A miniaturized electrothermal array for rapid analysis of temperature preference behaviors in ecology and ecotoxicology

Due to technical limitations, there have been minimal studies performed on thermal preferences and thermotactic behaviors of aquatic ectotherm species commonly used in ecotoxicity testing. In this work, we demonstrate an innovative, purpose-built and miniaturized electrothermal array for rapid thermal preference behavioral tests. We applied the novel platform to define thermal preferences in multiple invertebrate and vertebrate species. Specifically, Dugesia notogaea (freshwater planarians), Chironomus tepperi (nonbiting midge larvae), Ostracoda (seed shrimp), Artemia franciscana (brine shrimp), Daphnia carinata (water flea), Austrochiltonia subtenuis (freshwater amphipod), Physa acuta (freshwater snail), Potamopyrgus antipodarum (New Zealand mud snail) and larval stage of Danio rerio (zebrafish) were tested. The Australian freshwater water fleas, amphipods, snail Physa acuta as well as zebrafish exhibited the most consistent preference to cool zones and clear avoidance of zones >27 °C out of nine species tested. Our results indicate the larval stage of zebrafish as the most responsive species highly suitable for prospective development of multidimensional behavioral test batteries. We also showcase preliminary data that environmentally relevant concentrations of pharmaceutical pollutants such as non-steroidal anti-inflammatory drug (NSAID) ibuprofen (9800 ng/L) and insecticide imidacloprid (4600 ng/L) but not anti-depressant venlafaxine (2200 ng/L) and (iv) anticonvulsant medications gabapentin (400 ng/L) can perturb thermal preference behavior of larval zebrafish. Collectively our results demonstrate the utility of simple and inexpensive thermoelectric technology in rapid exploration of thermal preference in diverse species of aquatic animals. We postulate that more broadly such technologies can also have added value in ecotoxicity testing of emerging contaminants.

Parental effects manifest as alterations in offspring phenotype resulting from the parental phenotype and/or parental environment. We evaluated the effects of parental diet quality and mating strategy on the toxicant tolerance of offspring in Biomphalaria glabrata snails. We raised snails either individually (self-fertilizing) or in groups of three (outcrossing) on a diet of uncooked lettuce, fish food, cooked lettuce, or cooked lettuce plus fish food. We then exposed their offspring to cadmium and malathion challenges. Cadmium tolerance varied with parental diet and was greater in the offspring of outcrossing snails than self-fertilizing snails. Malathion tolerance was not affected by parental diet but was greater in the offspring of outcrossing snails. These results indicate that offspring responses to stressors are heavily influenced by parental experience, but may depend on the specific stressor and the mechanism of action and/or detoxification.

Little is known about effects of maternally transferred contaminants in snakes. The purpose of this study was to evaluate sublethal effects of maternally transferred mercury (Hg) on neonatal northern watersnakes (Nerodia sipedon). We captured 31 gravid females along a historically Hg-contaminated river. Following birth, we measured litter Hg concentrations and assessed locomotor performance, foraging ability (i.e., number of prey eaten, latency to first strike, strike efficiency, and handling time), and learning (i.e., change in foraging measures over time) in their offspring (n = 609). Mercury concentrations in offspring negatively correlated with motivation to feed and strike efficiency. Over time, strike efficiency and latency to strike decreased for all snakes in the study. However, offspring from contaminated areas maintained consistently lower efficiencies than reference individuals. This study is the first to examine sublethal behavioral effects of maternally transferred contaminants in snakes and suggests that maternally transferred Hg negatively affects offspring behavior.

Total cadmium (Cd) concentrations in four ornithogenic coral-sand sedimentary profiles displayed a strong positive correlation with guano-derived phosphorus, but had no correlation with plant-originated organic matter in the top sediments. These results indicate that the total Cd distributions were predominantly controlled by guano input. Bioavailable Cd and zinc (Zn) had a greater input rate in the top sediments with respect to total Cd and total Zn, and a positive correlation with total organic carbon (TOC) derived from plant humus. Multi-regression analysis showed that the total Cd and TOC explained over 80% of the variation of bioavailable Cd, suggesting that both guano and plant inputs could significantly influence the distribution of bioavailable Cd, and that plant biocycling processes contribute more to the recent increase of bioavailable Cd. A pollution assessment indicates that the Yongle archipelago is moderately to strongly polluted with guano-derived Cd.

