The interference of nonylphenol (NP) with humans and animals, especially in hormone systems, has been well-studied. There is rarely any record of its effect on bacteria, which dominate in various environments. In our study, we employed Pseudomonas aeruginosa PAO1 as a model microorganism and took its common lifestyle biofilm, mainly regulated by quorum sensing (QS), as a cut-in point to investigate the effect of NP (1, 5, 10 mg L⁻¹) on bacteria. The results showed that more than 5 mg L⁻¹ of NP did interfere with biofilm formation and affected bacterial QS. In detail, the LasI/R circuit, but not the RhlI/R circuit, was considerably obstructed. The decrease in lasI and lasR expression resulted in a significant reduction in N-3-oxo-dodecanoyl homoserine lactone (3OC₁₂-HSL) signals and the downstream production of elastases. Docking results indicated the binding of NP with LasR protein, simulating the binding of 3OC₁₂-HSL with LasR protein, which explained the obstruction of the LasIR circuit. We concluded that NP competed with 3OC₁₂-HSL and blocked 3OC₁₂-HSL binding with the LasR protein, resulting in a direct interference in bacterial biofilm formation. This is the first report of NP interference with bacterial signaling, which is not only helpful to understand the effect of NP on various ecosystems, but is also beneficial to enrich our knowledge of inter-kingdom communication.

The release of hexavalent chromium [Cr(VI)] into water bodies poses a major threat to the environment and human health. However, studies of the biological response to Cr(VI) are limited. In this study, a toxic bacterial mechanism of Cr(VI) was investigated using Pannonibacter phragmitetus BB (hereafter BB), which was isolated from chromate slag. The maximum Cr(VI) concentrations with respect to the resistance and reduction by BB are 4000 mg L−1 and 2500 mg L−1, respectively. In the BB genome, more genes responsible for Cr(VI) resistance and reduction are observed compared with other P. phragmitetus strains. A total of 361 proteins were upregulated to respond to Cr(VI) exposure, including enzymes for Cr(VI) uptake, intracellular reduction, ROS detoxification, DNA repair, and Cr(VI) efflux and proteins associated with novel mechanisms involving extracellular reduction mediated by electron transfer, quorum sensing, and chemotaxis. Based on metabolomic analysis, 174 metabolites were identified. Most of the upregulated metabolites are involved in amino acid, glucose, lipid, and energy metabolisms. The results show that Cr(VI) induces metabolite production, while metabolites promote Cr(VI) reduction. Overall, multi-enzyme expression and metabolite production by BB contribute to its high ability to resist/reduce Cr(VI). This study provides details supporting the theory of Cr(VI) reduction and a theoretical basis for the efficient bioremoval of Cr(VI) from the environment.

Noble metal–based nanomaterials (NMNs), such as platinum nanoparticles (Pt@NPs) and palladium nanoparticles (Pd@NPs), are increasingly being used as antibacterial agents. However, little information is available on bacterial resistance to NMNs. In this study, owing to their oxidase-like and peroxidase-like properties, both Pt@NPs and Pd@NPs induce reactive oxygen species (ROS) and manifest antibacterial activities: 6.25 μg/mL of either Pt@NPs or Pd@NPs killed >50% of Staphylococcus aureus strain ATCC29213. However, Pseudomonas aeruginosa strain PAO1 completely resisted 12.5 μg/mL of Pt@NPs and 6.25 μg/mL of Pd@NPs. Compared to the non-NMN groups, these NMNs promoted 2–3-fold upregulation of the quorum sensing (QS) gene lasR in strain PAO1. In fact, the lasR gene upregulation induced a 1.5-fold reduction in ROS production and increased biofilm formation by 11% (Pt@NPs) and 27% (Pd@NPs) in strain PAO1. The ΔlasR mutants (lasR gene knock out in strain PAO1), became sensitive to NMNs. The survival rates of ΔlasR mutants at 12.5 μg/mL Pt@NPs and Pd@NPs treatments were only 77% and 58%, respectively. This is the first report indicating that bacteria can resist NMNs through QS. Based on these results, evaluation of the ecological risks of using NMNs as antibacterial agents is necessary.

