Bioactivity of MWCNT in Conidia of Entomopathogenic Fungus Isaria fumosorosea

The bioactivity of three kinds of multi-walled carbon nanotubes (MWCNT) towards the conidia of entomopathogenic fungus Isaria fumosorosea was examined in an in vitro study. Commercial—raw and functionalized—carboxylated MWCNT were applied. A fungal conidia suspension was placed in contact with dispersed MWCNT over different time-periods. After contact with the nanomaterial, the conidia were cultured on dishes and both the linear vegetative mycelium growth and the sporulation and germination of the spores derived from the culture were investigated. Also, the pathogenicity of the conidia after contact with MWCNT was examined in relation to test larvae. No fungistatic activity of MWCNT relative to I. fumosorosea conidia was demonstrated. Conidia contact with MWCNT resulted in the following changes in vital processes in the subsequent culture compared to the control standard culture: (1) raw MWCNT limited mycelium inoculation, but the growth rate observed later in the log-phase was more intense; (2) after 24-h conidia contact with all MWCNT types, the mycelium sporulated the most intensively; longer contact resulted in sporulation process limitation. Germination of conidia after contact with the MWCNT was not significantly modified. Raw MWCNT potentiated conidia pathogenicity towards test insects. It was observed that carboxylation of MWCNT reduces the bioactivity of this nanomaterial towards the investigated conidia.

Presence of industrial dyes and heavy metal as a contaminant in environment poses a great risk to human health. In order to develop a potential technology for remediation of dyes (Reactive remazol red, Yellow 3RS, Indanthrene blue and Vat novatic grey) and heavy metal [Cu(II), Ni(II), Cd(II), Zn(II), Cr(VI) and Pb(II)] contamination, present study was performed with entomopathogenic fungi, Beauveria bassiana (MTCC no. 4580). High dye removal (88–97%) was observed during the growth of B. bassiana while removal percentage for heavy metals ranged from 58 to 75%. Further, detailed investigations were performed with Pb(II) in terms of growth kinetics, effect of process parameters and mechanism of removal. Growth rate decreased from 0.118 h⁻¹ (control) to 0.031 h⁻¹, showing 28% reduction in biomass at 30 mg L⁻¹ Pb(II) with 58.4% metal removal. Maximum Pb(II) removal was observed at 30 °C, neutral pH and 30 mg L⁻¹ initial metal concentration. FTIR analysis indicated the changes induced by Pb(II) in functional groups on biomass surface. Further, microscopic analysis (SEM and atomic force microscopy (AFM)) was performed to understand the changes in cell surface morphology of the fungal cell. SEM micrograph showed a clear deformation of fungal hyphae, whereas AFM studies proved the increase in surface roughness (RSM) in comparison to control cell. Homogenous bioaccumulation of Pb(II) inside the fungal cell was clearly depicted by TEM-high-angle annular dark field coupled with EDX. Present study provides an insight into the mechanism of Pb(II) bioremediation and strengthens the significance of using entomopathogenic fungus such as B. bassiana for metal and dye removal.

The stored grain insects cause great damage to grains under storage conditions. Synthetic insecticides and fumigants are considered as key measures to control these stored grain insect pests. However, the major issue with these chemicals is grain contamination with chemical residues and development of resistance by insect pests to these chemicals. Biological control is considered as a potential alternative to chemical control especially with the use of pathogens, alone or in combination with selective insecticides. The present study was conducted to evaluate the synergism of Metarhizium anisopliae with diatomaceous earth (DE) and thiamethoxam against four insect pests on the stored wheat grains. In the first bioassay, the M. anisopliae was applied at 1.4 × 10⁴ and 1.4 × 10⁶conidia/ml alone and in integration with two concentrations (250 and 500 ppm) of tested DE. The tested fungus when combined with DE and thiamethoxam possessed synergistic impact as compared to their individual efficacy. Adult mortality increased with respect to increased exposure interval and doses. In the second bioassay, M. anisopliae was applied at 1.4 × 10⁴ conidia/ml individually and in combination with three concentrations (0.50, 0.75, and 1.00 ppm) of thiamethoxam. Results concluded that M. anisopliae integrated with DE and thiamethoxam provides more effective control of stored grain insect pests.

Mosquitoes transmit dreadful diseases, causing millions of deaths every year. Therefore, screening for larvicidal and pupicidal activity of microbial extracts attributes could lead to development of new and improved mosquito control methods that are economical and safe for nontarget organisms and are ecofriendly. Synthetic chemical insecticides occupy predominant position in control strategies. These hazardous chemicals exert unwarranted toxicity and lethal effects on nontarget organisms, develop physiological resistance in target, and cause adverse environmental effect. For vector control, fungal-mediated natural products have been a priority in this area at present. In the current study, effective larvicidal and pupicidal effect of mycosynthesized silver nanoparticles (Ag NPs) using an entomopathogenic fungi Trichoderma harzianum against developmental stages of the dengue vector Aedes aegypti was investigated. An attractive possibility of green nanotechnology is to use microorganisms in the synthesis of nanosilver especially Ag NPs. The mycosynthesized Ag NPs were characterized to find their unique properties through UV-visible spectrophotometer, X-ray diffraction analysis, Fourier transform infrared, and surface characteristics by scanning electron microscopy. To analyze the bioefficacy, different test concentrations for extracellular filtrate (0.2, 0.4, 0.6, 0.8, and 1.0 %) and Ag NPs (0.05, 0.10, 0.15, 0.20, and 0.25 %) were prepared to a final volume of 200 mL using deionized water; 20 larvae of each instars (I–IV) and pupa were exposed to each test concentration separately which included a set of control (distilled water) group with five replicates. Characterization of the synthesized Ag NPs were about 10–20 nm without aggregation. Susceptibility of larval instars to synthesized Ag NPs was higher than the extracellular filtrate of T. harzianum alone after 24-h exposure, where the highest mortality was recorded as 92 and 96 % for first and second instars and 100 % for third, fourth instars, and pupa. Lethal concentration 50 values of 0.079, 0.084, 0.087, 0.068, and 0.026 % were recorded for I–IV instars and pupa, respectively, when exposed to Ag NPs at 0.25 % concentration. Toxicity was exhibited against first (1.076 %), second (0.912 %), third (0.770 %), fourth (0.914 %) instars larvae, and pupa (0.387 %) with extracellular filtrate at a concentration of 1 % that was three- to fourfold higher compared to Ag NPs; no mortality was observed in the control. The present study is the first report on effective larvicidal and pupicidal activity of Ag NPs synthesized from an entomopathogenic fungi T. harzianum extracellular filtrate and could be an ideal ecofriendly, single-step, and inexpensive approach for the control of A. aegypti.
