Diversity and abundance of bacterial pathogens in urban rivers impacted by domestic sewage

In developing countries, many urban rivers are suffering from heavy contamination by untreated sewage, which implies great microbial risks. However, information regarding the bacterial pathogen diversity and distribution in urban rivers is highly limited. In this study, 41 water samples of fifteen rivers and eight samples from two sewage treatment plants in Changzhou City of Yangtze River Delta were sampled. Next-generation sequencing and a self-built reference pathogen database were used to investigate the diversity of enteric and environmental pathogens. The results indicated that the studied urban rivers were harboring diverse potential pathogen species, which primarily included enteric pathogens in Arcobacter and Bacteroides, and environmental pathogens in Acinetobacter, Aeromonas and Pseudomonas. Quantification of twelve pathogens/indicators of interest by qPCR showed that Escherichia coli, Enterococcus faecalis, Campylobacter jejuni, Arcobacter cryaerophilus, Acinetobacter johnsonii, Acinetobacter lwoffii and Aeromonas spp. were abundant, with median values ranging from 3.30 to 5.85 log10 copies/100 mL, while Salmonella, Legionella pheumophila, Mycobacterium avium, Pseudomonas aeruginosa and Staphylococcus aureus were infrequently quantified. The pollution of nutrients and human intestinal microorganisms indicated by specific markers were found to be prevalent but with different levels in the rivers. The correlation analyses revealed that the diversity (p < 0.01) and concentrations (p < 0.05) of the enteric pathogens highly correlated to the human fecal marker abundances, which indicated that the enteric pathogens in the urban rivers were likely to have originated from domestic sewage. The environmental pathogens, which are different from the enteric ones, showed various distribution patterns, and some of them were more abundant in the rivers of rich nutrient. Our findings provide a comprehensive understanding of the bacterial pathogen distribution and influencing factors in urban rivers that are impacted by domestic sewage, thereby establishing the foundation for urban water management.

Wild birds have been shown to be significant sources of numerous types of pathogens that are relevant to humans and agriculture. The presence of large numbers of migratory birds in such a sensitive and important ecosystem as the Platte River in central Nebraska, USA, could potentially serve a significant source of bird-derived pathogens in the water/sediment and riverine environment. In 2009 and 2010, a study was completed to investigate the potential water-quality impacts of Sandhill Cranes and Snow Geese on the microbial water quality of the central Platte River during their spring migration period. Fecal material, river-bottom sediment, and water samples were collected from January through May of each year during the spring migration season of Sandhill Cranes in the Central Flyway of North America. Results indicate that several types of fecal indicator bacteria and from a range of viral, protozoan, and bacterial pathogens, Campylobacter jejuni were present in Sandhill Crane excreta, and at significantly higher frequency and densities in water and sediments when the Sandhill Cranes were present, particularly during evening roosts within the Platte River environment. Therefore, further investigation of the health significance of avian pathogens is warranted for the Platte River in Central Nebraska during migration of Sandhill Cranes and other waterfowl.

The aim of this study was the evaluation of the occurrence of pathogenic Campylobacter, Escherichia coli O157:H7, E. coli virulence genes and Salmonella spp. in different wastewater treatment plants (WWTPs) using a method based on an enrichment step and PCR. This method was sensitive enough to detect low levels (∼2 CFU100 ml⁻¹ of raw sewage) of all the investigated pathogens. In the WWTP samples, E. coli O157:H7 DNA and the eae gene were never found, but 33 % of influents and effluents exhibited amplicons corresponding to Shiga-like toxin I. Twenty-five percent of the influent and 8 % of the effluent exhibited the presence of Shiga-like toxin II. Campylobacter jejuni and C. coli DNA were identified in 50 and 25 % of the influents and in 8 and 25 % of the effluents, respectively. Salmonella spp. DNA was present in all the samples. Considering the results obtained, the method tested here offers a reliable and expeditious tool for evaluating the efficiency of the effluent treatment in order to mitigate contamination risk. Influent contamination by Salmonella spp. and Campylobacter spp. provides indirect information about their circulation; moreover, their presence in effluents underlines the role of WWTPs in the contamination of the receiving surface waters, which affects public health directly or indirectly.

Typical recreational water risk to swimmers is assessed using epidemiologically derived correlations by means of fecal indicator bacteria (FIB). It has been documented that concentrations of FIB do not necessarily correlate well with protozoa and viral pathogens, which pose an actual threat of illness and thus sometimes may not adequately assess the overall microbial risks from water resources. Many of the known pathogens have dose-response relationships; however, measuring water quality for all possible pathogens is impossible. In consideration of a typical freshwater receiving secondarily treated effluent, we investigated the level of consistency between the indicator-derived correlations and the sum of risks from six reference pathogens using a quantitative microbial risk assessment (QMRA) approach. Enterococci and E. coli were selected as the benchmark FIBs, and norovirus, human adenovirus (HAdV), Campylobacter jejuni, Salmonella enterica, Cryptosporidium spp., and Giardia spp. were selected as the reference pathogens. Microbial decay rates in freshwater and uncertainties in exposure relationships were considered in developing our analysis. Based on our exploratory assessment, the total risk was found within the range of risk estimated by the indicator organisms, with viral pathogens as dominant risk agents, followed by protozoan and bacterial pathogens. The risk evaluated in this study captured the likelihood of gastrointestinal illnesses only, and did not address the overall health risk potential of recreational waters with respect to other disease endpoints. Since other highly infectious pathogens like hepatitis A and Legionella spp. were not included in our analysis, these estimates should be interpreted with caution.

An outbreak of food-poisoning caused by Campylobacter jejuni occured in Omachi city, Nagano prefecture during November and December, 1981. The symptoms common to the 462 patients were abdominal pain (87.2%), diarrhoea (53.9%), fever (42.6%), nausea (28.6%), headache (9.5%) and vomiting (5.6%). Positive rate of isolation of Campylobacter jejuni from the stools of patients in acute stage and convalescent stage was 36.7%, Sera from 6 patients showed elevated agglutinin liters, ranging from 1 : 40 to 1 : 160, against the formalinized antigens of the isolates. Contamination of a lunch served in the school in November 25th was presumed to be the cause of the outbreak. This was the first report on the outbreak of food-poisoning by this microorganism in Nagano prefecture.