Bacterial quorum sensing signal molecules N-acyl homoserine lactones (AHLs) (C10-HSL, 3-OXO-C10-HSL and 3-OH-C10-HSL) as possible chemical cues were employed to investigate the role in the formation of fouling diatom-biofilm (Cylindrotheca sp.). Results showed that AHLs promoted Chlorophyll a (Chl.a) and extracellular polymeric substance (EPS) contents in the diatom-biofilm. In the presence of AHLs-inhibitor 3, 4-Dibromo-2(5)H-furanone, which was used to avoid the possible interference of AHLs from bacteria, AHLs also increased the Chl.a and EPS contents. Scanning electron microscope and confocal laser scanning microscope analysis further demonstrated that AHLs promoted the formation of the diatom-biofilm. Non-invasive micro-test technique showed that AHLs promoted Ca2+ efflux in Cylindrotheca sp., which implied that Ca2+ might be correlated with AHLs-induced positive effect on the formation of diatom-biofilm. This study provides direct evidences that AHLs play an important role in developing the diatom-biofilm and AHLs-inhibitors might be promising active agents in marine antifouling.

Hydrodynamic cavitation treatment was used for the functional inactivation of quorum-sensing lactone molecules of Pseudomonas aeruginosa. Hydroxyl radicals formed as well as the shear effects during the cavitation process induced the inactivation of the signal molecules through hydrolysis reaction coupled with bacterial destruction. Concentration of two different types of homoserine lactones (HSL) molecules was tested after the treatment at various rotational speeds. It was found that the strongest effects can be achieved at speeds > 2000 rpm. This value is considered as an onset speed of dominant cavitation, and it is in agreement with literature data. The experimental trends were in agreement with the calculations based on the finite element modelling, which show a significant increase in average shear stress at higher rotational speeds. Overall, the work has demonstrated the possible effects of hydrodynamic cavitation on the quorum-sensing molecules of Pseudomonas aeruginosa for the first time.

The cell density-dependent gene expression in gram-negative bacteria is through the activity of acyl homoserine lactone signal molecules. The novel strain Pseudomonas putida JMQS1 isolated from detergent-contaminated soil exhibited quorum sensing along with its ability to degrade phenol. The response to Chromobacterium violaceum DSTS-1 mutant biosensor and luxI and luxR gene-specific amplification was used to characterize the quorum sensing property of the isolate. A regulation in the synthesis of various acyl homoserine lactone molecules, viz C₆HSL in the initial stages of phenol stress, C₈HSL during degradation, and 3OC₁₂HSL on completion of degradation could be identified by liquid chromatography-quadrupole time of flight. Thin-layer chromatography, Fourier transform infrared spectroscopy, and gas chromatography mass spectrometry confirmed the complete degradation of phenol in 48–56 h. P. putida JMQS1 exhibited adaptation over phenol stress through the selective activation of the quorum sensing signal molecules depending on the changing physiological conditions. This study proposes an efficient method for enhancing the degradation of toxic organic pollutants by the supplementation of acyl homoserine lactone signal molecules.

Chlorella vulgaris and Bacillus licheniformis consortium was added to synthetic wastewater with exogenous vitamin B₁₂. In the presence of 100 ng/L vitamin B₁₂, removal efficiencies of TN, NH₃-N, PO₄³—P, and COD were 80.1%, 76.8%, 87.9%, and 76.7%, respectively. The functional groups on the cell surface of the consortium, including -NH, -CH₃, C=O, C=C, and P-O-C, increased with 100 ng/L vitamin B₁₂. These functional groups improved the biological adsorption of the consortium; however, higher concentrations of vitamin B₁₂ resulted in an occlusion of the functional groups. Furthermore, there were 5 significantly enriched protein pathways, namely carbon fixation in photosynthetic organisms; amino acid metabolic pathways; the pathway of one carbon pool by folate; nitrogen metabolism; and photosynthesis. Most proteins in these pathways were upregulated, which enhanced carbon fixation and photosynthesis in the algal cells. Simultaneously, B₁₂ promoted significant upregulation of proteins associated with the quorum-sensing pathway, which promoted the interaction between algae and bacteria.

