Melatonin (M) is a pleiotropic molecule that improves plant growth and increases heavy metal tolerance. The role of M for improving plant growth and tolerance under cadmium (Cd) stress, and mitigation of Cd-induced toxicity has not yet been sufficiently examined. Therefore, here we conducted a glasshouse experiment to explore the influence of various M dosages on Cd detoxification and stress-tolerance responses of Brassica napus under high Cd content (30 mg kg⁻¹). The effects of M on the modulation of Cd tolerance in B. napus plants have been investigated using various growth attributes, Cd accumulation and tolerance indices, and secondary metabolic parameters. We found that Cd stress inhibited root growth (by 11.9%) as well as triggered reactive oxygen species accumulation (by 31.2%) and MDA levels (by 18.7%); however, exogenous M substantially alleviated the adverse effect of oxidative stress by decreasing levels of H₂O₂ (by 38.7%), MDA (by 13.8%) and EL (by 1.8%) in the Cd-stressed plants, as compared to the M-untreated plants (control). Interestingly, exogenous M reduced Cd accumulation in roots (∼48.2–58.3-fold), stem (∼2.9–5.0-fold) and leaves (∼4.7–6.6-fold) compared to control plants, which might be due to an M-induced defense and/or detoxification response involving a battery of antioxidants. Overall, addition of the exogenous M to the Cd-stressed plants profoundly enhanced Cd tolerance in B. napus relative to control plants. These results suggested the biostimulatory role (at the physiological and molecular level) of M in improving growth, Cd tolerance, and Cd detoxification in B. napus, which indicate the potentiality of M for green remediation of Cd contaminated soils. This green trial would provide a reference for producing renewable bioenergy crops under Cd stress in contaminated soils. However, these recommendations should be verified under field conditions and the potential mechanisms for the interaction between Cd and M should be explicitly explored.

Grape pomace (GP) is a low-value by-product that contains a significant amount of high value-added products. The huge amount of non-edible residues of GP wastes (seeds, skins, leaves and, stems) produced by wine industries causes’ environmental pollution, management issues as well as economic loss. Studies over the past 15–20 years revealed that GP could serve as a potential source for valuable bioactive compounds like antioxidants, bioactive, nutraceuticals, single-cell protein, and volatile organic compounds with an increasing scientific interest in their beneficial effects on human and animal health. However, the selection of appropriate techniques for the extraction of these compounds without compromising the stability of the extracted products is still a challenging task for the researcher. Based on the current scenario, the review mainly summarizes the novel applications of winery wastes in many sectors such as agriculture, pharmaceuticals, cosmetics, livestock fields, and also the bio-energy recovery system. We also summarize the existing information/knowledge on several green technologies for the recovery of value-added by-products. For the promotion of many emerging technologies, the entrepreneur should be aware of the opportunities/techniques for the development of high-quality value-added products. Thus, this review presents systematic information on value-added by-products that are used for societal benefits concerning the potential for human health and a sustainable environment.

A comprehensive study of the removal of selected biologically active compounds (pharmaceuticals and pesticides) from different water types was conducted using bare TiO₂ nanoparticles and TiO₂/polyaniline (TP-50, TP-100, and TP-150) nanocomposite powders. In order to investigate how molecular structure of the substrate influences the rate of its removal, we compared degradation efficiency of the initial substrates and degree of mineralization for the active components of pharmaceuticals (propranolol, and amitriptyline) and pesticides (sulcotrione, and clomazone) in double distilled (DDW) and environmental waters. The results indicate that the efficiency of photocatalytic degradation of propranolol and amitriptyline was higher in environmental waters: rivers (Danube, Tisa, and Begej) and lakes (Moharač, and Sot) in comparison with DDW. On the contrary, degradation efficacy of sulcotrione and clomazone was lower in environmental waters. Further, of the all catalysts applied, bare TiO₂ and TP-100 were found to be most effective in the mineralization of propranolol and amitriptyline, respectively, while TP-150 appeared to be the most efficient in terms of sulcotrione and clomazone mineralization. Also, there was no significant toxicity observed after the irradiation of pharmaceuticals or pesticides solutions using appropriate catalysts on rat hepatoma (H-4-II-E), mouse neuroblastoma (Neuro-2a), human colon adenocarcinoma (HT-29), and human fetal lung (MRC-5) cell lines. Subsequently, detection and identification of the formed intermediates in the case of sulcotrione photocatalytic degradation using bare TiO₂ and TP-150 showed slightly different pathways of degradation. Furthermore, tentative pathways of sulcotrione photocatalytic degradation were proposed and discussed.

