In this study, the lethal toxicity, behavioral responses and hematotoxicity of formulated chlorpyrifos on Clarias gariepinus was evaluated. C. gariepinus fingerlings were exposed to 0.2 mg/L, 0.25 mg/L, 0.3 mg/L, 0.35 mg/L and 0.4 mg/L of the active ingredient chlorpyrifos to determine the lethal concentrations and behavioral effects. C. gariepinus juveniles (38.84±7.67g) were then exposed to 0.0256 mg/L and 0.0128 mg/L for 14 days to study somatic indices and haematological effects. The 24h, 48h, 72h and 96h LC50 were estimated as 0.292 (0.210 – 0.376) mg/L, 0.275 (0.252 – 0.297) mg/L, 0.263 (0.242 – 0.282) mg/L, and 0.256 (0.235 – 0.275) mg/L respectively. Hyper activity, loss of equilibrium, erratic swimming, trembling, respiratory distress and poor startle response were observed in fingerlings in response to acute toxic stress of chlorpyrifos. Liver somatic index (LSI) of exposed juveniles increased significantly (p<0.05) compared with control, while there was no statistically significant difference in all the haematological parameters of the exposed fishes compared with the control (p<0.05). The results indicate that the chlorpyrifos formulation was highly toxic and induced behavioral changes in C gariepinus fingerlings, while sub-lethal concentrations induced inflammation in the liver but had no effect on haematological parameters of Clarias gariepinus juveniles. LSI was sensitive to the sub-lethal concentrations and could serve as indicators or exposure to organophosphate insecticides.

Hypoxia (low oxygen) often occurs in aquatic ecosystems that receive effluent from municipal wastewater treatment plants (WWTP). The combination of hypoxia and WWTP effluent could impair fish health, because WWTP effluent contains multiple contaminants that could disrupt the physiological pathways fish use to cope with hypoxia, but the interactive effects of these stressors on fish physiology are poorly understood. We have examined this issue by exposing mummichog killifish (Fundulus heteroclitus) to hypoxia (5 and 2 kPa O₂) and/or 100% WWTP effluent for 21 days in a full factorial design. We then measured hypoxia tolerance, whole-animal metabolism, gill morphology, haematology, and tissue metabolites. In clean water, killifish responded to chronic hypoxia with improvements in hypoxia tolerance, as reflected by increases in time to loss of equilibrium at 0.5 kPa (tLOE). These improvements occurred in association with increases in the exposed surface of gill lamellae that resulted from a regression of interlamellar cell mass (ILCM). Concurrent exposure to wastewater attenuated the increases in tLOE and gill remodeling in chronic hypoxia, and nearly depleted brain glycogen stores. Therefore, exposure to WWTP effluent can disrupt the physiological mechanisms fish use to cope with chronic hypoxia and impair hypoxia tolerance. Our research suggests that the combination of stressors near WWTPs can have interactive effects on the physiology and health of fish.

Aquatic ecosystem health is the main concern to increasing pesticides application to control agricultural pests as it is the ultimate receptor of such materials. This study evaluated the impact of new metal-insecticide, the [Mg(hesp)₂(phen)], referred as MgHP, on fish using physiological, genetic, biochemical, and morphological biomarkers. The fish, Prochilodus lineatus, was exposed to 0 (control), 1, 10, 100, 1000 μg L⁻¹ MgHP, for 24 and 96 h. MgHP was not lethal but caused genotoxicity, altered hematological variables and, the activity of antioxidant and biotransformation enzymes and histology of liver, depending on concentration and time exposure. Hematocrit and erythrocyte number (RBC) increased without change hemoglobin content resulting in changes in hematimetric indexes after 24 h; after 96 h, only RBC was changed. Erythrocyte nuclear abnormalities and crenate cells increased after 24 h but, not after 96 h. Erythrocytes and hepatocytes indicated instability in DNA integrity however, the absence of micronuclei suggested DNA damage repairment. After 24 h, the antioxidant defense system and the phase II biotransformation enzyme was responsiveness and catalase activity decreased at high MgHP concentrations; the antioxidant response was triggered after 96 h. Hepatocyte hypertrophy, intracellular cytoplasmic substances, cytoplasm degeneration, melanomacrophage and hyperemia increased in fish exposed from 10 μg L⁻¹ to higher MgHP concentrations; the organ alteration index increased as MgHP concentration increased showing dose-dependence. Most of hematological and genotoxic effects occurred after 24 h exposure evidencing potential recover capability of organism by activation of the antioxidant defense system and DNA repairment mechanisms. Nevertheless, the histopathological changes in the liver was maintained over time at high MgHP concentrations, a concentration usually no environmental relevant. In conclusion, this data reinforced the importance of continuing research on MgHP effects in other organisms considering the promising use of such compound to control the leaf-cutter ants and other insects.

