Hematological responses of Goldfish (Carassiusauratus) to different acute concentrations of Silver Sulfate as a toxicant

This study aimed to evaluate the efficacy of silver sulfate (AgSO4) as a toxicant in goldfish (Carassiusauratus). One hundred and forty-seven live specimens of C. auratus were obtained and exposed to 1, 10, 100, 500, 1000 and 2000 ppm of AgSO4for 96 hours. There was one control group (no AgSO4) and three replicates. The physicochemical properties of water and the following parameters were constant: pH: 7.56±0.45 (TS1); temperature: 19±1°C; hardness: 293±2.35 ppm and dissolved oxygen: 8.80 ±0.06 mg L-1 (DO-5510). LC1, LC10, LC30, LC50, LC70, LC90 and LC99 were calculated in 24, 48, 72 and 96 hours. For assessing the impact ofAgSO4 on physiological responses of goldfish hematological indices, blood glucose and cortisol levels were measured. Results showed that LC50 96-h of AgSO4 for goldfish was 687.81 ppm. In addition, the use of AgSO4induces a significant decrease in MCHE after 48 hours, MCV and MCH after 96 hours and lymphocyte after 96 hours in contrast to the control group (P<0.05). Furthermore, increased lymphocyte was significant after 24 hours exposure (P<0.05). In addition, glucose increased significantly at P<0.05 with time increase24 hours after experiment but this (). In conclusion, the study showed that acute toxicity of AgSO4induced hematological alterations in goldfish and offers a tool for the evaluation oftoxicity-derived alterations.

We couple current findings of pesticides in surface and groundwater to the history of pesticide usage, focusing on the potential contribution of legacy pesticides to the predicted ecotoxicological impact on benthic macroinvertebrates in headwater streams. Results suggest that groundwater, in addition to precipitation and surface runoff, is an important source of pesticides (particularly legacy herbicides) entering surface water. In addition to current-use active ingredients, legacy pesticides, metabolites and impurities are important for explaining the estimated total toxicity attributable to pesticides. Sediment-bound insecticides were identified as the primary source for predicted ecotoxicity. Our results support recent studies indicating that highly sorbing chemicals contribute and even drive impacts on aquatic ecosystems. They further indicate that groundwater contaminated by legacy and contemporary pesticides may impact adjoining streams. Stream observations of soluble and sediment-bound pesticides are valuable for understanding the long-term fate of pesticides in aquifers, and should be included in stream monitoring programs.

We analyzed the spatial distribution of an antifouling biocide, 4,5-dichloro-2-n-octyl-4-isothiazolin-3-one (Sea-Nine 211) in the surface water and sediments of Hiroshima Bay, Japan to determine the extent of contamination by this biocide. A quantitative estimate of the environmental concentration distribution (ECD) and species sensitivity distributions (SSDs) for marine organisms were derived by using a Bayesian statistical model to carry out a probabilistic ecological risk analysis, such as calculation of the expected potentially affected fraction (EPAF). The spatial distribution analysis supported the notion that Sea-Nine 211 is used mainly for treatment of ship hulls in Japan. The calculated EPAF suggests that approximately up to a maximum of 0.45% of marine species are influenced by the toxicity of Sea-Nine 211 in Hiroshima Bay. In addition, estimation of the ecological risk with a conventional risk quotient method indicated that the risk was a cause for concern in Hiroshima Bay.

BDE209 and Pb are ubiquitous contaminants at e-waste recycling sites (EWRSs). This study aimed to determine acute and sub-acute toxicity to earthworm Eisenia fetida induced by BDE209 and Pb in natural soil. Results demonstrated that the inhibition of Pb on growth and reproduction of earthworms followed a dose-dependent pattern. Earthworms exposed to 100 mg kg−1 of BDE209 displayed avoidance responses, while the soil indicated a more obvious decline of habitat function with the increase of Pb level. Comet assay suggested that increasing concentrations of Pb exposure resulted in a gradual increase in the tail length and olive tail moment, which meant that the degree of DNA damage was promoted. BDE209 addition could reduce the damage; therefore the joint effects of both chemicals showed antagonistic. These results revealed that joint exposure (BDE209-Pb) could elicit pronounced biochemical and physiological responses in earthworms, and the DNA damage might be potential molecular biomarker of the two pollutants.

Difenoconazole is a widely used triazole fungicide, its extensive application may potentially cause toxic effects on non-target organisms. To investigate the effect of difenoconazole on cholesterol content and related mechanism, adult zebrafish were exposed to environmental related dosage (0.1, 10 and 500 μg/L) difenoconazole. The body weight and hepatic total cholesterol (TCHO) level was tested at 7, 15 and 30 days post exposure (dpe). The expressions of eight cholesterol synthesis genes and one cholesterol metabolism gene were assessed via Quantitative PCR method. The significant decrease of TCHO level in male zebrafish liver was observed at 15 and 30 dpe, which was accompanied by apparent hepatic cholesterol-genesis genes expression decline. In comparison with males, female zebrafish showed different transcription modification of tested genes, and the cholesterol content maintain normal level during the whole exposure.