Urinary tract infections are the utmost common bacterial infections caused by Proteus mirabilis, Pseudomonas aeruginosa, Escherichia coli, and Serratia marcescens. These uropathogens resist the action of several antibiotics due to their ability to form biofilms. Most of these bacterial pathogens use the quorum sensing (QS) machinery to co-ordinate their cells and regulate several virulence factors and biofilm formation. On the other hand, the anti-quorum sensing (anti-QS) and antibiofilm potential of silver nanoparticles have been well reported against certain bacterial pathogens, but to the best of our knowledge, no report is available against the pathogenicity of uropathogens in particular S. marcescens and P. mirabilis. Therefore, the present study is primarily focused on the anti-QS and antibiofilm potential of Piper betle-based synthesized silver nanoparticles (PbAgNPs) against S. marcescens and P. mirabilis. Initially, the silver nanoparticles were synthesized by the aqueous extract of P. betle and characterized by UV-absorbance spectroscopy, XRD, FT-IR, SEM, TEM, and DLS. The synthesized silver nanoparticles were assessed for their anti-QS activity and the obtained results revealed that the PbAgNPs inhibited the QS-mediated virulence factors such as prodigiosin, protease, biofilm formation, exopolysaccharides and hydrophobicity productions in uropathogens. The gene expression analysis divulged the downregulation of fimA, fimC, flhD, and bsmB genes in S. marcescens and flhB, flhD, and rsbA genes in P. mirabilis, respectively. The in vivo Caenorhabditis elegans assays revealed the non-toxic and anti-adherence efficiency of PbAgNPs. Furthermore, the non-toxic effect of PbAgNPs was also confirmed through peripheral blood mononuclear cells and normal lung epithelial cells. Therefore, the contemporary study demonstrates the use of PbAgNPs as a possible alternative toward conventional antibiotics in controlling QS and biofilm-related uropathogen infections.

To better construct a bioaugmented system for tobacco wastewater treatment, activated sludge was inoculated with different concentrations of the nicotine-degrading bacterium Pseudomonas sp. HF-1. The results showed that inoculum concentrations of 0.55 ± 0.01 and 1.10 ± 0.03 mg/g (dry weight of strain HF-1/dry weight of activated sludge) were best to ensure strain HF-1 survival and successful bioaugmentation. The release pattern of autoinducer (AI) for quorum sensing in the bioaugmented system was also investigated. During the period of HF-1 inoculation, compared with failed bioaugmented systems, AI-2 was significantly increased in the successful systems, suggesting that AI-2-mediated bacterial communication played an important role in the colonization of HF-1. When inoculation of strain HF-1 was stopped, the amount of AI-2 decreased and leveled out in all systems. Notably, there was a greater than threefold increase of short-chain AHLs in failed bioaugmented systems, but no increase in successful ones, implying that the fluctuation of short-chain AHLs could be an indicator of the failure of bioaugmentation. Thus, AI-2-mediated quorum sensing could be implemented to facilitate HF-1 colonization.

In this paper, the effects of cerium oxide nanoparticles (CeO₂ NPs) on the group bacterial behaviors were elaborated. After 36-h cultivation, the biofilm biomass was enhanced by the sub-lethal concentrations of 0.5 and 2 mg/L CeO₂ NP exposure. Meanwhile, the promoted production of total amino acids in microbes further resulted in the increased surface hydrophobicity and percentage aggregation. To resist the CeO₂ NPs stress, the biofilm exhibited a double-layer microstructure, with the protein (PRO) and living cells occupying the bottom, the polysaccharide (PS), and dead cells dominating the top. The bacterial diversity was highly suppressed and Citrobacter and Pseudomonas from the phylum of γ-Proteobacteria strongly dominated the biofilm, indicating the selective and enriched effects of CeO₂ NPs on resistant bacteria. The stimulated inherent resistance of biofilm was reflected by the reduced adenosine triphosphate (ATP) content after 4 h exposure. The increased levels of reactive oxygen species (ROS) in the treatments of 8 h CeO₂ NP exposure led to the upregulated quorum sensing signals of acylated homoserine lactone (AHL) and autoinducer 2 (AI-2), beneficial to mitigating the environmental disturbance of CeO₂ NPs. These results provide evidences for the accelerating effects of CeO₂ NPs on biofilm formation through oxidative stress, which expand the understanding of the ecological effects of CeO₂ NPs.
