Metronidazole enhances steatosis-related early-stage hepatocarcinogenesis in high fat diet-fed rats through DNA double-strand breaks and modulation of autophagy

Nonalcoholic fatty liver disease is a hepatic disorder with deposition of fat droplets and has a high risk of progression to steatosis-related hepatitis and irreversible hepatic cancer. Metronidazole (MNZ) is an antiprotozoal and antimicrobial agent widely used to treat patients infected with anaerobic bacteria and intestinal parasites; however, MNZ has also been shown to induce liver tumors in rodents. To investigate the effects of MNZ on steatosis-related early-stage hepatocarcinogenesis, male rats treated with N-nitrosodiethylamine following 2/3 hepatectomy at week 3 were received a control basal diet, high fat diet (HFD), or HFD containing 0.5% MNZ. The HFD induced obesity and steatosis in the liver, accompanied by altered expression of Pparg and Fasn, genes related to lipid metabolism. MNZ increased nuclear translocation of lipid metabolism-related transcription factor peroxisome proliferator-activated receptor gamma in hepatocytes, together with altered liver expression of lipid metabolism genes (Srebf1, Srebf2, Pnpla2). Furthermore, MNZ significantly increased the number of preneoplastic liver foci, accompanied by DNA double-strand breaks and late-stage autophagy inhibition, as reflected by increased levels of γ-H2AX, LC3, and p62. Therefore, MNZ could induce steatosis-related hepatocarcinogenesis by inducing DNA double-strand breaks and modulating autophagy in HFD-fed rats.

Availability of clean and safe freshwater has become a looming global concern. The accelerated demography, industrialization, and climate changes contaminate the meager freshwater reserves. Pollution of water bodies is significantly detrimental to health, ecology, economy, and society. The rising number of malnutrition cases, stunted growth, hepatitis, gastroenteritis, skin ailments, cholera, respiratory disorders, liver malfunction, eye infections, and mortality have been attributed to exposure to compromised water. Thus, optimized, durable, and inexpensive wastewater treatment and remediation processes are necessary. Current conventional treatment strategies suffer from several drawbacks, which may be mitigated through nanotechnological intercession, promising sustainability. Nanomaterials include nanosorbents, carbon nanotubes, nanocomposites, nanofibers, graphene, nanodendrimers, nanomembranes, and nanocatalysts. They have unique properties that make attractive alternatives for wastewater remediation, purification, and contamination detection through pollutant-specific nanosensors and detectors. This review discusses water pollution, its impacts, conventional treatment strategies, nanotechnological contributions, venture possibilities, and associated commercial opportunities.

Nowadays, water-borne diseases including hepatitis remain the critical health challenge due to the inadequate supply of potable and safe water for human activities. The major cause is that the pathogenic microorganisms causing diseases have developed resistance against common techniques used by sewage water treatment plants for water disinfection. Therefore, there is a need to improve these conventional water treatment techniques by taking into consideration the application of nanotechnology for wastewater purification. The main aim of this paper is to provide a review on the synthesis of biopolymer-inorganic nanoparticle composites (BINCs), their used as antimicrobial compounds for water disinfection, as well as to elaborate on their antimicrobial mechanism of action. The microbial properties affecting the activity of antimicrobial compounds are also evaluated.

