Development of chitosan-coated nanoemulsions of two sulfides present in onion (Allium cepa) essential oil and their nematicidal activities against the pine wood nematode, Bursaphelenchus xylophilus

Pine wood nematode, Bursaphelenchus xylophilus, is a plant parasitic nematode which causes severe damage to several Pinus species. Two natural compounds, dipropyl trisulfide (DPTS) and methyl propyl trisulfide (MPTS), showed strong nematicidal activity against the pine wood nematode, presenting 4.24 and 17.81 μg/mL LC₅₀ values, respectively. However, hydrophobicity and low stability have limited their practical use in the field as nematicides. To overcome these problems, chitosan-coated nanoemulsions of DPTS and MPTS were developed. The optimum chitosan concentration for the delivery system of the two sulfides was 0.5%. Optimized chitosan-coated nanoemulsions of sulfides have a uniform size distribution (mean diameter = 203.7 and 207.7 nm, mean polydispersity index = 0.176 and 0.178) with sufficient colloidal stability (mean zeta potential = +40 and +45 mV). The LC₅₀ values of DPTS and MPTS nanoemulsions coated with 0.5% chitosan against the pine wood nematode were 5.01 and 16.60 μg/mL, respectively. In addition, chitosan coating improved the long-term storage stability and persistence of nematicidal activity of the nanoemulsions. This study indicates that the chitosan-coated nanoemulsion is a suitable formulation for sulfides as novel nematicides against the pine wood nematode for field application.

The essential oil (EO) of Cupressus sempervirens was obtained by hydrodistillation and analysed using gas chromatography–flame ionization detection (GC–FID) and gas chromatography–mass spectrometry (GC–MS). Two monoterpenes, α-pinene (49.1%) and δ-3-carene (21.4%), and one sesquiterpene hydrocarbon, α-cedrol (5.1%), were isolated as the EO major terpenes. An oil-in-water nanoemulsion (particle size 71.2 nm) was produced from the EO through a low-energy method. The EO, its nanoemulsion and its main constituents showed mosquitocidal and biochemical effects against Culex quinquefasciatus Say, the common vector of lymphatic filariasis parasites. All treatments showed dose-dependent bioactivity, and adults were more susceptible to the EO products than the larvae. The nanoemulsion showed superior activity, followed by the crude EO and α-cedrol. At 40 μg/ml, the nanoemulsion caused 100% larval mortality, while the EO and α-cedrol required twice this concentration to achieve the same larval mortality. The LC₅₀ values were 8.4, 16.1, 15.1, 30.7 and 53.4 μg/ml at 24 h after exposure for the nanoemulsion, crude oil, α-cedrol, δ-3-carene and α-pinene, respectively. For adults, 20.0 μl/l nanoemulsion caused 100% mortality, while twice this concentration of the EO was required to achieve the same effect. The LC₅₀’s against adults ranged between 6.2 and 40.4 μl/l. EO products prominently repelled mosquitoes at concentrations between 0.75 and 6.0 μl/cm². The EO products caused remarkable inhibition of Cx. quinquefasciatus acetylcholinesterase activity but were safer towards the non-target aquatic species Gambusia affinis. These results recommend the use of C. sempervirens EO, its nanoemulsion and main terpenes as natural tools to control Cx. quinquefasciatus.

The essential oil (EO) was hydrodistilled from of Deverra tortuosa aerial parts. Fifty-six components amounting 99.3% were identified in EO through using gas chromatography–flame ionization detection (GC–FID) and (GC–MS). Phenylpropanoids, dillapiole (41.6%), elemicin (7.3%) and myristicin (5.1%), and the monoterpene, sabinene (4.2%) were identified as the major terpenes. An oil-in-water nanoemulsion (particle size 70.3 nm) was developed from EO adopting a low-energy method. The EO products showed insecticidal and biochemical effects against the cowpea weevil Callosobruchus maculatus. Based on a 48-h exposure period, the oil nanoemulsion exhibited a superior contact bioactivity (LC₅₀ = 10.3 µg/cm²), followed by EO (LC₅₀ = 23.1 µg/cm²), dillapiole (LC₅₀ = 27.8 µg/cm²), and myristicin (LC₅₀ = 37.1 µg/cm²). Upon fumigation, nanoemulsion and EO were superior as fumigants (LC₅₀ after 48 h were 6.9 and 14.3 µl/l, respectively). Test materials showed a residual bioactivity against C. maculatus, where EO, dillapiole, and myristicin showed the strongest grain protecting activity. EO products significantly inhibited acetylcholinesterase (AChE) activity of C. maculatus adults. Test products were safe toward the non-target earthworms and did not alter the viability of cowpea seeds. There are evidences for the potential of using EO of D. tortuosa and its nanoemulsion and phenylpropanoids as natural grain protectants against C. maculatus.

