy was used to identify the Cd chemical environment in the crease. Cadmium distribution was characterized by strong accumulation in the crease and by non-negligible dissemination in the endosperm. Inside the crease, Cd accumulated most in the pigment strand where it was mainly associated with sulfur ligands. High-resolution maps highlighted very specific accumulation areas of some nutrients in the germ, for instance Mo in the root cortex primordia and Cu in the scutellum. Cadmium loading into the grain appears to be highly restricted. In the grain, Cd co-localized with several nutrients, notably Mn and Zn, which challenges the idea of selectively removing Cd-enriched fractions by dedicated milling process.

International audience | Due to the dramatic quantity of plastic debris released into our environment, one of the biggest challenges of the next decades is to trace and quantify microplastics (MPs) in our environments, especially to better evaluate their capacity to transport other contaminants such as trace metals. In this study, trace elements (Fe, Cu, Zn, As, Cd, Sn, Sb, Pb, and U) were analyzed in the microplastic subsurface (200 μm) using laser ablation inductively coupled plasma mass spectrometry (LA-ICP-MS). Microplastics subjected to the marine environment were collected on beaches (Guadeloupe) exposed to the north Atlantic gyre. We established a strategy to discriminate sorbed contaminants from additives based on the metal concentration profiles in MP subsurface using qualitative and quantitative approaches. A spatiotemporal correlation of the sorption pattern was proposed to compare MPs in terms of relative exposure time and time-weighted average concentrations in the exposure media.

The quality of freshwater undoubtedly reflects the health of our surrounding environment, society, and economy, as these are supported by various freshwater ecosystems. Monitoring efforts have therefore been considered a vital means of ensuring the ecological health of freshwater environments. Nevertheless, most aquatic environmental monitoring strategies largely focus on bulk water sampling for analysis of physicochemical and key biological indicators, which for the most part do not consider pollution events that occur at any time between sampling events. Because benthic biofilms are ubiquitous in aquatic environments, pollution released during sporadic events may be absorbed by these biofilms, which can act as repositories of pollutants. The aim of this study was to assess whether benthic biofilm monitoring could provide an efficient way of properly characterizing the extent of pollution in aquatic environments. Here, bulk water and benthic biofilms were sampled from three Hong Kong streams having various pollution profiles, and subsequently compared via high-resolution microscopy, metagenomic analysis, and analytical chemistry. The results indicated that biofilms were, indeed, reservoirs of environmental pollutants, having different profiles compared with that of the corresponding bulk water samples. Moreover, the results also suggested that biofilms sampled in polluted areas were characterized by a higher species richness. While the analytical testing of benthic biofilms still needs further development, the integration of chemical-pollutant profiles and biofilm sequencing data in future studies may provide unique perspectives for understanding and identifying pollution-related biofilm biomarkers.

Natural and synthetic chemicals are essential to our daily lives, food supplies, health care, industries and safe sanitation. At the same time protecting marine ecosystems and seafood resources from the adverse effects of chemical contaminants remains an important issue. Since the 1970s, monitoring of persistent, bioaccumulative and toxic (PBT) chemicals using analytical chemistry has provided important spatial and temporal trend data in three important contexts; relating to human health protection from seafood contamination, addressing threats to marine top predators and finally providing essential evidence to better protect the biodiversity of commercial and non-commercial marine species. A number of regional conventions have led to controls on certain PBT chemicals over several years (termed ‘legacy contaminants’; e.g. cadmium, lindane, polycyclic aromatic hydrocarbons [PAHs] and polychlorinated biphenyls [PCBs]). Analytical chemistry plays a key role in evaluating to what extent such regulatory steps have been effective in leading to reduced emissions of these legacy contaminants into marine environments. In parallel, the application of biomarkers (e.g. DNA adducts, CYP1A-EROD, vitellogenin) and bioassays integrated with analytical chemistry has strengthened the evidence base to support an ecosystem approach to manage marine pollution problems. In recent years, however,the increased sensitivity of analytical chemistry, toxicity alerts and wider environmental awareness has led to a focus on emerging chemical contaminants (defined as chemicals that have been detected in the environment, but which are currently not included in regulatory monitoring programmes and whose fate and biological impacts are poorly understood). It is also known that natural chemicals (e.g. algal biotoxins) may also pose a threat to marine species and seafood quality. Hence complex mixtures of legacy contaminants, emerging chemicals and natural biotoxins in marine ecosystems represent important scientific, economic and health challenges. In order to meet these challenges and pursue cost-effective scientific approaches that can provide evidence necessary to support policy needs (e.g. the European Marine Strategy Framework Directive), it is widely recognised that there is a need to (i) provide marine exposure assessments for priority contaminants using a range of validated models, passive samplers and biomarkers; (ii) integrate chemical monitoring data with biological effects data across spatial and temporal scales (including quality controls); and (iii) strengthen the evidence base to understand the relationship between exposure to complex chemical mixtures, biological and ecological impacts through integrated approaches and molecular data (e.g. genomics, proteomics and metabolomics). Additionally, we support the widely held view that (iv) that rather than increasing the analytical chemistry monitoring of large number of emerging contaminants, it will be important to target analytical chemistry towards key groups of chemicals of concern using effects-directed analysis. It is also important to evaluate to what extent existing biomarkers and bioassays can address various classes of emerging chemicals using the adverse outcome pathway (AOP) approach now being developed by the Organization for Economic Cooperation and Development (OECD) with respect to human toxicology and ecotoxicology.

