Biota monitoring under the Water Framework Directive: On tissue choice and fish species selection

The study addresses the topic of suitable matrices for chemical analysis in fish monitoring and discusses the effects of data normalization in the context of the European Water Framework Directive (WFD). Differences between species are considered by comparing three frequently monitored species of different trophic levels, i.e., chub (Squalius cephalus, n = 28), (bream, Abramis brama, n = 11), and perch (Perca fluviatilis, n = 19) sampled in the German Danube. The WFD priority substances dioxins, furans and dioxin-like polychlorinated biphenyls (PCDD/F + dl-PCB), polybrominated diphenyl ethers (PBDE), α-hexabromocyclododecane (α-HBCDD), hexachlorobenzene (HCB), mercury (Hg), and perfluorooctane sulfonic acid (PFOS) as well as non-dioxin-like (ndl)-PCB were analyzed separately in fillet and carcass and whole body concentrations were calculated. Hg was analyzed in individual fish fillets and carcasses, all other substances were determined in pool samples, which were compiled on the basis of fish size (3 chub pools, 1 bream pool, 2 perch pools). The data were normalized to 5% lipid weight (or 26% dry mass in the case of Hg and PFOS) for comparison between matrices and species.Hg concentrations were generally higher in fillet than in whole fish (mean whole fish-to-fillet ratio: 0.7) whereas all other substances were mostly higher in whole fish. In the case of lipophilic substances these differences leveled after lipid normalization.Significant correlations (p ≤ .05) were detected between Hg and fish weight and age. Hg concentrations varied least among younger fish. PCDD/F, dl-PCB, ndl-PCB, PBDE, α-HBCDD and HCB correlated significantly (p ≤ .05) with lipid concentrations. Fillet-to-whole fish conversion equations and/or conversion factors were derived for all substances except α-HCBDD. Although more data also for individual fish would be desirable the results are nevertheless a step on the way to translate fillet concentrations of priority substances to whole fish concentrations.

Dioxins and dioxin-like polychlorinated biphenyls (dl-PCBs) in fish fillet counteract the health benefits of fish products. In this study, food waste was used as a protein alternative to replacing fishmeal commonly used in the commercial fish feed, aiming to cultivate Sabah grouper with acceptable levels of dioxins and dl-PCBs. The concentrations of dioxins and dl-PCBs, as well as the fish growth performance, were compared between the fish groups fed with food waste-based feed (FWBF) and commercial feed (Nanyu®, control). The results showed that the concentrations of polychlorinated dibenzo-p-dioxins (PCDDs) (1.22 pg/g dry weight (d.w.)) and non-ortho-dl-PCBs (13.0 pg/g d.w.) were significantly lower (p < 0.05) in the FWBF than in the control feed (commercial feed) (PCDDs: 2.35 pg/g d.w.; non-ortho-dl-PCBs: 27.2 pg/g d.w.). The growth performance of the fish group fed with the FWBF was comparable to that fed with the control feed. There were no significant differences between the WHO₂₀₀₅-TEQ values of different fish fillets (1.00, 1.11, and 1.10 pg WHO₂₀₀₅-TEQ/g d.w. for FWBF group, control feed group, and local market fish, respectively). Based on the guidelines provided by European Food Safety Authority (ESFA) and U.S. Environmental Protection Agency (USEPA), the fish fed with the FWBF were safe for human consumption (hazard index values: 0.284–0.522; cancer risk range: 2.59–2.97 × 10⁻⁵). The findings of this study suggest that food waste could serve as an alternative protein source for cultivating Sabah grouper with acceptable levels of dioxins and dl-PCBs.

Mercury (Hg) and its bioaccumulation are important in evaluating the health risk through fish consumption. In the Marano and Grado Lagoon a historical contamination originating from both mining and industrial sources is present.In this study the Hg levels in the grass goby and the protective effect of selenium (Se) were determined as a function of size, sex, sampling sites and season.Mercury often exceeded the limit for commercialisation, whereas Se was mostly constant being homeostatically regulated.The mean Se/Hg molar ratio showed about 40% of data >1.0 and the HBVSe index was on average equal to 0.0. Thus, grass goby of this area is not a Se source food indicated for pregnant and/or breastfeeding women, but its occasional consumption would appear to have no contraindications if integrated with other Se rich foods: the ingestion of about 10 fish fillets per week can be safely consumed by adults.

Halogenated natural products (HNPs) are frequently detected in marine organisms. High HNP concentrations have previously been found in marine mammals from the Great Barrier Reef, Australia, including in the blubber of herbivorous dugongs (Dugong dugon). To identify the source of HNPs we initially focused on the analysis of Australian seagrass (Halophila ovalis) which serves as the principal food source for dugongs. GC/MS analysis of the seagrass indicated the presence of several organobromine compounds. One compound was identified as 2,3,4,5-tetrabromo-1-methylpyrrole (TBMP) by synthesis. Subsequent analysis of semipermeable membrane devices demonstrated that the photo-sensitive TBMP is widespread in the Great Barrier Reef (Queensland, Australia). The detection of larger TBMP concentrations in fish fillets from Chile and traces in mussels from New Zealand indicated that this potential HNP may be distributed throughout the Southern Pacific Ocean.
