In this study, the accumulation and toxicity effect of 1–7 μM of Hg was determined during 24–72 h on two strains of Chlamydomonas reinhardtii, CC-125 and CC-503 as a cell wall-deficient mutant, by monitoring the growth rate and the maximum quantum yield of Photosystem II. In addition, the level of extracytoplasmic polyphosphates (polyP related to the cell wall) was determined to understand the polyP physiological role in Hg-treated algal cells. The results showed that the polyP level was higher in the strain CC-125 compared to CC-503. When algal cells were exposed to 1 and 3 μM of Hg, the accumulation of Hg was correlated with the degradation of polyP for both strains. These results suggested that the degradation of polyP participated in the sequestration of Hg. In fact, this mechanism might explain at 72 h the recovery of the polyP level, the efficiency of maximum PSII quantum yield, the low inhibition of growth rate, and the low accumulated Hg in algal biomass. Under the effect of 5 and 7 μM of Hg, the degradation of polyP was complete and could not be recovered, which was caused by a high accumulation and toxicity of Hg already at 24 h. Our results demonstrated that the change of polyP level was correlated with the accumulation and effect of Hg on algal cells during 24–72 h, which can be used as a biomarker of Hg toxicity. Therefore, this study suggested that extracytoplasmic polyP in C. reinhardtii contributed to the cellular tolerance for Hg.

The phenolic compounds have posed public concern for potential threats to human health and ecosystem. Tert-butylphenols (TBPs), as one group of emerging contaminants, showed potential endocrine disrupting effects and aquatic toxicities. In the present study, we detected concentrations of 2,4-DTBP ranging from <0.001 to 0.057 μg/L (detection limit: 0.001 μg/L) in drinking water source from the Qiantang River in East China in April 2016. The endocrine disrupting effects of 2-TBP, 2,4-DTBP and 2,6-DTBP toward human estrogen receptor α (ERα), androgen receptor (AR) and thyroid hormone receptor β (TRβ) were evaluated using human recombinant two-hybrid yeast bioassay. Their aquatic toxicities were investigated with indicator organisms including Photobacterium phosphoreum, Vibrio fischeri and freshwater green alga Chlamydomonas reinhardtii. 2-TBP and 2,4-DTBP exhibited moderate antagonistic effects toward human ERα and AR in a concentration-dependent manner. 2-TBP significantly inhibited the light emission of P. phosphoreum. 2-TBP, 2,4-DTBP and 2,6-DTBP significantly inhibited the growth of C. reinhardtii and reduced the chlorophyll content. Our results suggest the potential adverse effects of TBPs on human health and aquatic organisms. The data will facilitate further risk assessment of TBPs and related contaminants.

Recently, there has been a growing concern that climate change may rapidly and extensively alter global ecosystems with unknown consequences for terrestrial and aquatic life. While considerable emphasis has been placed on terrestrial ecology consequences, aquatic environments have received relatively little attention. Limited knowledge is available on the biological effects of increments of seawater temperature and pH decrements on key ecological species, i.e., primary producers and/or organisms representative of the basis of the trophic web. In the present study, we addressed the biological effects of global warming and ocean acidification on two model organisms, the microbenthic marine ciliate Euplotes crassus and the green alga Dunaliella tertiocleta using a suite of high level ecological endpoint tests and sub-lethal stress measures. Organisms were exposed to combinations of pH and temperature (TR1: 7.9[pH], 25.5 °C and TR2: 7.8[pH], 27,0 °C) simulating two possible environmental scenarios predicted to occur in the habitats of the selected species before the end of this century. The outcomes of the present study showed that the tested scenarios did not induce a significant increment of mortality on protozoa. Under the most severe exposure conditions, sub-lethal stress indices show that pH homeostatic mechanisms have energetic costs that divert energy from essential cellular processes and functions. The marine protozoan exhibited significant impairment of the lysosomal compartment and early signs of oxidative stress under these conditions. Similarly, significant impairment of photosynthetic efficiency and an increment in lipid peroxidation were observed in the autotroph model organism held under the most extreme exposure condition tested.

