Influence of environmental factors on the response of a natural population of Daphnia magna (Crustacea: Cladocera) to spinosad and Bacillus thuringiensis israelensis in Mediterranean coastal wetlands

The present study was undertaken to assess the impact of a candidate mosquito larvicide, spinosad (8, 17 and 33 μg L−1) on a field population of Daphnia magna under natural variations of water temperature and salinity, using Bti (0.16 and 0.50 μL L−1) as the reference larvicide. Microcosms (125 L) were placed in a shallow temporary marsh where D. magna was naturally present. The peak of salinity observed during the 21-day observation period may have been partly responsible for the decrease of daphnid population density in all the microcosms. It is also probably responsible for the absence of recovery in the microcosms treated with spinosad which caused a sharp decrease of D. magna abundance within the first two days following treatment whereas Bti had no effect. These results suggest that it may be difficult for a field population of daphnids to cope simultaneously with natural (water salinity and temperature) and anthropogenic (larvicides) stressors. To evaluate precision and reliability of recently introduced Metal Pollution Index as a method for metal pollution monitoring of surface waters, a series of toxicity tests with Daphnia magna have been undertaken. The sampling sites (the River Danube wetlands and Hydrosystem DTD (Danube - Tisza - Danube), Serbia, Yugoslavia) for this study have been chosen according to previously recorded high MPI values. Chemical characterization and WET (whole effluent toxicity) testing of selected waste waters at the chosen sampling sites, and the recipient water toxicity testing upstream and downstream the discharger evaluated the validity of MPI, and, on the other hand, discriminated the effects of background pollution. The results of acute and chronic toxicity tests of water and sediment corresponded well with MPI values, which leads to a conclusion that MPI could be regarded as a reliable and precise method for metal pollution monitoring of surface waters.

The popular paradigm in trophic dynamic theory is that contemporary autochthonous organic matter (e.g., phytoplankton) sustains consumer growth, whereas aged allochthonous organic matter is conceptually considered recalcitrant resources that may only be used to support consumer respiration but suppress consumer growth. This resource-age paradigm has been challenged by a growing body of recent evidence that ancient (radiocarbon depleted) organic carbon (OC) released from glaciers and permafrost can be incorporated by consumers in aquatic systems. However, little information is available regarding the food quality of ancient terrestrial OC and how it impacts the growth of consumers in lakes. Here, ancient dissolved organic carbon (DOC) was extracted from frozen soils in an alpine lake catchment. The contents of polyunsaturated fatty acids (PUFAs) in soil DOC increased significantly after bioconversion by heterotrophic bacteria. The utilization of soil DOC by heterotrophic bacteria also increased the total phosphorus concentration in the systems. Gammaproteobacteria and Betaproteobacteria showed a strong negative correlation with the percentage contents of fluorescent components, including humic-like and tyrosine-like components. Daphnia magna were fed Auxenochlorella vulgaris and ancient DOC plus heterotrophic bacteria. The contents of PUFAs and the growth of zooplankton were influenced by the pre-conversion time of ancient DOC by bacteria. When ancient DOC was pre-converted by bacteria for 27 days, D. magna fed on the mixed diets showed the highest body length (3.40 mm) and intrinsic rate of increase in population (0.49 d⁻¹). Our findings provide direct evidence that ancient terrestrial OC can be an important subsidy for lake secondary production, which have important implications for food webs in high-altitude and polar lakes.

Various modern products have metallic nanoparticles (MNPs) embedded to enhance products performance. Technological advances enable nowadays even multiple hybrid nanoparticles. Consequently, the future co-release of multiple MNPs will inevitably result in the presence of MNP mixtures in the environment. An important question is if the responses of mixtures of MNPs can be dealt with in a similar way as with the responses of biota to mixtures of metal salts. Moreover, natural organic matter (NOM) is an important parameter affecting the behavior and effect of MNPs. Herein, we determined the joint toxicity and accumulation of copper nanoparticles (CuNPs) and zinc oxide nanoparticles (ZnONPs) in Daphnia magna in the absence and presence of Suwannee River natural organic matter (SR-NOM), compared to the joint toxicity and accumulation of corresponding metal salts. The results of toxicity testing showed that the joint toxicity of CuNPs + ZnONPs was greater than the single toxicity of CuNPs or ZnONPs. The joint toxic action of CuNPs + ZnONPs was additive or more-than-additive for D. magna. A similar pattern was found in the toxicity of the mixtures of Cu- and Zn-salts from the literature data. The presence of SR-NOM had no significant impact on the joint toxicity of CuNPs + ZnONPs. The calculated component-specific contribution to overall toxicity indicated that SR-NOM increased the relative contribution of dissolved ions released from the MNPs to the toxicity of the binary mixtures at high-effect concentrations of individual MNPs. Moreover, dissolved Zn-ions released from the ZnONPs were found to dominate the joint toxicity of CuNPs + ZnONPs in the presence of SR-NOM. Furthermore, the results of the accumulation experiment displayed that the presence of SR-NOM significantly enhanced the accumulation of either CuNPs or ZnONPs in D. magna exposed to the MNP mixtures.