Isoprostanes (IsoPs) are a class of oxidation products naturally formed in vivo that are indicative of endogenous oxidative stress. In individuals with chronic and oxidative stress related diseases, IsoPs are increased to pathological levels. Since they are excreted through urine into sewage systems, IsoPs can be detected in wastewater treatment plants' (WWTPs) effluents and thus can be used to evaluate the health status of a given population. The underlying principle is that higher isoprostanes WWTPs’ levels correspond to populations undergoing higher levels of oxidative stress, and thus disease. However, IsoPs are not eliminated by WWTPs and will end up being released into the aquatic environment, where they will be available for uptake by aquatic species. Being bioactive molecules, it has been suggested that IsoPs in the environment may elicit oxidative stress in aquatic organisms. In this context, we have critically reviewed the available data on IsoPs as products and effectors of toxicity, and propose the new concept of “circular toxicity”. In general, IsoPs excreted by humans as a consequence of oxidative stress are released into the aquatic environment where they may interact with aquatic organisms and induce the production of more IsoPs. These stress markers, in turn, will also be excreted, increasing the already high levels of stressors in the aquatic environment and thus create an escalating cycle of oxidative stress.

We utilized volcanic CO₂ vents at Castello Aragonese off Ischia Island as a natural laboratory to investigate the effect of lowered pH/elevated CO₂ on the bioactivities of extracts from fleshy brown algae Sargassum vulgare C. Agardh. We analysed the carbohydrate levels, antioxidant capacity, antibacterial, antifungal, antiprotozoal, anticancer properties and antimutagenic potential of the algae growing at the acidified site (pH ∼ 6.7) and those of algae growing at the nearby control site Lacco Ameno (pH∼8.1). The results of the present study show that the levels of polysaccharides fucoidan and alginate were higher in the algal population at acidified site. In general, extracts for the algal population from the acidified site showed a higher antioxidant capacity, antilipidperoxidation, antibacterial, antifungal, antiprotozoal, anticancer activities and antimutagenic potential compared to the control population. The increased bioactivity in acidified population could be due to elevated levels of bioactive compounds of algae and/or associated microbial communities. In this snapshot study, we performed bioactivity assays but did not characterize the chemistry and source of presumptive bioactive compounds. Nevertheless, the observed improvement in the medicinal properties of S. vulgare in the acidified oceans provides a promising basis for future marine drug discovery.

Due to the public and environmental health impact of cyanotoxins, investigations have been focused on finding environmental friendly algaecides from aquatic plants. The present study had the objective to evaluate the population control and physiological response of Microcystis aeruginosa (Kützing) Kützing (strain BCCUSP232) exposed to Pistia stratiotes L. extracts. Aqueous and ethanolic extracts of P. stratiotes at different concentrations (10, 25, and 50 mg L⁻¹) were submitted to M. aeruginosa and reduced significantly (p<0.05) the cyanobacterium cell density. The ethanolic extract presented the greatest growth inhibition of the strain at the highest concentration. During exposure to P. stratiotes extracts, intracellular hydrogen peroxide levels, malondialdehyde content, and antioxidant enzymes (peroxidase, catalase, and glutathione S-transferase) activities increased in M. aeruginosa, while total protein concentration decreased when compared to the control group. Superoxide dismutase (SOD) activities presented a sharp decline, suggesting superoxide radical and peroxide accumulation. This implied that SOD was a target for bioactive substance(s) from aqueous and ethanolic extracts of P. stratiotes. Phytochemical screening of the extracts revealed that the ethanolic extract presented 93.36 mg gallic acid equivalent (GAE) per gram dry weight (g⁻¹ DW) total polyphenols and 217.33 mg rutin equivalent (RE) per gram dry weight total flavonoids, and for the aqueous extract, 5.19 mg GAE g⁻¹ DW total polyphenols and 11.02 mg RE g⁻¹ DW total flavonoids were detected. Gas chromatography (GC)/mass spectrometry (MS) analyses of the ethanolic and aqueous extracts presented palmitic acid ethyl ester as major allelochemical. In view of these results, it can be concluded that P. stratiotes showed potential in controlling M. aeruginosa populations.

Global deterioration of water, air, and soil quality by the release of toxic chemicals from anthropogenic pollutants is becoming a serious global problem. The extensive use of copper oxide nanoparticles (CuO NPs) can be environmentally hazardous when these NPs enter the atmosphere. The present study aimed to evaluate the role of CuO NPs on plant growth, photosynthetic capacity, and bioactive compounds, as well as their transcriptional level changes in Brassica rapa seedlings. Chlorophyll, carotenoid, and sugar content decreased, while proline and anthocyanins were significantly enhanced in the CuO NP-treated seedlings compared with the untreated controls. Reactive oxygen species (ROS), malondialdehyde (MDA), and hydrogen peroxide (H₂O₂) production were also enhanced in the seedlings exposed to CuO NPs, which could have caused DNA damage that was detected by a DNA laddering assay. The glucosinolate (GSL) and phenolic compound content were significantly increased in CuO NP-treated seedlings compared with that in control seedlings. Transcriptional variation of genes associated with oxidative stress (CAT, POD, and GST), R2R3-type MYB involved in GSL (BrMYB28, BrMYB29, BrMYB34, and BrMYB51), and phenolic compounds (ANS, PAP1, PAL, and FLS) biosynthesis was analyzed using real-time polymerase chain reaction. Significant upregulation of CAT, POD, GST, BrMYB28, BrMYB29, BrMYB34, BrMYB51, ANS, PAP1, PAL, and FLS genes was observed in seedlings exposed to different concentrations of CuO NPs relative to the untreated seedlings. Therefore, we suggest that the use of CuO NPs could stimulate the toxic effects and enhance phytochemicals (i.e., glucosinolates and phenolic compounds) in B. rapa.

