Characterization of microplastic litter in the gastrointestinal tract of Solea solea from the Adriatic Sea

Micro-plastic particles in the world's oceans represent a serious threat to both human health and marine ecosystems. Once released into the aquatic environment plastic litter is broken down to smaller pieces through photo-degradation and the physical actions of waves, wind, etc. The resulting particles may become so small that they are readily taken up by fish, crustaceans and mollusks. There is mounting evidence for the uptake of plastic particles by marine organisms that form part of the human food chain and this is driving urgent calls for further and deeper investigations into this pollution issue.The present study aimed at investigating for the first time the occurrence, amount, typology of microplastic litter in the gastrointestinal tract of Solea solea and its spatial distribution in the northern and central Adriatic Sea. This benthic flatfish was selected as it is a species of high commercial interest within the FAO GFCM (General Fisheries Commission for the Mediterranean) area 37 (Mediterranean and Black Sea) where around 15% of the overall global Solea solea production originates.The digestive tract contents of 533 individuals collected in fall during 2014 and 2015 from 60 sampling sites were examined for microplastics. These were recorded in 95% of sampled fish, with more than one microplastic item found in around 80% of the examined specimens. The most commonly found polymers were polyvinyl chloride, polypropylene, polyethylene, polyester, and polyamide, 72% as fragments and 28% as fibers. The mean number of ingested microplastics was 1.73 ± 0.05 items per fish in 2014 and 1.64 ± 0.1 in 2015. PVC and PA showed the highest densities in the northern Adriatic Sea, both inshore and off-shore while PE, PP and PET were more concentrated in coastal areas with the highest values offshore from the port of Rimini.

Per- and polyfluorinated alkyl substances (PFAS) are highly persistent chemicals, which pose a potential risk for aquatic wildlife due to their bioaccumulative behaviour and toxicological effects. Although the distribution of PFAS in marine environments has been studied worldwide, little is known on the contamination of PFAS in the southern North Sea. In the present study, the bioaccumulation and trophic transfer of Perfluoroalkyl acids (PFAAs) was studied in liver and muscle tissue of seven fish species and in whole-body tissue of two crustacean species, collected at 10 sites in the Belgian North Sea. Furthermore, the human and ecological health risks were examined. Overall, perfluorooctane sulfonate (PFOS) was predominant in all matrices and other long-chain PFAS were frequently detected. Mean PFOS concentrations ranged from <LOQ to 107 ng/g (ww) in fish liver, from <LOQ to 24 ng/g ww in fish muscle and from 0.29 to 5.6 ng/g ww in crustaceans. Elevated perfluorotridecanoic acid (PFTrDA) concentrations were detected in fish liver from the estuarine and coastal region (<LOQ-116 ng/g ww), indicating a specific point source of this compound. Based on stable isotope analysis, no distinctive trophic transfer patterns of PFAS could be identified which implies that the bioconcentration of PFAS from the surrounding abiotic environment is most likely dominating over the biomagnification in the studied biota. The consumption of commercially important species such as the brown shrimp (Crangon crangon), plaice (Pleuronecta platessa), sole (Solea solea) and whiting (Merlangus merlangus) might pose potential health risks if it exceeds 17 g/day, 18 g/day, 26 g/day and 43 g/day respectively. Most PFOS measurements did not exceed the QSbᵢₒₜₐ,ₕₕ of 9.1 ng/g ww, however, the benchmark of 33 ng/g ww targeting the protection of wildlife from secondary poisoning was exceeded for 43% and 28% of the samples in plaice and sole.

On December 12th, 1999, the oil tanker Erika sank off the southern coast of Brittany (France), releasing 20 00030 000 tons of heavy oil in the open sea. Among the affected coastal habitats were important nurseries for numerous flatfish species and particularly for the common sole, Solea solea. To investigate the potential impact of the spill on this economically significant resource, we employed Fry's concept of metabolic scope for activity to assess the deleterious effect of fuel exposure on the functional integrity of juvenile sole. Fish were captured from uncontaminated areas and experimentally exposed to contamination conditions mimicking those encountered during the weeks that followed the Erika spill. Using respirometry techniques we measured basal and active metabolic rates, and calculated aerobic metabolic scope, in control and fuel-exposed sole. We then compared the ability of control and fuel-exposed sole to face an episode of reduced oxygen availability. We found that whereas basal metabolic rate was not altered in fuel-exposed fish, active metabolic rate (−27%), and therefore aerobic metabolic scope, were impacted. These changes in metabolic scope were viewed as indicating changes in fishes' ability to face environmental contingencies. Finally the ability of sole to face an episode of reduced oxygen availability was found to be significantly altered following fuel exposure as indicated by a 65% increase in the critical oxygen level. It is concluded that fuel-exposed sole are functionally impaired and less able to face environmental challenges. The link between these results and the recently reported fall in the abundance of the year class that suffered the Erika oil spill is discussed.

