Understanding the ecological status of aquatic ecosystems and the impact of anthropogenic contamination requires correlating exposure to toxicants with impact on biological communities. Several tools exist for assessing the ecotoxicity of substances, but there is still a need for new tools that are ecologically relevant and easy to use. We have developed a protocol based on the substrate-induced respiration of a river biofilm community, using the MicroResp™ technique, in a pollution-induced community tolerance approach. The results show that MicroResp™ can be used in bioassays to assess the toxicity toward biofilm communities of a wide range of metals (Cu, Zn, Cd, Ag, Ni, Fe, Co, Al and As). Moreover, a community-level physiological profile based on the mineralization of different carbon substrates was established. Finally, the utility of MicroResp™ was confirmed in an in-situ study showing gradient of tolerance to copper correlated to a contamination gradient of this metal in a small river. Chromium pollution is a serious environmental problem with few cost-effective remediation strategies available. Crambe abyssinica (a member of Brassicaseae), a non-food, fast growing high biomass crop, is an ideal candidate for phytoremediation of heavy metals contaminated soils. The present study used a PCR-Select Suppression Subtraction Hybridization approach in C. abyssinica to isolate differentially expressed genes in response to Cr exposure. A total of 72 differentially expressed subtracted cDNAs were sequenced and found to represent 43 genes. The subtracted cDNAs suggest that Cr stress significantly affects pathways related to stress/defense, ion transporters, sulfur assimilation, cell signaling, protein degradation, photosynthesis and cell metabolism. The regulation of these genes in response to Cr exposure was further confirmed by semi-quantitative RT-PCR. Characterization of these differentially expressed genes may enable the engineering of non-food, high-biomass plants, including C. abyssinica, for phytoremediation of Cr-contaminated soils and sediments.

Elevated levels of copper and zinc in sediment have been shown to adversely affect estuarine infauna. We investigated the additivity of the combined effects of copper and zinc on infaunal recolonisation through a manipulative field experiment in Orewa estuary, New Zealand, using defaunated sediment discs treated with these metals. The nature of their combined effects varied among infaunal taxa and the particular variables being examined. Additive effects were detected for species richness, for the mean log abundances of the polychaetes Prionospio sp. and Scoloplos cylindrifer and for the multivariate response of the community as a whole. Antagonistic effects were detected for the mean log abundances of total infauna and the polychaete Heteromastus sp. Characterising the potentially interactive nature of the combined effects of multiple heavy metals is essential in order to build predictive models of future environmental impacts of metal accumulation in estuarine sediments.

In order to achieve efficient phytoextraction of heavy metals using trees, the metal allocation to aboveground tissues needs to be characterised. In his study, the distribution of heavy metals, macro- and micronutrients and the metal micro-localisation as a function of the leaf position and heavy metal treatment were analysed in poplars grown on soil with mixed metal contamination. Zinc was the most abundant contaminant in both soil and foliage and, together with cadmium, was preferentially accumulated in older foliage whereas excess copper and lead were not translocated. Changes in other element concentrations indicated an acceleration in aging as a consequence of the metal treatment. Excess zinc was irregularly accumulated inside leaf tissues, tended to saturate the veins and was more frequently stored in cell symplast than apoplast. Storage compartments including metabolically safe and sensitive subcellular sites resulted in sizable metal accumulation as well as stress reactions.

Over the past three decades heavy metal pollution has increased substantially in Cochin estuary, south west coast of India. Here we studied the distribution, diversity and enzyme expression profile of culturable microbial population along a pollution gradient. The distribution of resistance against 5 mM concentration of Zn, Co, Ni and Cu was observed among 90–100% of bacterial isolates retrieved from highly polluted Eloor, whereas it was less than 40% in Vypin and Munambam. Similarly, there was a difference in the distribution and diversity of bacterial phyla with predominance of Proteobacteria in Eloor and Firmicutes in Munambam and Vypin. We observed that 75–100% of the organisms retrieved from Eloor had low levels of expression for hydrolytic enzyme. In conclusion, the heavy metal pollution in Cochin estuary brought in reduction/adaptation in the distribution, diversity and enzyme expression profile of bacteria, which may impart adverse impacts on ecosystem functioning.

The concentrations of heavy metals (Cr, Cd, Hg, Cu, Zn, Pb and As) in the water, sediment, and fish were investigated in the middle and lower reaches of the Yangtze River, China. Potential ecological risk analysis of sediment heavy metal concentrations indicated that six sites in the middle reach, half of the sites in the lower reach, and two sites in lakes, posed moderate or considerable ecological risk. Health risk analysis of individual heavy metals in fish tissue indicated safe levels for the general population and for fisherman but, in combination, there was a possible risk in terms of total target hazard quotients. Correlation analysis and PCA found that heavy metals (Hg, Cd, Pb, Cr, Cu, and Zn) may be mainly derived from metal processing, electroplating industries, industrial wastewater, and domestic sewage. Hg may also originate from coal combustion. Significant positive correlations between TN and As were observed.

The leaves of date palms were evaluated as a possible biomonitor of heavy metal contamination in Ma’an city, Jordan. Concentrations of (Fe), (Pb), (Zn), (Cu), (Ni), and (Cr) were determined in washed and unwashed leaves and soil samples collected from different sites with different degrees of metal contamination (urban, suburban, industrial, highway and rural sites); separate leaves were taken from outside the city to be used as a control sample. Samples collected from industrial sites were found to have high concentrations of all metals except those of Cu, Ni and Pb, which were found at high levels in the highway site samples which is associated with the road traffic. The difference between unwashed and washed samples showed that metal pollutants exist as contaminants, particularly Pb, Zn and Ni, which varied in concentration, depending on the source of the metal.

Soils were sampled in three types of wetlands from the young (A) and old (B) reclaimed regions of the Pearl River Estuary. They were analyzed for total concentrations of heavy metals to investigate their distributions and pollution levels in both regions. Results showed that most heavy metals in ditch and riparian wetlands did not significantly differ from those in reclaimed wetlands in A region, while significantly lower for Cd, Cu, Pb, and Zn in reclaimed wetlands in B region, suggesting higher effects of long-term reclamation. Iron, Cr and Cu were identified as metal pollutants of primary concern and had higher contributions to the total toxic units compared to other metals. Almost all metals exceeded their lowest effect levels and Fe and Cr even exceeded the severe effect levels. Multivariate analysis shows that Fe and Mn are controlled by parent rocks and other metals mainly originate from anthropogenic source.

The role of extracellular polymeric substances (EPS) in Cd adsorption by Bacillus subtilis and Pseudomonas putida was investigated using a combination of batch adsorption experiments, potentiometric titrations, Fourier transform infrared spectroscopy (FTIR). An increased adsorption capacity of Cd was observed for untreated bacteria relative to that for EPS-free bacteria. Surface complexation modeling of titration data showed the similar pKₐ values of functional groups (carboxyl, phosphate and hydroxyl) between untreated and EPS-free bacteria. However, site concentrations on the untreated bacteria were found to be higher than those on the EPS-free bacteria. FTIR spectra also showed that no significant difference in peak positions was observed between untreated and EPS-free bacteria and carboxyl and phosphate groups were responsible for Cd adsorption on bacterial cells. The information obtained in this study is of fundamental significance for understanding the interaction mechanisms between heavy metals and biofilms in natural environments.