With continuous development of pesticide dosage forms, emulsifiable concentrates using large amounts of organic solvents are gradually obsoleted. Nanoemulsions with high water content have been developed and the preparation processes also evolved, but these processes still exist some problems, such as poor controllability and high energy consumption. Microfluidic is a controllable nanoemulsion preparation system which mainly applied to pharmaceutical synthesis. In this study, the pesticide phoxim nanoemulsion was prepared by microfluidic technology. The optimized formulation of phoxim nanoemulsion was composed of Tween 80 and pesticide emulsifier 500 as surfactant, hexyl acetate as oil, and n-propanol as co-surfactant. Moreover, when the flow rates of water and oil in the microfluidic system were adjusted to 5 μL/min and 20 μL/min, phoxim nanoemulsion was obtained with a cloud point/boiling point of 109 °C, a particle size of 21.5 ± 0.8 nm and a potential value of − 18.7 ± 0.6 mV. Furthermore, the nanoemulsion had a rapid release effect in vitro which could be fitted by the Ritger-Peppas model. The feeding toxicity of the phoxim nanoemulsion was higher than that of commercial formulation while the contact killing effect was higher than that of the active ingredient. Therefore, pesticide dosage was reduced and the insecticidal effect was enhanced by using phoxim nanoemulsions. These results also confirm the potential of microfluidics as a green process to produce pesticide nanoemulsions.

Aedes aegypti is the main vector of yellow fever, chikungunya, Zika, and dengue worldwide and is managed by using chemical insecticides. Though effective, their indiscriminate use brings in associated problems on safety to non-target and the environment. This supports the use of plant-based essential oil (EO) formulations as they are safe to use with limited effect on non-target organisms. Quick volatility and degradation of EO are a hurdle in its use; the present study attempts to develop nanoemulsions (NE) of Trachyspermum ammi EO and its constituent thymol using Tween 80 as surfactant by ultrasonication method. The NE of EO had droplet size ranging from 65 ± 0.7 to 83 ± 0.09 nm and a poly dispersity index (PDI) value of 0.18 ± 0.003 to 0.20 ± 0.07 from 1 to 60 days of storage. The NE of thymol showed a droplet size ranging from 167 ± 1 to 230 ± 1 nm and PDI value of 0.30 ± 0.03 to 0.40 ± 0.008 from 1 to 60 days of storage. The droplet shape of both NEs appeared spherical under a transmission electron microscope (TEM). The larvicidal effect of NEs of EO and thymol was better than BEs (Bulk emulsion) of EO and thymol against Ae. aegypti. Among the NEs, thymol (LC₅₀ 34.89 ppm) had better larvicidal action than EO (LC₅₀ 46.73 ppm). Exposure to NEs of EO and thymol causes the shrinkage of the larval cuticle and inhibited the acetylcholinesterase (AChE) activity in Ae. aegypti. Our findings show the enhanced effect of NEs over BEs which facilitate its use as an alternative control measure for Ae. aegypti.

Meningitis is an inflammation of the protective membranes called meninges and fluid adjacent the brain and spinal cord. The inflammatory progression expands all through subarachnoid space of the brain and spinal cord and occupies the ventricles. The pathogens like bacteria, fungi, viruses, or parasites are main sources of infection causing meningitis. Bacterial meningitis is a life-threatening health problem that which needs instantaneous apprehension and treatment. Nesseria meningitidis, Streptococcus pneumoniae, and Haemophilus flu are major widespread factors causing bacterial meningitis. The conventional drug delivery approaches encounter difficulty in crossing this blood-brain barrier (BBB) and therefore are insufficient to elicit the desired pharmacological effect as required for treatment of meningitis. Therefore, application of nanoparticle-based drug delivery systems has become imperative for successful dealing with this deadly disease. The nanoparticles have ability to across BBB via four important transport mechanisms, i.e., paracellular transport, transcellular (transcytosis), endocytosis (adsorptive transcytosis), and receptor-mediated transcytosis. In this review, we reminisce distinctive symptoms of meningitis, and provide an overview of various types of bacterial meningitis, with a focus on its epidemiology, pathogenesis, and pathophysiology. This review describes conventional therapeutic approaches for treatment of meningitis and the problems encountered by them while transmitting across tight junctions of BBB. The nanotechnology approaches like functionalized polymeric nanoparticles, solid lipid nanoparticles, nanostructured lipid carrier, nanoemulsion, liposomes, transferosomes, and carbon nanotubes which have been recently evaluated for treatment or detection of bacterial meningitis have been focused. This review has also briefly summarized the recent patents and clinical status of therapeutic modalities for meningitis.