This study extends our knowledge of the bioaccumulation of polycyclic aromatic hydrocarbons (PAHs) in marine organisms and investigates its possible determinants. PAH levels were measured in Solea solea tissue and in marine sediments collected from three areas of the northern Adriatic Sea characterized by different anthropic impacts (Venetian Lagoon, Po Delta, and fishing grounds off Chioggia). The possibility of differential PAH bioaccumulation in different tissues (muscle, liver and gills) was investigated by seeking relationships between mean individual and total PAH concentrations in tissue and sediment samples, the physicochemical properties of PAHs (rings and Kow), and some key biological variables (lipid content of tissues, body size, habitat). The present study demonstrated that the lipid content might not be the only determinant of PAH bioaccumulation in common sole tissues. The habitat characteristics, the tissue types and some physicochemical properties of compounds were closely related to PAH bioaccumulation.

One of the aims of this study is the development of a pretreatment method for additives, plasticizers and other components of micro-litter (APFs), and small microplastics (SMPs <100 μm) in the gastrointestinal tract (GIT) of five of the most widely distributed and consumed commercial fish species, Engraulis encrasiculos, Sardina pilchardus, Mullus surmuletus, Solea solea, and Sparus aurata. The second aim was to develop a simultaneous quantification and identification method via Micro-FTIR of APFs and SMPs ingested by these commercial fish species. The distribution of SMPs and APFs is characteristically different for each species investigated. E. encrasiculos and S. pilchardus had a higher weight of SMPs than the other species investigated. Regarding APFs, the highest abundance was observed in E. encrasiculos. This study highlights the importance of studying additives and plasticizers that can be used as efficient proxies of microplastics, as shown by the presence of vulcanizing agents such as Vanax®.

Mercury (Hg) is a contaminant of global concern in marine ecosystems, notably due to its ability to accumulate and concentrate in food webs. Concentrations of total mercury (THg), methylmercury (MeHg) and inorganic mercury (IHg) were assessed and compared in different tissues (liver, muscle, and gonads) of three common fish species (hake Merluccius merluccius, red mullet Mullus surmuletus, and sole Solea solea) from the continental shelf from the southern part of the Bay of Biscay. Several studies investigated Hg concentration in fish muscle, but few assessed concentrations in other organs, despite the importance of such data to understand contaminant organotropism and metabolization. Results showed that trophic position and feeding habitat are required to understand the variability of Hg concentration in muscle between fish species. In addition, high MeHg/THg ratio in muscle could be explained by the predatory behavior of the studied fish species and the biomagnification of this Hg species within the food web, MeHg. Despite differences between species, Hg concentration was always higher in muscle (from 118 ± 64 to 338 ± 101 ng g⁻¹ w.w.) and liver (from 122 ± 108 to 271 ± 95 ng g⁻¹ w.w.). These results can be related to physiological processes especially the MeHg detoxification strategies.

Sole and hake, together with sediments, were collected during two campaigns along the Basque continental shelf to study the utility of two existing histopathological indices for assessing the biological effects of contaminants to implement the European Marine Strategy Framework Directive. Hepatic and gonadal histopathology were measured, and metal and/or organic contaminants were determined in both liver and sediments. Sediments from the Basque continental shelf were found to be moderately impacted by metals but non-impacted by organic compounds. Metal bioaccumulation and histopathological lesions in liver were higher in sole than in hake, although non-specific and early non-neoplastic toxicopathic lesions were observed in both species. No gross alterations were recorded in gonad. The two histopathological indices applied were highly correlated in both organs but the lack of correlation between sediment contamination levels, bioaccumulation and histopathological indices suggests that other factors, rather than pollution alone, are responsible for the biological effects observed.

