Arsenic (As) contamination and bioaccumulation are a serious threat to agricultural plants. To address this issue, we checked the efficacy of As tolerant plant growth promoting bacteria (PGPB), zinc oxide nanoparticles (ZnO NPs) and oxalic acid (OA) in Luffa acutangula grown on As rich soil. The selected most As tolerant PGPB i.e Providencia vermicola exhibited plant growth promoting features i.e solubilzation of phosphate, potassium and siderophores production. Innovatively, we observed the synergistic effects of P. vermicola, ZnO NPs (10 ppm) and OA (100 ppm) in L. acutangula grown on As enriched soil (150 ppm). Our treatments both as alone and in combination alleviated As toxicity exhibited by better plant growth and metabolism. Results revealed significantly enhanced photosynthetic pigments, proline, relative water content, total sugars, proteins and indole acetic acid along with As amelioration in L. acutangula. Furthermore, upregulated plant resistance was manifested with marked reduction in the lipid peroxidation and electrolyte leakage and pronounced antagonism of As and zinc content in leaves under toxic conditions. These treatments also improved level of nutrients, abscisic acid and antioxidants to mitigate As toxicity. This marked improvement in plants’ defense mechanism of treated plants under As stress is confirmed by less damaged leaves cell structures observed through the scanning electron micrographs. We also found substantial decrease in the As bioaccumulation in the L. acutangula shoots and roots by 40 and 58% respectively under the co-application of P. vermicola, ZnO NPs and OA in comparison with control. Moreover, the better activity of soil phosphatase and invertase was assessed under the effect of our application. These results cast a new light on the application of P. vermicola, ZnO NPs and OA in both separate and combined form as a feasible and ecofriendly tool to alleviate As stress in L. acutangula.

Soil Cd and Zn contamination has become a serious environmental problem. This work explored the performance of wood vinegar (WV) in enhancing the phytoextraction of Cd/Zn by hyperaccumulator Sedum alfredii Hance. Rhizosphere chemical properties, enzyme activities and bacterial community were analyzed to determine the mechanisms of metal accumulation in this process. Results demonstrated that, after 120 days growth, different times dilution of WV increased the shoot biomass of S. alfredii by 85.2%–148%. In addition, WV application significantly increased soil available Cd and Zn by lowing soil pH, which facilitated plant uptake. The optimal Cd and Zn phytoextraction occurred from the 100 times diluted WV (D100), which increased the Cd and Zn extraction by 188% and 164%, compared to CK. The 100 and 50 times diluted WV significantly increased soil total and available carbon, nitrogen and phosphorus, and enhancing enzyme activities of urease, acid phosphatase, invertase and protease by 10.1–21.4%, 29.1–42.7%,12.2–38.3% and 26.8–85.7%, respectively, compared to CK. High-throughput sequencing revealed that the D 100 significantly increased the bacterial diversity compared to CK. Soil bacterial compositions at phylum, family and genera level were changed by WV addition. Compared to CK, WV application increased the relative abundances of genus with plant growth promotion and metal mobilization function such as, Bacillus, Gemmatimonas, Streptomyces, Sphingomonas and Polycyclovorans, which was positively correlated to biomass, Cd/Zn concentrations and extractions by S. alfredii. Structural equation modeling analysis showed that, soil chemical properties, enzyme activities and bacterial abundance directly or indirectly contributed to the biomass promotion, Cd, and Zn extraction by S. alfredii. To sum up, WV improved phytoextraction efficiency by enhancing plant growth, Cd and Zn extraction and increasing soil nutrients, enzyme activities, and modifying bacterial community.

Atrazine has been used on Chinese farmlands for a long time and over a wide range. The concentration of atrazine (1.86–1100 mg kg⁻¹) has exceeded the allowable limit in the soil (1.0 mg kg⁻¹), and concern is increasing about the potential harm to farmland soil. Four treatments (AT₀, AT₆, AT₁₀, AT₁₆) were established to reveal the effects of the long-term application of atrazine on soil health. The results showed a nonlinear regulation of the atrazine residue concentrations in the four treatments. The highest concentration of atrazine residue was in AT₆, at 167 mg kg⁻¹, and the lowest concentration of atrazine residue was in AT₁₆, at 102 mg kg⁻¹, but there was no significant difference between AT₁₀ and AT₁₆. The soil urease activity decreased significantly with the increase in the years of atrazine application, the saccharase and cellulase activities in the AT₆ were significantly higher than those observed in the other three treatments, the catalase activity gradually decreased with the increase in atrazine application years, and the activity in AT₆ was significantly higher than that in AT₁₆. A total of 238 genera were identified by Illumina MiSeq sequencing, and 28 dominant genera were screened. Atrazine significantly increased the relative abundance of Actinobacteria and contributed to the relative abundance of Rubrobacter, Blastococcus, Promicromonospora, Jiangella, Psychroglaciecola and Acetobacteraceae_uncultured, which exhibited significantly higher abundance in AT₁₆ than in AT₀. Although there were atrazine-degrading bacteria in the soil, and the atrazine residue decreased with the increase in application years, the concentration of the atrazine residue was still nearly 100 times higher than the allowable limit in the soil, which is a great threat to the soil health.