Dissolution of silver nanoparticles (AgNP with carbonate or citrate coating, total Ag 1–5 μM) was examined in the presence of the ligands cysteine, chloride and fulvic acids and of the oxidant hydrogen peroxide (H2O2) at low concentrations at pH 7.5. Dissolved Ag was separated from AgNP by ultrafiltration. Cysteine in the concentration range 0.2–5 μM resulted in an initial increase of dissolved Ag within few hours. Chloride (up to 0.1 mM) and fulvic acids (up to 15 mg L−1) had little effect on the dissolution of AgNP within hours to days. In contrast, very rapid dissolution within 1–2 h of both carbonate and citrate coated AgNP was observed in the presence of H2O2 in the concentration range 0.1–10 μM, under dark or light conditions. The high efficiency of H2O2 in dissolving AgNP is likely to be of importance in toxic effects of AgNP to algae, as H2O2 is produced and released into solution by algae.

The Gulf of Riga, river Daugava and several interconnected lakes around the City of Riga, Latvia, form a dynamic brackish-freshwater system favouring occurrence of toxic cyanobacteria. We examined bioaccumulation of microcystins and nodularin-R in aquatic organisms in Latvian lakes, the Gulf of Riga and west coast of open Baltic Sea in 2002–2007. The freshwater unionids accumulated toxins efficiently, followed by snails. In contrast, Dreissena polymorpha and most lake fishes (except roach) accumulated much less hepatotoxins. Significant nodularin-R concentrations were detected also in marine clams and flounders. No transfer of nodularin-R and microcystins between lake and brackish water systems took place. Lake mussels can transfer hepatotoxins to higher organisms, and also effectively remove toxins from the water column. Obvious health risks to aquatic organisms and humans are discussed.

Chronic effects of the ionic liquid [C4mim][Cl] (mp 73 °C) towards the microalga, Scenedesmus quadricauda were studied by flow cytometry, monitoring multiple endpoints of cell density, esterase activity, membrane integrity, reactive oxygen species and chlorophyll fluorescence. Toxicity was clearly in evidence, and although increased esterase activity indicated hormesis during initial exposure to [C4mim][Cl], inhibition of both esterase activity and chlorophyll fluorescence became apparent after 3 days. Cell density was also decreased by culturing with [C4mim][Cl], but this effect was clearly concentration-dependent and only became significant during the second half of the experiment. In contrast, [C4mim][Cl] had only a modest effect on reactive oxygen species (ROS) and caused little damage to cell membranes.

In the present study, we conducted a 2 week microcosm experiment with a natural freshwater bacterial community to assess the effects of titanium dioxide nanoparticles (TiO2-NPs) at various concentrations (0, 1, 10 and 100 mg/L) on planktonic and sessile bacteria under dark conditions. Results showed an increase of planktonic bacterial abundance at the highest TiO2-NP concentration, concomitant with a decrease from that of sessile bacteria. Bacterial assemblages were most affected by the 100 mg/L TiO2-NP exposure and overall diversity was found to be lower for planktonic bacteria and higher for sessile bacteria at this concentration. In both compartments, a 100 mg/L TiO2-NPs exposure induced a decrease in the ratio between the Betaproteobacteria and Bacteroidetes. For planktonic communities, a decrease of Comamonadaceae was observed concomitant with an increase of Oxalobacteraceae and Cytophagaceae (especially Emticicia). For sessile communities, results showed a strong decrease of Betaproteobacteria and particularly of Comamonadaceae.

Bisphenol A (BPA) is a ubiquitous pollutant around the globe, but whether environmental concentrations have toxic effects remains controversial. BPA interferes with a number of nuclear receptor pathways, including several that mediate animal responses to environmental input. Because thermal acclimation is regulated by these pathways in fish, we hypothesized that the toxicity of BPA would change with ambient temperature. We exposed zebrafish (Danio rerio) to ecologically relevant and artificially high concentrations of BPA at two acclimation temperatures, and tested physiological responses at two test temperatures that corresponded to acclimation temperatures. We found ecologically relevant concentrations of BPA (20 μg l−1) impair swimming performance, heart rate, muscle and cardiac SERCA activity and gene expression. We show many of these responses are temperature-specific and non-monotonic. Our results suggest that BPA pollution can compound the effects of climate change, and that its effects are more dynamic than toxicological assessments currently account for.