Fish cultivation in rice fields is a valuable resource in some rural areas of the world. Fish is a source of protein and an additional source of income for local farmers. However, the use of pesticides may impact fish and consumer health. The aim of this study was to evaluate exposure and effect biomarkers in native fish inhabiting a rice field during a production cycle. Samples of fish, water and sediment from a rice field in Santa Fe, Argentina were collected during a cultivation season (at the beginning: November 2017, in the middle: December 2017 and at the end: February 2018). At each sampling period, fish biomarkers of effect (biometric indices, hematological parameters, energy reserves, oxidative stress and neurotoxicity) were assessed together with pesticide screening in water, sediment, and fish samples. Only herbicides were present in water and sediment samples in agreement with land treatment before rice sowing stage, where only herbicides were applied. In general, the greatest water concentrations of bentazone, glyphosate and aminomethylphosphonic acid (AMPA), and the lowest sediment glyphosate and AMPA levels were observed at the beginning of the farming cycle. Fish bioaccumulated AMPA residues at all sampling periods and showed biological responses to cope with a stressful environment. Alterations in hematological parameters, mobilization of energetic reserves and activation of the antioxidant system were detected. However, no oxidative damage nor neurotoxic effects were present along the production cycle. Under a real exposure scenario, the present work demonstrates that biological changes are induced in fish to cope with stressors present in a rice field. Fish-rice coculture is an efficient and ecologically sustainable approach to increase food supplies, and a better understanding of the effect of this particular environment on fish would allow a greater and safer development of this promising productive activity in South American rice producing countries.

Adult coho salmon (Oncorhynchus kisutch) prematurely die when they return from the ocean to spawn in urban watersheds throughout northwestern North America. The available evidence suggests the annual mortality events are caused by toxic stormwater runoff. The underlying pathophysiology of the urban spawner mortality syndrome is not known, and it is unclear whether closely related species of Pacific salmon are similarly at risk. The present study co-exposed adult coho and chum (O. keta) salmon to runoff from a high traffic volume urban arterial roadway. The spawners were monitored for the familiar symptoms of the mortality syndrome, including surface swimming, loss of orientation, and loss of equilibrium. Moreover, the hematology of both species was profiled by measuring arterial pH, blood gases, lactate, plasma electrolytes, hematocrit, and glucose. Adult coho developed behavioral symptoms within a few hours of exposure to stormwater. Various measured hematological parameters were significantly altered compared to coho controls, indicating a blood acidosis and ionoregulatory disturbance. By contrast, runoff-exposed chum spawners showed essentially no indications of the mortality syndrome, and measured blood hematological parameters were similar to unexposed chum controls. We conclude that contaminant(s) in urban runoff are the likely cause of the disruption of ion balance and pH in coho but not chum salmon. Among the thousands of chemicals in stormwater, future forensic analyses should focus on the gill or cardiovascular system of coho salmon. Because of their distinctive sensitivity to urban runoff, adult coho remain an important vertebrate indicator species for degraded water quality in freshwater habitats under pressure from human population growth and urbanization.

Exposure to essential and non-essential elements may be elevated for green sea turtles (Chelonia mydas) that forage close to shore. Biomonitoring of trace elements in turtle blood can identify temporal trends over repeated sampling events, but any interpretation of potential health risks due to an elevated exposure first requires a comparison against a baseline. This study aims to use clinical reference interval (RI) methods to produce exposure baseline limits for essential and non-essential elements (Na, Mg, K, Ca, Ti, V, Cr, Mn, Fe, Co, Ni, Cu, Zn, As, Se, Mo, Cd, Sb, Ba, and Pb) using blood from healthy subadult turtles foraging in a remote and offshore part of the Great Barrier Reef. Subsequent blood biomonitoring of three additional coastal populations, which forage in areas dominated by agricultural, urban and military activities, showed clear habitat-specific differences in blood metal profiles relative to the those observed in the offshore population. Coastal turtles were most often found to have elevated concentrations of Co, Mo, Mn, Mg, Na, As, Sb, and Pb relative to the corresponding RIs. In particular, blood from turtles from the agricultural site had Co concentrations ranging from 160 to 840 μg/L (4–25 times above RI), which are within the order expected to elicit acute effects in many vertebrates. Additional clinical blood biochemistry and haematology results indicate signs of a systemic disease and the prevalence of an active inflammatory response in a high proportion (44%) of turtles from the agricultural site. Elevated Co, Sb, and Mn in the blood of these turtles significantly correlated with elevated markers of acute inflammation (total white cell counts) and liver dysfunction (alkaline phosphatase and total bilirubin). The results of this study support the notion that elevated trace element exposures may be adversely affecting the health of nearshore green sea turtles.