Silver nanoparticles (AgNPs) are widely used because of their excellent antibacterial properties. They are, however, easily discharged into the water environment, causing potential adverse environmental effects. Meta-transcriptomic analyses are helpful to study the transcriptional response of prokaryotic and eukaryotic aquatic microorganisms to AgNPs. In the present study, microcosms were used to investigate the toxicity of AgNPs to a natural aquatic microbial community. It was found that a 7-day exposure to 10 μg L⁻¹ silver nanoparticles (AgNPs) dramatically affected the structure of the microbial community. Aquatic micro eukaryota (including eukaryotic algae, fungi, and zooplankton) and bacteria (i.e., heterotrophic bacteria and cyanobacteria) responded differently to the AgNPs stress. Meta-transcriptomic analyses demonstrated that eukaryota could use multiple cellular strategies to cope with AgNPs stress, such as enhancing nitrogen and sulfur metabolism, over-expressing genes related to translation, amino acids biosynthesis, and promoting bacterial-eukaryotic algae interactions. By contrast, bacteria were negatively affected by AgNPs with less signs of detoxification than in case of eukaryota; various pathways related to energy metabolism, DNA replication and genetic repair were seriously inhibited by AgNPs. As a result, eukaryotic algae (mainly Chlorophyta) dominated over cyanobacteria in the AgNPs treated microcosms over the 7-d exposure. The present study helps to understand the effects of AgNPs on aquatic microorganisms and provides insights into the contrasting AgNPs toxicity in eukaryota and bacteria.

Climbazole (CBZ) is an antibacterial and antifungal agent widely used in personal care products. In this study, we investigated the interactions between climbazole (CBZ) and freshwater microalgae Scenedesmus obliquus (S. obliquus). Dose-effect relationships between CBZ concentrations and growth inhibitions or chlorophyll a content were observed. After 12 days of incubation, the algae density and chlorophyll a content in 2 mg/L treatment group was 56.6% and 15.8% of those in the control group, respectively. Biotransformation was the predominant way to remove CBZ in the culture solution, whereas the contribution of bioaccumulation and bioadsorption were negligible. More than 88% of CBZ was removed by S. obliquus across all treatments after 12 days of incubation, and the biotransformation of CBZ followed the first order kinetic model with half-lives of approximately 4.5 days at different treatments. CBZ-alcohol (CBZ-OH) was the only biotransformation product identified in algal solution. Moreover, the toxicity of biotransformation products was much lower than its corresponding precursor compound (CBZ). The results of this study revealed that S. obliquus might have a great impact on the environmental fates of CBZ and could be further applied to remove organic pollutants in aquatic environment.

Carbon nanotubes (CNTs) are considered promising materials in nanotechnology. We quantified CNT accumulation by the alga Desmodesmus subspicatus. Cells were exposed to radiolabeled CNTs (14C-CNTs; 1 mg/L) to determine uptake and association, as well as elimination and dissociation in clear media. Attenuated total reflection Fourier-transform infrared spectroscopy (ATR-FTIR) was used to detect effects of CNTs on algae. CNT-cell interactions were visualized by electron microscopy and related to alterations in their cell composition. A concentration factor of 5000 L/kg dry weight was calculated. Most of the material agglomerated around the cells, but single tubes were detected in the cytoplasm. Computational analyses of the ATR-FTIR data showed that CNT treated algae differed from controls at all sampling times. CNT exposure changed the biochemical composition of cells. The fact that CNTs are bioavailable for algae and that they influence the cell composition is important with regard to environmental risk assessment of this nanomaterial.

Antibiotics have been widely detected in the ocean and have various impacts on the environment, while knowledge of their chronic influence on phytoplankton, especially red tide algae, is still limited. Dinoflagellates and green algae are common phytoplankton in marine ecosystems. The former is the main red tide algae, and the latter is an important primary producer. We investigated the long-term responses of two representative algae, Prorocentrum lima and Chlorella sp., to two common antibiotics (sulfamethoxazole (SMX) and norfloxacin (NFX)) at environmentally relevant levels (10 and 100 ng/L) during simulated natural conditions. The cell density and activities of three antioxidant enzymes (superoxide dismutase (SOD), catalase (CAT) and peroxidase (POD)) were analyzed. The results showed that the influence of each antibiotic on Chlorella sp. was not significant (p > 0.05) during the first 10 days, but the influence of the antibiotics later began to show significant inhibition (p < 0.05) compared with the control group, especially during mixed exposure. P. lima was not inhibited, but its cell density increased. SMX had a superior stimulation effect on P. lima. The three enzymes activities of P. lima increased, and the antioxidant mechanism was not seriously impacted. However, for Chlorella sp., the activity of SOD increased while the activities of CAT and POD decreased, suggesting that this algae’s antioxidant system was unbalanced due to oxidative stress. Based on our results, the growth of P. lima was different from green algae Chlorella sp. as well as other inhibited marine algae (such as diatom, golden algae) studied in previous studies. Therefore, as a typical pollutant in the ocean, antibiotics may play a positive role in the bloom of dinoflagellate red tides.