Improving digestibility, fermentation characteristics, and reducing greenhouse biogases to protect the environment without the use of synthetic materials is an important goal of modern-day farming and nutritionist. Plant extracts are capable of solving these. This is due to the digestive enzymes and the bioactive components capable of performing antimicrobial functions inherent in these plants. This study was aimed to investigate the effect of standard maize substrate treated with selected herbs and spices extracts on ruminal environmental biogas production and pressure during fermentation via biogas production technique. Herbs (Azadirachta indica leaves (T1), Moringa oleifera leaves (T2), Ocimum gratissimum leaves (T3) and spices (Allium sativum bulb (T4), Zingiber officinale rhizome (T5)) were harvested, air dried, and milled using standard procedures. Methanolic extracts of the herbs and spices were prepared and used as additives at different concentrations (50, 100, and 150 μL) to the maize substrate for in vitro biogas production. Data were analyzed using regression analysis. There were significant (P < 0.05) differences across all the treatments on the volume and pressure of biogas. The pressure and volume of biogas when compared with the levels tested showed differences (P < 0.05) across all the treatments for the prediction of volume from pressure of biogas. The pressure and volume of gas produced in vitro increased (P < 0.05) and biogases decreased (P < 0.05) by the substrate treated with herbs and spices but for the drum stick leaves which was similar for the levels of concentration tested. This means that the level tested had a pronounced mitigation effect on pressure of biogas and volume of biogas produced. It was concluded that the herb and spice extracts have the potential to improve rumen fermentation and reduce the production of biogases in ruminant diet.

The aim of this study was to investigate the lead (Pb)-induced lipid metabolism impairment and its amelioration using plant-based therapeutic interventions. Pb-induced hepatotoxicity can disturb the normal levels of natural antioxidant enzymes including glutathione (GSH) and superoxide dismutase (SOD) exerting a crucial impact on membrane unsaturated fatty acids (FA), hence leading to lipid peroxidation. Furthermore, Pb toxicity can also alter the regulation of various hormones involved in the synthesis of 3-hydroxy-methyl glutaryl CoA (HMG-CoA reductase), leading to an impairment in normal levels of serum cholesterol and other associated conjugated lipid molecules such HDL-cholesterol, LDL-cholesterol and VLDL-cholesterol. In this study, the lipoprotein fractions, cholesterol, triglyceride (TGs) and biomarkers of liver functions were estimated by employing respective assay kits. The levels of antioxidant enzymes, FFAs and HMG-CoA reductase were determined by employing sandwich ELISA method. The administration of PbAc in experimental rats induced a significant disturbance in lipid profile (P < 0.05) accompanying a significant reduction in natural antioxidant defence system (P < 0.05). The significant alteration in the levels of serum antioxidant enzymes can lead to membrane lipid peroxidation that is reflected by a significantly (P < 0.05) high level of serum MDA in PbAc-induced experimental rats. However, the administration of resveratrol proved therapeutically effective in the treatment of Pb toxicity. Overall, the results of this study accompanying histopathological examination had proved the ameliorating effect of resveratrol in Pb-induced lipid metabolism impairment by adopting vitamin C as a standard therapeutic intervention.

Crocin (C₄₄H₆₄O₂₄) is an isolated bioactive molecule of saffron extract. It has different pharmacological effects such as antioxidant and anti-inflammatory activities. In the present study, radioprotective property of crocin was investigated in the rat liver. Thirty-two rats were equally divided into four groups: (1) control (normal saline), (2) crocin (200 mg/kg), (3) γ-rays (6Gy), and (4) crocin plus γ-rays-treated groups. The liver histopathology, serum transaminases (ALT and AST), alkaline phosphatase (ALP), tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β), and hepatic lipid peroxidation, superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GPX) have been assessed. The histopathological result of hepatic tissue in group 3 showed hydropic degeneration and this progressed to focal or spotty necrosis through the lobule. Moreover, some sinusoids are distended with blood or with leukocytic infiltrations. Other cases in group 3 showed periportal leukocytic infiltrations and necrosis extended out from the portal tract to involve hepatic lobules with fibrinous necrosis in portal vessels, while the examination of hepatic tissues of group 4 showed reduced deformities, irregular arrangement, congested hepatic vessels, and necrosis in hepatocytes. The results also showed significant decreased level of liver function activities, inflammatory markers, lipid peroxidation, and increased levels of liver antioxidants enzymes in group 4. Crocin showed moderate protective effect against γ-rays-induced liver toxicity. The antioxidant effect of crocin may be a major reason for its positive impact on liver parameters. Graphical abstract .