A monitoring network was established in streams within a catchment near the Costa Rican Pacific coast (2008–2011) to estimate the impact of pesticides in surface water (84 samples) and sediments (84 samples) in areas under the influence of melon and watermelon production. A total of 66 (water) and 47 (sediment) pesticides were analyzed, and an environmental risk assessment (ERA) was performed for four taxa (algae, Daphnia magna, fish and Chironomus riparius). One fungicide and seven insecticides were detected in water and/or sediment; the fungicide azoxystrobin (water) and the insecticide cypermethrin (sediments) were the most frequently detected pesticides. The insecticides endosulfan (5.76 μg/L) and cypermethrin (301 μg/kg) presented the highest concentrations in water and sediment, respectively. The ERA revealed acute risk in half of the sampling points of the melon-influenced area and in every sampling point from the watermelon-influenced area. Safety levels were exceeded within and around the crop fields, suggesting that agrochemical contamination was distributed along the catchment, with potential influence of nearby crops. Acute risk was caused by the insecticides chlorpyrifos, cypermethrin and endosulfan to D. magna, fish and C. riparius; the latter was the organism with the overall highest/continuous risk. High chronic risk was determined in all but one sampling point, and revealed a higher number of pesticides of concern. Cypermethrin was the only pesticide to pose chronic risk for all benchmark organisms. The results provide new information on the risk that tropical crops pose to aquatic ecosystems, and highlight the importance of including the analysis of sediment concentrations and chronic exposure in ERA.

Thermal discharge and heatwaves under climate change may increase water temperature. In this study, the individual and combined effect of elevated temperature and cadmium (Cd) toxicity on somatic growth and reproduction of Daphnia magna was evaluated using a simplified dynamic energy budget model (DEBtox). The model predicted that the maximum body length (Lₘ) would be shorter (3.705 mm) at an elevated temperature of 25 °C than at 20 °C (3.974 mm), whereas the maximum reproduction rate (R˙m) would be higher at 25 °C (5.735) than at 20 °C (5.591). The somatic growth and reproduction of D. magna were significantly (p < 0.05) reduced with increasing Cd concentrations, and the reduction was greater at 25 than at 20 °C. Potentiation of Cd toxicity by elevated temperature was correctly simulated by assuming four toxicological modes of action influencing assimilation, somatic maintenance and growth, and reproduction. Overall, the population growth rate of D. magna was expected to decrease linearly with increasing Cd concentrations, and the decrease was expected to be higher at 25 than at 20 °C. These findings suggest a significant ecological risk of toxic metals at elevated temperature, with a mechanistic interpretation of the potentiation effect using a DEBtox modeling approach.

There is limited information regarding the toxicity of the trace element thallium (Tl) to aquatic biota, most of which assesses acute toxicity and bioaccumulation. The relative lack of chronic Tl toxicity data compromises the establishment of water quality criteria for this trace metal. In the presented work, chronic toxicity endpoints (final body weight (a proxy measure of growth), survival, and reproduction) and Tl body burden were measured in the freshwater crustacean Daphnia magna during a 21-day exposure to dissolved Tl. Thallium caused complete mortality in daphnids between exposure concentrations of 424 and 702 μg L⁻¹. In contrast with previously published work examining acute Tl toxicity, exposure to Tl for 21 days was not associated with changes in whole-body potassium concentration. This was despite a 710-fold increase in Tl body burden in animals exposed to 424 μg L⁻¹ relative to the control. Median effect concentrations (EC₅₀’s) for growth and reproduction (total neonates produced), were 1.6 (95% confidence interval: 1.0–3.1) and 11.1 (95% confidence interval: 5.5–21.8) μg Tl L⁻¹, respectively. A no observable effect concentration (NOEC) of 0.9 μg Tl L⁻¹ for growth, and a NOEC range of 0.9–83 μg Tl L⁻¹ for a variety of reproductive metrics, was measured. A lowest observable effect concentration (LOEC) of 8.8 μg Tl L⁻¹ was determined for the effects of Tl on growth and most of the reproductive endpoints examined. These data indicate that under controlled laboratory conditions D. magna is significantly less sensitive to Tl than the species on which the current Canadian Council of Ministers of the Environment regulatory guideline value of 0.8 μg L⁻¹ is based.