Concerns regarding microplastic contamination have spread from aquatic environments to terrestrial systems with a growing number of studies have been reported. Notwithstanding, the potential effects on soil ecosystems remain largely unexplored. In this study, the effects of polyethylene microplastics on soil enzymatic activities and the bacterial community were evaluated, and the microbiota colonizing on microplastics were also investigated. Microplastic amendment (2000 fragments per kg soil) significantly increased the urease and catalase activities in soil after 15 days, and no discernible alteration of invertase activities was detected. Results from high-throughput sequencing of 16S rRNA revealed that the alpha diversities (richness, evenness, and diversity) of the microbiota in soil were not obviously changed by the PE amendment, whereas the diversity indexes of microbiota on plastic fragments were significantly lower than those in the control and amended soils. Different taxonomic composition was observed in between the control and amended soils after 90 days of incubation. Bacterial assemblages with distinct community structure colonized the PE microplastics. Additionally, several taxa including plastic-degrading bacteria and pathogens were more abundant on microplastics. Simultaneously, the predicted functional profiles showed that the pathways of amino acid metabolism and xenobiotics biodegradation and metabolism were higher on the microplastics. These results indicated that microplastics in soil, compared with those in aquatic environments, can also act as a distinct microbial habitat, potentially altering the ecological functions of soil ecosystems.

In this study, the tips of Myriophyllum aquaticum (M. aquaticum) plants were planted in open-top plastic bins and treated by simulated wastewater with various ammonium-N concentrations for three weeks. The contents of related carbohydrates and key enzyme activities of carbon metabolism were measured, and the mechanisms of carbon metabolism regulation of the ammonia tolerant plant M. aquaticum under different ammonium-N levels were investigated. The decrease in total nonstructural carbohydrates, soluble sugars, sucrose, fructose, reducing sugar and starch content of M. aquaticum were induced after treatment with ammonium-N during the entire stress process. This finding showed that M. aquaticum consumed a lot of carbohydrates to provide energy during the detoxification process of ammonia nitrogen. Moreover, ammonia-N treatment led to the increase in the activitives of invertase (INV) and sucrose synthase (SS), which contributed to breaking down more sucrose to provide substance and energy for plant cells. Meanwhile, the sucrose phosphate synthase (SPS) activity was also enhanced under stress of high concentrations of ammonium-N, especially on day 21. The result indicated that under high-concentration ammonium-N stress, SPS activity can be significantly stimulated by regulating carbon metabolism of M. aquaticum, thereby accumulating sucrose in the plant body. Taken together, M. aquaticum can regulate the transformation of related carbohydrates in vivo by highly efficient expression of INV, SPS and SS, and effectively regulate the osmotic potential, thereby delaying the toxicity of ammonia nitrogen and improving the resistance to stress. It is very important to study carbon metabolism under ammonia stress to understand the ammonia nitrogen tolerance mechanism of M. aquaticum.

Natural aging alters the surface physicochemical properties of biochars, which can affect the retention of heavy metals. This work investigated the effect of biochar aging on stabilization of heavy metals (Cd and Ni) and soil enzyme activities simulated with laboratory wet–dry (WD) and freeze–thaw (FT) cycling. A wheat straw (WS) biochar and a corn straw (CS) biochar were subjected to 30 WD or FT cycles, and Cd- and Ni-contaminated alkaline soils amended with the two fresh biochars (at 5% w/w) were subjected to 30-day constant moisture incubation and 30 WD or FT cycles. WD and FT aging caused slight reduction in the pH of the biochars, significant increases in their O contents and surface areas, and formation of new carbonate minerals. WS biochar was more effective than CS biochar at reducing the phytoavailable Cd in the soil, with reduction of 12.1%, 14.6%, and 12.9% under constant moisture incubation, WD aging, and FT aging, respectively. Reduction in phytoavailability of Ni by the addition of biochars was observed only under WD aging, by 17.0% and 18.5% in the presence of WS and CS biochars, respectively. Biochar amendment also reduced the distribution of Cd in the acid soluble and reducible fractions in all aging regimes. The addition of biochars decreased catalase activity in almost all aging regimes and invertase activity under FT aging, but increased urease activity under FT aging. Comparison of the enzyme activities in the soils amended with biochars under constant moisture and accelerated aging conditions indicates WD aging significantly decreased the activities of catalase, invertase, and urease in all treatments, while FT aging significantly increased urease activity in all treatments. These findings suggest that biochars can stabilize Cd in alkaline soils under changing environmental conditions, although the activities of some soil enzymes could be negatively impacted.