Toxic elements (arsenic, cadmium, lead, mercury, selenium, thallium) are a group of contaminants that are known to elicit developmental, reproductive, general health, and immune system effects in reptiles, even at low concentrations. Reptiles, including marine turtles, are susceptible to accumulation of toxic elements due to their long life span, low metabolic rate, and highly efficient conversion of prey into biomass. The objectives of this study were to (1) document concentrations of arsenic, cadmium, lead, mercury, selenium, and thallium in whole blood and keratin from nesting loggerhead sea turtles (Caretta caretta) from Casey Key, Florida and document correlations thereof and (2) correlate whole blood toxic element concentrations to various hematological and plasma biochemistry analytes. Baselines for various hematological and plasma analytes and toxic elements in whole blood and keratin (i.e., scute) in nesting loggerheads are documented. Various correlations between the toxic elements and hematological and plasma biochemistry analytes were identified; however, the most intriguing were negative correlations between arsenic, cadmium, lead, and selenium with and α- and γ-globulins. Although various extrinsic and intrinsic variables such as dietary and feeding changes in nesting loggerheads need to be considered, this finding may suggest a link to altered humoral immunity. This study documents a suite of health variables of nesting loggerheads in correlation to contaminants and identifies the potential of toxic elements to impact the overall health of nesting turtles, thus presenting important implications for the conservation and management of this species.

Trace elements are pollutants of both natural and anthropogenic origin which can influence negatively on ecosystem and wildlife health. We evaluated trace element in blood samples of gray seal (Halichoerus grypus) stranded in the Gulf of Riga and their influence on their health status through hematological and biochemical profiles. Zn showed the highest levels followed by Cu > Se > Pb > THg > As. Cr and Cd were not detected. Most trace element levels were generally comparable to those reported in seal species; however, high Pb values were observed in those sample showing detectable concentrations (<0.046–257.6 μg/kg ww). Significant positive correlations were found between trace elements concentrations and various biochemical parameters, including Se-ASAT, Se:Hg-ASAT, Cu-TP, Cu-ALB, CuCa, Zn-ALAT, ZN-LDH, ZnP, Zn-Segment neutrophils, and Pb-CK. Nevertheless, most relationships were not strong enough (p > 0.04) to assume a toxicological implication. Despite its limitations, this information could serve as the baseline for future research.

The present field study aimed to determine the extent of pollution in the Red Sea coast of Yemen Republic using a battery of biomarkers in sea catfish, Arius thalassinus, originating from a reference site in comparison with a polluted site. We reported the concentration of heavy metals in some vital fish organs and their effects on the morphological, hematological, biochemical and oxidative stress biomarkers accompanied by the examination of histopathological alterations. The obtained results showed clear signs of stress in fish from a polluted site. Linear correlation analysis exhibited that the biomarkers response could be linked to the detected metals bioaccumulation. In addition, principal component analysis showed a clear separation of sampling sites in two different assemblages. Semi-quantitative analysis for the observed histopathological lesions revealed that gills were the most affected organs with signs of severe alterations. This field investigation provides a baseline data on pollution status in this region.

Palmyra Atoll, in the Central Pacific, is a unique marine ecosystem because of its remarkably intact food web and limited anthropogenic stressors. However during World War II the atoll was structurally reconfigured into a military installation and questions remain whether this may have impacted the health of the atoll’s ecosystems and species. To address the issue we assessed green sea turtle (n=157) health and exposure to contaminants at this foraging ground from 2008 to 2012. Physical exams were performed and blood was sampled for testosterone analysis, plasma biochemistry analysis, hematology and heavy metal exposure. Hematological and plasma chemistries were consistent with concentrations reported for healthy green turtles. Heavy metal screenings revealed low concentrations of most metals, except for high concentrations of iron and aluminum. Body condition indices showed that <1% of turtles had poor body condition. In this study, we provide the first published blood values for a markedly healthy sea turtle population at a remote Central Pacific Atoll.