The present work aimed to remove azithromycin (AZM) from the contaminated aqueous system using a water/ethanol/transcutol/Capryol-90 green nanoemulsion. The drug is identified as a potential pharmaceutical contaminant detrimental for flora and fauna of aquatic lives as well as human health. Green nanoemulsions were tailored and characterized for thermodynamic stability, size, polydispersity index (PDI), zeta potential, viscosity, refractive index (RI), and morphological assessment using a transmission electron microscopy (TEM). Moreover, nanoemulsions were investigated for percent removal efficiency (%RE) and factors affecting percent removal efficiency (%RE). The results suggested that the developed green nanoemulsions (ANE1–ANE5) were transparent (˂ 200 nm) and stable. ANE5 exhibited the lowest value of globular size (49 nm), PDI (0.17), viscosity (~ 93 cP), and optimum zeta potential (−27.8 mV). The value of %RE depended upon the content of water and Capryol-90 of the nanoemulsion. Furthermore, the value of %RE was found to be increased with increased content of water, whereas this was decreased on increasing the Capryol-90 content in the nanoemulsions. Similarly, on decreasing the values of size and viscosity, the %RE values were observed to be increased. There was insignificant impact of the duration of exposure time on %RE. Thus, the maximum %RE value (96.8%) was obtained by ANE5 from the aqueous solution after 20 min of contact time with ANE5. Thus, this method could be a promising approach to remove AZM from the contaminated water and serve as an alternative to conventional methods.

The control of storage insect pests is largely based on synthetic pesticides. However, due to fast growing resistance in the targeted insects, negative impact on humans and non-target organisms as well as the environment, there is an urgent need to search some safer alternatives of these xenobiotics. Many essential oils (EOs) and their bioactive compounds have received particular attention for application as botanical pesticides, since they exhibited high insecticidal efficacy, diverse mode of action, and favourable safety profiles on mammalian system as well as to the non-target organisms. Data collected from scientific articles show that these EOs and their bioactive compounds exhibited insecticidal activity via fumigant, contact, repellent, antifeedant, ovicidal, oviposition deterrent and larvicidal activity, and by inhibiting/altering important neurotransmitters such as acetylcholine esterase (AChE) and octopamine or neurotransmitter inhibitor γ-amino butyric acid (GABA), as well as by altering the enzymatic [superoxide dismutase (SOD), catalase (CAT), peroxidases (POx), glutathione-S-transferase (GST) and glutathione reductase (GR)] and non-enzymatic [glutathione (GSH)] antioxidant defence systems. However, in spite of promising pesticidal efficacy against storage pests, the practical application of EOs and their bioactive compounds in real food systems remain rather limited because of their high volatility, poor water solubility and susceptibility towards degradation. Nanoencapsulation/nanoemulsion of EOs is currently considered as a promising tool that improved water solubility, enhanced bio-efficacy, stability and controlled release, thereby expanding their applicability.

In the present scenario, the utilization of petroleum fuel is expanding forcefully worldwide in the vitality store and plays a highly hazardous role in the ecological system. Biofuel stands out among the most tenable keys for this issue. The lemongrass oil is used as a biofuel because of low density and viscosity when compared with diesel. The lemongrass oil is extracted by steam distillation process. In the present investigation, partially stabilized zirconium, due to its higher thermal conductivity, is selected as coating material. The top surface of the piston and the inlet and exhaust valves are coated up to the preferred thickness of 500 μm by the plasma spray technique. The lemongrass emulsion fuel is prepared in the proportion of 94% of lemongrass oil, 5% of water, and 1% of surfactant span 80. The nanoparticles of cerium oxide were used with lemongrass oil (LGO) nano-emulsion in the measurement of 30 ppm. The four-stroke diesel engine execution, ignition, and the outflow extent were contrasted in the diesel and lemongrass oil (LGO) compared with the base diesel engine. The performance characteristic curves of lemongrass-cerium oxide nano-emulsion fuel show the increase in brake thermal efficiency of 17.21% when compared with the mineral diesel fuel. The emission characteristics of lemongrass-cerium oxide nano-emulsion fuel show a drop in hydrocarbon and carbon monoxide emission by 16.21% and 15.21%, respectively, when compared with base diesel fuel and also there is a decrease in oxides of nitrogen and smoke emission by 24.1% and 6.3%, respectively, when compared to mineral diesel fuel.

Using organic insecticides including plant oils, it is possible to design a new perspective for the control of insect pests. In this research, nanoemulsion formulations of Mentha piperita, wild-type essential oil (EO) were prepared utilizing high-energy ultrasonication process. Physicochemical properties of nanoemulsions were precisely studied by measurement various parameters including pH, viscosity, conductivity, and zeta potential. Experimental design by the aid of response surface methodology (RSM) was used to highlight the physicochemical roles of EO percentage (1% to 5% (v/v)) and surfactant concentration (3% to 15% (v/v)) for achieving minimum droplet diameter with high physical stability. The nanoemulsion formulations were then characterized using dynamic light scattering, transmission electron microscopy, and optical clarity. Afterward, an appropriate model between the variable factors (EO percentage and surfactant concentration) and the response (hydrodynamic particle size) was statistically developed. Under the optimum conditions, nanoemulsion with hydrodynamic particle size less than 10 nm with high physical stability is obtainable. Bioassay experiments were carried out to elucidate the effects of nanoemulsion on the cotton aphid. Synthesized nanoemulsion formulations showed relatively high contact toxicity (average value of LC₅₀ was about 3879.5 ± 16.2 μl a.i./L) against the pest. On the basis of the obtained results, prepared nanoemulsion using M. piperita is potentially applicable as organic insecticides against cotton aphid. Graphical abstract