Controlling blooms of toxigenic phytoplankton, including cyanobacteria, is a high priority for managers of aquatic systems that are used for drinking water, recreation, and aquaculture production. Although a variety of treatment approaches exist, hydrogen peroxide (H2O2) has the potential to be an effective and ecofriendly algaecide given that this compound may select against cyanobacteria while not producing harmful residues. To broadly evaluate the effectiveness of H2O2 on toxigenic phytoplankton, we tested multiple concentrations of H2O2 on (1) four cyanobacterial cultures, including filamentous Anabaena, Cylindrospermopsis, and Planktothrix, and unicellular Microcystis, in a 5-day laboratory experiment and (2) a dense cyanobacterial bloom in a 7-day field experiment conducted in a nutrient-rich aquaculture pond. In the laboratory experiment, half-maximal effective concentrations (EC50) were similar for Anabaena, Cylindrospermopsis, and Planktothrix (average EC50 = 0.41 mg L−1) but were ∼10x lower than observed for Microcystis (EC50 = 5.06 mg L−1). Results from a field experiment in an aquaculture pond showed that ≥1.3 and ≥ 6.7 mg L−1 of H2O2 effectively eliminated Planktothrix and Microcystis, respectively. Moreover, 6.7 mg L−1 of H2O2 reduced microcystin and enhanced phytoplankton diversity, while causing relatively small negative effects on zooplankton abundance. In contrast, 20 mg L−1 of H2O2 showed the greatest negative effect on zooplankton. Our results demonstrate that H2O2 can be an effective, rapid algaecide for controlling toxigenic cyanobacteria when properly dosed.

Although fluoride occurs naturally in the environment, excessive amounts of fluoride in freshwater and terrestrial ecosystems can be harmful. We evaluated the toxicity of fluoride compounds on the growth, viability, and photosynthetic capacity of freshwater (Chlamydomonas reinhardtii and Pseudokirchneriella subcapitata) and terrestrial (Chlorococcum infusionum) algae. To measure algal growth inhibition, a flow cytometric method was adopted (i.e., cell size, granularity, and auto-fluorescence measurements), and algal yield was calculated to assess cell viability. Rhodamine123 and fluorescein diacetate were used to evaluate mitochondrial membrane potential (MMA, ΔΨm) and cell permeability. Nine parameters related to the photosynthetic capacity of algae were also evaluated. The results indicated that high concentrations of fluoride compounds affected cell viability, cell organelle potential, and photosynthetic functions. The cell viability measurements of the three algal species decreased, but apoptosis was only observed in C. infusionum. The MMA (ΔΨm) of cells exposed to fluoride varied among species, and the cell permeability of the three species generally decreased. The decrease in the photosynthetic activity of algae may be attributable to the combination of fluoride ions (F−) with magnesium ions (Mg2+) in chlorophyll. Our results therefore provide strong evidence for the potential risks of fluoride compounds to microflora and microfauna in freshwater and terrestrial ecosystems.

Oil spill causes extreme environmental damage, from aquatic life to seabirds, disrupting the entire ecosystem. Herein, we have synthesized high scale, economical and bio-compatible, green algae mediated Titanium oxide (TiO₂) nanoparticles and Polyacrylonitrile (PAN) nanofiber mats. We have studied the effect of encapsulation and coating of TiO₂ nanoparticles over nanofiber mats for highly efficient oil spill adsorption. TiO₂ encapsulated and coated PAN (TECP) nanofibers showed a maximum of 62.34 g g⁻¹ adsorption capacity of petroleum oil from the water surface. Moreover, the composite mats show maximum adsorption within 45 s for up to 5 repeated cycles. Further, it has been observed that the adsorption capacity has increased by increasing the weight of the composite nanofiber mats, which confirms its commercial applicability. Thus, this work provides rapid, large-scale, economical, bio-compatible, and highly effective adsorbents for oil spill cleaning and extraction over natural waterbodies.