Phytoextraction using Celosia argentea Linn. can potentially decontaminate Cd-contaminated soils. However, most earlier studies have been conducted at laboratory scale and for a relatively short remediation period. To evaluate the phytoextraction efficiency of C. argentea combined with different soil amendments (ammonium chloride, Bacillus megaterium, and citric acid), an 18-month field experiment was carried out in a farmland soil contaminated with 3.68 mg kg⁻¹ Cd by mine tailings in southern China. Soil Cd concentrations were decreased by 6.34 ± 0.73% after the three harvestings (with no amendments), which was 2.27 times that of the no-planting control (p < 0.05). Application of ammonium chloride, B. megaterium, and citric acid increased the overall Cd reduction rate in soil by 40.5%, 46.1%, and 105%, respectively. The application of citric acid decreased total Cd in soil by up to 16.9% in the rhizosphere soil and 13.0% in the bulk soil. The highest annual shoot biomass yield and Cd extraction amount reached 8.79 t ha⁻¹ and 273 g ha⁻¹. Acid-soluble Cd fraction in the rhizosphere was significantly lower compared to that in the bulk soil (p < 0.05), which indicates that mobile Cd in the rhizosphere was taken up by the roots vastly. C. argentea phytoextraction also improved soil metabolic functions by increasing the activities of soil enzymes (urease, invertase, phosphatase, and catalase). These findings demonstrate that Cd phytoextraction using C. argentea with the application of soil amendments can greatly improve the quality of Cd-contaminated soils.

Fomesafen, a long-lived protoporphyrinogen-oxidase inhibitor, specially developed for post-emergence control of broad-leaf weeds, is used widely in soybean fields in northern China (Dayan and Duke, 2010). The impact of fomesafen on microbial communities in rhizosphere soils, however, is unknown. In this study we examined fomesafen degradation as well as its effects in the rhizosphere of soybean plants grown in a greenhouse. Fomesafen had shorter half-life in rhizosphere soil than previously reported for bulk soil from the same location (87 vs 120 days). The enzyme activity of soil extracts and the microbial community composition of 16S rRNA genes (16S) amplified from soil DNA were also investigated. Although not immediately apparent, both the high (37.5 mg kg⁻¹) and low (18.75 mg kg⁻¹) doses of fomesafen significantly decreased urease and invertase activities in the rhizosphere soil from days 30 and 45 respectively until the end of the experiment (90 days). Analysis of 16S amplicons demonstrated that fomesafen had a dose dependent effect, decreasing alpha diversity and altering beta diversity. Significant phylum level decreases were observed in five of the ten phyla that were most abundant in the control. Proteobacteria was the only phylum whose relative abundance increased in the presence of fomesafen, driven by increases in the genera Methylophilacaea, Dyella, and Sphingomonas. The functional implications of changes in 16S abundance as predicted using PICRUSt suggested that fomesafen enriched for enzymes involved in xenobiotic metabolism and detoxification (cytochrome P450s and glutathione metabolism). Our data suggest that, despite being degraded more rapidly in the rhizosphere than in bulk soil, fomesafen had long-lasting functional impacts on the soil microbial community.

Nutrient loading is a leading cause of global seagrass decline, triggering shifts from seagrass- to macroalgal-dominance. Within seagrass meadows of Xincun Bay (South China Sea), we found that nutrient loading (due to fish farming) increased sediment microbial biomass and extracellular enzyme activity associated with carbon cycling (polyphenol oxidase, invertase and cellulase), with a corresponding decrease in percent sediment organic carbon (SOC), suggesting that nutrients primed microorganism and stimulated SOC remineralization. Surpisingly, however, the relative contribution of seagrass-derived carbon to bacteria (δ13Cbacteria) increased with nutrient loading, despite popular theory being that microbes switch to consuming macroalgae which are assumed to provide a more labile carbon source. Organic carbon sources of fungi were unaffected by nutrient loading. Overall, this study suggests that nutrient loading changes the relative contribution of seagrass and algal sources to SOC pools, boosting sediment microbial biomass and extracellular enzyme activity, thereby possibly changing seagrass blue carbon.

Seagrass meadows are experiencing worldwide declines mainly because of nutrient enrichment, which always result in macroalgae bloom and consequently periodic collapse and decomposition. However, effects of macroalgae decay on the sediment organic carbon (SOC) sequestration capacity remain unknown. Depending on the macroalgae biomass in eutrophic seagrass meadows of South China Sea, we carried out a laboratory chamber experiment to investigate the sediment labile organic carbon (OC) compositions and the influencing SOC transformation enzyme activity variations of seagrass meadows in response to common macroalgae bloom species (Cladophora spp.) decomposition. Although the dehydrogenase and β-glucosidase activities were not affected by macroalgae decomposition, the macroalgae decomposition significantly elevated the salt-extractable carbon (SEC) content, SEC/SOC, levels of invertase and polyphenol oxidase activities, and the CO₂ release. Overall, this study indicates that macroalgae decomposition stimulates the SOC transformation, and therefore, it is not benefit for SOC sequestration within seagrass meadows of the South China Sea.