The feasibility of assessing copper accumulation in agricultural soils using avoidance tests with a Canadian strain of Folsomia candida was investigated under laboratory conditions. The avoidance response to nominal copper sulfate concentrations of 0, 200, 800, 1600 and 3200 mg kg⁻¹ in OECD soil was inconsistent between trials with the standard plastic cup or a modified Petri dish method requiring less soil. However, combined results from three Petri dish trials decreased variability and provided a 75% avoidance level, close to the 80% criterion proposed for avoidance tests. A Copper avoidance EC₅₀ₛ of 18 mg kg⁻¹was obtained using the Petri dish method whether tests were conducted with or without light. While Petri dish tests have potential as a cheap tool to distinguish metal contaminated soils from uncontaminated soils they would be unsuitable for tracking or quantifying changes in metal concentrations. throughout remediation. The effect of different sewage concentrations (0, 20, 60 and 100%), vegetation (Bare, Avicennia marina or Rhizophora mucronata) and immersion periods (immersion/emersion period of 12/12 h or 3/3 days just for 100%) conditions were studied for 6 months on survival and growth rates of Terebralia palustris (Linnaeus, 1767). Gastropods' activity and ecosystem engineering preformed at bare and A. marina planted cells and 3 sewage conditions (0, 20 and 60%) were determined. Survival rates were higher than 70% in all treatments. Growth rate decreased significantly with increasing sewage concentrations (mainly at unplanted conditions) and longer immersion periods. A complete shift (from immersion to emersion periods) and a significant decrease in mobility and consequently its engineer potential, due to sewage contamination, lead to a 3–4 fold decrease in the amount of sediment disturbed. Sewage contamination, primary producers' abundance and environmental conditions may have influenced the gastropods survival, growth and its ecosystem engineering potential. The São Domingos mine (Portugal) is, potentially, a good site for ecotoxicological studies, due to a pH and metal gradient of acid mine drainage. In this study, the toxicity of several mine sediments was evaluated using the aquatic oligochaete Lumbriculus variegatus as a test organism. Our hypothesis was that exposure to contaminated sediments would cause behavioural early warning responses in L. variegatus. Five sites, with pH ranging from 2.5 to 6.5, and with associated metals, were investigated. The results showed poor sediment quality in most of the collected sediments and Fe, S and As were the dominant elements in the samples. High mortalities were observed, ranging from 32.6 to 100%, indicating severe contamination. The collected sediments did not support good L. variegatus growth and significantly changed its behaviour. Early warning responses consisted of decreased locomotion and decreased peristaltic movements. A behaviour inhibition will affect the ecosystem balance by limiting the organisms' ability to avoid capture, which leads to a higher risk of predation. Behavioural responses of the aquatic oligochaeta Lumbriculus variegatus may be used to detect early warning responses.

There is currently a lack of ecotoxicity tests adapted to earthworm species of higher ecological relevance and whose endpoints could be directly related to their ecological role in the soil. We propose a new and relatively simple ecotoxicity test based on the estimation of cast production (CP) by Lumbricus terrestris under laboratory conditions. CP was found to be linearly correlated to earthworm biomass and to be greatly influenced by soil water content. Azinphos-methyl had no effect on CP at all the concentrations tested. Significant decreases were observed at the normal application rate for other pesticides with (imidacloprid, carbaryl, methomyl) or without (ethyl-parathion and chlorpyrifos-ethyl) a clear concentration–effect response. For the highest concentration tested, reduction in CP varied between 35 and 67%. CP is straightforward and rapidly measured and ecologically meaningful. We thus believe it to be of great use as an endpoint in ecotoxicity testing. Cast production of Lumbricus terrestris is affected by pesticides under laboratory conditions.

Epifaunal communities associated with macroalgae were exposed to storm water pulses using a custom made irrigation system. Treatments included Millipore® freshwater, freshwater spiked with trace metals and seawater controls to allow for the relative importance of freshwater inundation, trace metals and increased flow to be determined. Experimental pulses created conditions similar to those that occur following real storm water events. Brief storm water pulses reduced the abundance of amphipods and gastropods. Freshwater was the causative agent as there were no additional effects of trace metals on the assemblages. Laboratory assays indicated that neither direct nor latent mortality was likely following experimental pulses and that epifauna readily avoid storm water. Indirect effects upon epifauna through salinity-induced changes to algal habitats were not found in field recolonisation experiments. Results demonstrate the importance of examining the effects of pulsed contaminants under realistic exposure conditions and the need to consider ecologically relevant endpoints. Brief storm water pulses trigger avoidance response in mobile epifauna due to the inundation of freshwater.

Although many studies have focused on trace metals accumulation, investigations of talitrid amphipods as biomarkers are rare. This study explores the possibility of using the solar orientation capacity of Talitrus saltator as a behavioural marker of exposure to two essential (Cu and Zn) and two non-essential (Cd and Hg) metals. LC50 analyses performed before the solar orientation tests showed that the 72 h LC50 for Hg was 0.02 ppm while the 96 h LC50 values for Cu, Cd and Zn were 13.28 ppm, 27.66 ppm, and 62.74 ppm, respectively. The presence of metals in seawater affects the solar orientation capacity of T. saltator in a concentration-dependent manner and according to the toxicity ranking of the metals (Hg > Cu > Cd > Zn). Therefore, the solar orientation capacity of T. saltator seems to be a promising behavioural marker for exposure to trace metals. Solar orientation capacity is a promising behavioural marker for exposure to trace metals in sandhoppers.