The determination of trace metals and organic contaminants in environmental samples, such as water, air, soil, and sediment, is until today a challenging process for the analytical chemistry. Metal-organic frameworks (MOFs) are novel porous nanomaterials that are composed of metal ions and an organic connector. These materials are gaining more and more attention due to their superior characteristics, such as high surface area, tunable pore size, mechanical and thermal stability, luminosity, and charge transfer ability between metals and ligands. Among the various applications of MOFs are gas storage, separation, catalysis, and drug delivery. Recently, MOFs have been successfully introduced in the field of sample preparation for analytical chemistry and they have been used for sample pretreatment of various matrices. This review focuses on the applications of MOFs as novel adsorbents for the extraction of organic compounds from environmental samples.

Combining bioassays and analytical chemistry screenings is a powerful approach to assess emerging organic micropollutants which are the main contributors to toxic potentials in complex mixtures of water matrices. The aim of this study was to assess the cytotoxic effect of the occurrence of emerging organic micropollutants discharged into river water through industrial wastewater and treated effluents. The cytotoxic effects of surface water, treated effluents, and industrial wastewater were assessed using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) assay. Then, organic micropollutants of various chemical groups were identified using a detailed non-target screening based on gas chromatography coupled with a mass spectrometry detector (GC/MS). A significant cytotoxic effect on human intestinal epithelial Caco-2 cells was observed for all the samples. Caco-2 cell viability decreased by 17.99, 33.77, and 24.54 % for surface water, treated wastewater, and industrial water, respectively. The organic chemical compounds responsible for this toxic potential were identified using non-target chemical screening. Statistical correlation between cytotoxicity and the presence of emerging contaminants revealed that the cytotoxic effect was mainly due (r ≥ 0.42) to the occurrence of cyclopentasiloxane, decamethyl and cyclohexasiloxane, dodecamethyl, D-limonene, and ergoline-8-methanol, 8,9-didehydro-6-methyl while cytotoxicity was highly negatively correlated (r ≤ −0.42) to 2-ethylhexyl salicylate, 3-isopropoxy-1,1,1,7,7,7-hexamethyl-3,5,5-tris(trimethyl siloxy)tetrasiloxane, 6-acetyl-1,1,2,4,4,7-hexamethyltetralin, and (3-aminopropyltriethoxysilane. Seventy-six other compounds detected by GC/MS showed no correlation to cytotoxicity.

For the routine determination of metals in environmental samples, we require microwave-assisted digestion methods that yield 'total' or 'near-total' recoveries while avoiding the use of HF acid. As inductively coupled plasma mass spectrometry (ICP-MS) is the method of detection, it is desirable to minimize the use of HCl to avoid spectral interferences caused by high Cl- concentrations. Using certified reference materials, we performed a series of modifications to the US EPA method 3051 which included: increasing the temperature and durations of microwave digestion, varying the ratio of sample mass to acid volume, and alterations to the compositions of the acid digestion mixture. The experiments were conducted using urban particulate matter (NIST-1648), coal fly ash (NBS-1633) and six CANMET certified reference materials (Till-2, Till-3, Till-4, LKSD-1, LKSD-2 and LKSD-4), in two laboratories (Health Canada and Environment Canada) using different microwave digestion systems and different ICP-MS instruments. Our modified microwave-assisted nitric acid digestion method improved recoveries for Pb, Zn, V, Fe and Cu approaching 'total' recoveries in the same matrices determined using X-ray fluorescence (XRF) and instrumental neutron activation analysis (INAA) as reported in the certificates of analysis. Recoveries for other elements such as Cr and Ni compared well with 'near-total' recoveries yielded by traditional (non-assisted) acid digestion methods.