Edible muscle tissues of Solea solea, Mullus barbatus, and Sardina pilchardus marketed in Mersin were analyzed for their Cr (total), Mn, Fe, Ni, Cu, Zn, As (total), Cd, Sn, and Pb levels. Metal levels of the tissues were determined using inductively coupled plasma-mass spectrophotometric (ICP-MS) methods. Muscle levels of Cr, Mn, Fe, Ni, Cu, Zn, As, Sn, and Pb were determined as 0.19–2.80, 0.08–3.88, 0.93–25.76, 0.03–0.63, 0.01–1.96, 1.28–45.95, 0.49–25.26, 0.14–4.03, and 0.02–1.37 mg kg⁻¹ w.w., respectively. Cadmium levels were below detection limits in all the muscle samples taken. Mean metal levels of the tissues were compared with the provisional tolerable daily (PTDs) and weekly (PTWIs) intake limits. Mean metal levels taken by the consumption of analyzed tissues were below PTDs and PTWIs; hence, the fish species studied do not pose any risk for human consumption from the point of heavy metals.

Many studies have been carried out along mighty rivers with heavily industrialized watersheds to evaluate pollutants and their effects on freshwater organisms. However, their impact on marine organisms is virtually unknown. In order to cover this gap, Solea solea, one of the most important commercial fish species, together with sediments, were sampled during 2013–2015 offshore from the Ebro Delta river mouth. Fish health indicators (condition indices, histological tissue alterations, and parasite descriptors) were used to assess the potential effect of pollutants, an issue of particular interest in the area following the dredging activities taking place in the river upstream in 2013. No major histopathological alterations were detected, but perivascular inflammatory foci (PIF) were frequently observed, especially in 2014. The most prevalent and abundant parasites were acanthocephalans and digeneans within the digestive tract and copepods on the gills. Levels of trace metals from sediments and fish muscle were below the effects range median and reference levels accepted for human consumption, respectively. However, the lower levels of the hepatosomatic index, higher numbers of PIF, and variations in the abundance of parasites in 2014 and 2015 could suggest a pollutant exposure during these years. These results warn signs of toxicity, which could be associated with sediment leaks during the dredging activities.

The ²¹⁰Po radionuclide concentrations of some marine organisms (Eriphia verrucosa, Solea solea, Mytilus galloprovincialis, Ulva lactuca) obtained from the northern part of the Sea of Marmara (Tekirdağ) in 2019 are presented in this study. The spontaneous radiochemical processes for ²¹⁰Po deposition in these marine species were performed by using three various metal discs. The ²¹⁰Po deposition yields of different species on silver, stainless steel and aluminium discs was investigated, and then, the disc with the best chemical deposition yield was determined. The ²¹⁰Po activity concentrations of E. verrucosa samples were measured to be 6.3 ± 0.8 Bq kg⁻¹ on silver disc, 5.9 ± 1.4 Bq kg⁻¹ on stainless steel disc and 3.4 ± 2.1 Bq kg⁻¹ on aluminium disc. The ²¹⁰Po activity concentrations of S. Solea on silver, stainless steel and aluminium discs were found to be 15 ± 2 Bq kg⁻¹, 11 ± 3 Bq kg⁻¹ and 4.8 ± 2.4 Bq kg⁻¹, respectively. The ²¹⁰Po specific activities of M. galloprovincialis samples taken in port for silver, stainless steel and aluminium discs were 39 ± 4 Bq kg⁻¹, 23 ± 8 Bq kg⁻¹, and N.M. (not measurable), respectively. Also M. galloprovincialis samples collected outside the port had the ²¹⁰Po concentrations of 57 ± 6 Bq kg⁻¹ and 21 ± 8 Bq kg⁻¹ on silver and stainless steel. While the ²¹⁰Po concentrations for out of port U. lactuca samples were found to be 60 ± 6 Bq kg⁻¹ for silver, 49 ± 12 Bq kg⁻¹ for stainless steel and 39 ± 15 Bq kg⁻¹ for aluminium, the ²¹⁰Po concentrations for in port U. lactuca on silver were determined to be 19 ± 3 Bq kg⁻¹. The annual effective ingestion doses from the consumption of E. verrucosa, S. solea, M. galloprovincialis and U. lactuca species were also calculated.