Water pollution by recalcitrant compounds is an increasingly important problem due to the continuous introduction of new chemicals into the environment. Choosing appropriate measures and developing successful strategies for eliminating hazardous wastewater contaminants from industrial processes is currently a primary goal. Electroplating industry wastewater involves highly toxic cyanide (CN), heavy metal ions, oils and greases, organic solvents, and the complicated composition of effluents and may also contain biological oxygen demand (BOD), chemical oxygen demand (COD), SS, DS, TS, and turbidity. The availability of these metal ions in electroplating industry wastewater makes the water so toxic and corrosive. Because these heavy metals are harmful to living things, they must be removed to prevent them from being absorbed by plants, animals, and humans. As a result, exposure to electroplating wastewater can induce necrosis and nephritis in humans and lung cancer, digestive system cancer, anemia, hepatitis, and maxillary sinus cancer with prolonged exposure. For the safe discharge of electroplating industry effluents, appropriate wastewater treatment has to be provided. This article examines and assesses new approaches such as coagulation and flocculation, chemical precipitation, ion exchange, membrane filtration, adsorption, electrochemical treatment, and advanced oxidation process (AOP) for treating the electroplating industry wastewater. On the other hand, these physicochemical approaches have significant drawbacks, including a high initial investment and operating cost due to costly chemical reagents, the production of metal complexes sludge that needs additional treatment, and a long recovery process. At the same time, advanced techniques such as electrochemical treatment can remove various kinds of organic and inorganic contaminants such as BOD, COD, and heavy metals. The electrochemical treatment process has several advantages over traditional technologies, including complete removal of persistent organic pollutants, environmental friendliness, ease of integration with other conventional technologies, less sludge production, high separation, and shorter residence time. The effectiveness of the electrochemical treatment process depends on various parameters, including pH, electrode material, operation time, electrode gap, and current density. This review mainly emphasizes the removal of heavy metals and another pollutant such as CN from electroplating discharge. This paper will be helpful in the selection of efficient techniques for treatment based on the quantity and characteristics of the effluent produced.

The Santos and São Vicente Estuary has suffered extensively over the years from irregular industrial deposits. The present study aimed to evaluate liver disease prevalence and potential associated risk factors in four of the Estuary’s areas (Pilões and Água-Fria, Cubatão Center, Continental São Vicente, and Guarujá) and a reference area (Bertioga). This study consisted of a cross-sectional study design, in which a questionnaire was used to collect information in 820 households at each of the study areas. The proportion of total liver diseases, hepatitis, cirrhosis, and cancer (liver, biliary tract, and pancreas) per area were estimated. Pearson’s chi-square test and two proportion differences test were applied in order to evaluate associations between disease occurrence and areas and to test differences between two proportions, respectively. Single and multiple logistic regression models were applied to assess associations between disease prevalence and the different study areas. Liver disease prevalence was 1.5 % among all inhabitants and 1.4 % among those without any type of exposure. Among those who reported the presence of liver disease, a higher percentage of the participants that reported hepatitis (27.7 %) or other liver disease (48.7 %) did not report occupational or alcohol exposures. Hepatitis (77.8 %) was the most reported disease, and a statistical association between living in Pilões and Água-Fria and the occurrence of hepatitis was observed (Pearson’s χ ²: z = 18.1; p = 0.001). The consumption of locally-produced groceries (2.88; CI: 1.24–6.70) and water (5.88; CI: 2.24–15.45) were shown to be risk factors for the occurrence of liver disease. Thus, environmental exposure is still a public health problem present in the estuary region.

Congenital heart disease (CHD) is the most common birth defect. It is due to dysfunction of the heart and great vessels during embryo development stage, or the channel was not closed after birth. This study focuses on investigating the influence of virus infection and medicine history during pregnancy on the incidence rate of CHD of fetus. We conducted a retrospective birth cohort study of infant born in the maternal and child health hospital of Fanyu district in Guangzhou. Five thousand three hundred eighty one cases with complete medical records, including mothers, fathers, and infants, were enrolled. The exposure history of mothers to virus and medicine from 6 months before pregnancy to prenatal examination was investigated, including mflu, mumps, measles, rubella, chickenpox, and hepatitis and antibiotics, tocolytic agent, anticonvulsants, antipyretic and analgesic, antitumor drug, folic acid supplement, and contraceptive. The relationship between virus infection and medicine history during pregnancy and CHD was analyzed. There was statistical difference between a normal group and a defected group in influenza infection and tocolytic agent and contraceptive pill. The exposure history to influenza and medicines, such as tocolytic agent and contraceptive pill, during pregnancy influenced the incidence rate of CHD of fetus.