In ecological risk assessment, acute to chronic ratio (ACR) uncertainty factors are routinely applied to acute mortality benchmarks to estimate chronic toxicity thresholds. To investigate variability of aquatic ACRs, we first compiled and compared 56 and 150 pairs of acute and subchronic/chronic growth/reproductive toxicity data for fishes (Pimephales promelas (53), Danio rerio (2), and Oryzias latipes (1)) and the crustacean Daphnia magna, respectively, for 172 chemicals with different modes of action (MOA). We found that there were only significant relationships between P. promelas acute median lethal concentrations and growth lowest-observed effect concentrations for class 1 (nonpolar narcosis) chemicals, though significant relationships were demonstrated for D. magna to all Verhaar et al. MOA classes (Class 1: nonpolar narcosis, Class 2: polar narcosis, Class 3: reactive chemicals, and Class 4: AChE inhibitors and estrogenics). Probabilistic ecological hazard assessment using chemical toxicity distributions was subsequently employed for each MOA class to estimate acute and chronic thresholds, respectively, to identify MOA and species specific ecological thresholds of toxicological concern. Finally, novel MOA and species specific ACRs using both chemical toxicity distribution comparison and individual ACR probability distribution approaches were identified using representative MOA and chemical categories. Our data-driven approaches and newly identified ACR values represent robust alternatives to application of default ACR values, and can also support future research and risk assessment and management activities for other chemical classes when toxicity information is limited for chemicals with specific MOAs within invertebrates and fish.

The molecular mechanism of evaluating 17β-estradiol (E₂)-induced toxicity in female Daphnia magna has not been determined. In this study, the transcriptome of D. magna was analyzed after exposure to three different concentrations (0, 10, and 100 ng L⁻¹) of E₂ at 3, 6, and 12 h. The results showed 351–17,221 significantly up-regulated and 505–10,282 significantly down-regulated genes (P < 0.05). Overall, the selected largest 10,282 (10 ng L⁻¹vs control at 12 h) down-regulated and 17,221 (100 vs 10 ng L⁻¹) up-regulated genes were identified; following annotation, pathways in cancer and RNA transport were found to be enriched according to the interaction network. Among all completed comparisons, KEGG pathways related to the immune system, cancer, disease infection, and active compound metabolism were identified by short time series expression miner analysis. A different set of genes fluctuated in a “U”-shaped pattern over time and at different concentrations of E₂, whereas some genes associated with disintoxication showed a reverse “U”-shaped response as E₂ administration was increased. These results suggest that E₂ exposure caused transcriptional changes in the immune system, disintoxication, disease prevention, and the protein degradation pathway.

Water pollution by pesticides used in agriculture is currently a major concern both in Spain and in Europe as a whole, prompting the need to evaluate water quality and ecological risk in areas of intensive agriculture. This study involved monitoring pesticide residues and certain degradation products in surface and ground waters of the Denomination of Origin (DO) Jumilla vineyard area in Spain. Sixty-nine pesticides were selected and evaluated at twenty-one sampling points using a multi-residue analytical method, based on solid-phase extraction (SPE) and analysis by liquid chromatography coupled with mass spectrometry (LC-MS), providing reliable results. Twenty-six compounds from those selected were detected in the samples analyzed (eleven insecticides including one degradation product, nine herbicides, and six fungicides) and fifteen of them were found in concentrations over 0.1 μg L⁻¹ (upper threshold established by the EU for pesticides detected in waters for human consumption). Indoxacarb was present in more than 70% of the samples, being the most frequently detected compound in water samples. Some pesticides were ubiquitous in all the water samples. Ecotoxicological risk indicators, toxic units (TUs) and risk quotients (RQs), for algae, Daphnia magna and fish were calculated to estimate the environmental risk of the presence of pesticides in waters. The compounds with the highest risk were the herbicides pendimethalin, with RQ values > 1 for the three aquatic organisms, and diflufenican, posing a high risk for algae and fish, and the insecticide chlorpyrifos, with a high risk for Daphnia magna and fish. The ∑TUi determined for water at each sampling point posed only a high risk for the three aquatic organisms in a sample. These results are important for considering the selection of pesticides with less environmental risk in intensive agricultural areas.
