Declines in heart rate variability associated with short-term PM2.5 exposure were modified by blood pressure control and treatment: A multi-city panel study in China

Exposure to fine particulate matter (PM₂.₅) was associated with altered heart rate variability (HRV). However, whether blood pressure (BP) control and angiotensin II receptor blocker (ARB) treatment modifies the associations was seldom addressed. Therefore, we conducted a 3-phase panel study among 282 hypertensive subjects aged 35–74 years in four cities of China to address this issue. Real-time personal PM₂.₅ sampling and 24-h ambulatory electrocardiogram monitoring were performed repeatedly in 3 different seasons. Linear mixed-effects models were fitted overall and by control status of BP and ARB treatment to assess the associations between short-term PM₂.₅ exposure and HRV. The average hourly PM₂.₅ concentrations (Mean ± SD) ranged from 19.3 ± 18.2 μg/m³ to 99.4 ± 76.9 μg/m³ across study phases and cities. Generally, PM₂.₅ exposure was associated with decreased hourly and 24-h HRV. However, these adverse impacts were attenuated among patients with controlled BP (<140/90 mmHg). For each 10 μg/m³ increment in moving average of previous 2 days' (MA2d) PM₂.₅ exposure, 24-h SDNN (standard deviation of NN intervals) and rMSSD (root mean square of successive RR interval differences) decreased by 0.89% (95% CI: 0.19%–1.59%) and 2.98% (95% CI: 1.04%–4.89%) among patients with uncontrolled BP (≥140/90 mmHg), whereas no obvious declines were observed among those with controlled BP (Pdᵢffₑᵣₑₙcₑ = 0.007 and 0.022, respectively). Furthermore, ARB treatment alleviated or eliminated PM₂.₅-associated declines in hourly and 24-h HRV among those with uncontrolled BP. For instance, 24-h SDNN decreased by 1.31% (95% CI: 0.54%–2.07%) with a 10 μg/m³ increment in lag 2 days’ PM₂.₅ exposure in ARB nonusers, whereas no obvious changes were observed in ARB users (Pdᵢffₑᵣₑₙcₑ = 0.021). In conclusion, although PM₂.₅ exposure would decrease HRV, better BP control and ARB treatment could attenuate these adverse impacts, which provides supporting evidence for alleviating autonomic dysfunction of hypertension patients living in areas with high-level PM₂.₅.

Per- and polyfluoroalkyl substances (PFASs) are environmentally and biologically persistent anthropogenic chemicals linked to adverse health outcomes. Epidemiological data have revealed association between exposure to specific PFAS and disruption of insulin level in bodies. However, the effect of PFASs on insulin secretion and the responsible molecular mechanism are poorly understood. In the present study, we used perfluorooctane sulfonate (PFOS) as a representative PFAS family member to investigate its effect on the insulin secretion in mouse pancreatic β cells (β-TC-6). Our results showed that exposure to PFOS inhibited silent information regulator 1 (SIRT1) activity, and molecular simulation showed PFOS could fit into the pocket overlapped with the nicotinamide adenine dinucleotide (NAD⁺) binding cavity in SIRT1. PFOS exposure upregulated uncoupling protein 2 (UCP2) expression, and this upregulation was blunted in the presence of Ex-527, a SIRT1 specific inhibitor. The mitochondria membrane potential (ΔΨm), as well as the glucose-induced ATP production and Ca²⁺ influx decreased under PFOS treatment. PFOS continual exposure (48 h) impaired glucose stimulated insulin secretion (GSIS), while the gene expression of insulin was not significantly altered. Importantly, the SIRT1 activator and UCP2 inhibitor could partly reverse the PFOS-induced impairment of GSIS. Taken together, the results suggested that PFOS continual exposure could inhibit SIRT1 activity, and the SIRT1-UCP2 pathway mediated, at least partially, the PFOS induced GSIS impairment.

Associations between polycyclic aromatic hydrocarbons (PAHs) and respiratory diseases have been widely studied, but the effects of PAH on liver toxicity in adolescents are unclear. Here, 3194 adolescents with NHANES data from 2003 to 2016 were selected. PAH exposure was assessed by measuring PAH metabolites in urine. The outcome variables were the levels of alanine aminotransferase (ALT), aspartate amino transferase (AST) and gamma-glutamyl transpeptidase (GGT). The association between PAH exposure and liver function was evaluated by the weighted quantile sum (WQS) and logistic regression, and the associations between PAHs and inflammation and blood lipids were evaluated by linear regression. Covariates were adjusted for age, ethnicity, BMI, physical activity, family income, cotinine, and urinary creatinine. The results showed that for females, mixed PAH exposure was related to an increased ALT level (OR = 2.33, 95% CI 1.15, 4.72), and 2-fluorene contributed the most (38.6%). Urinary 2-fluorene was positively associated with an elevated ALT level (OR = 2.19 95% 1.12, 4.27, p for trend = 0.004). Mechanistically, 2-fluorene can cause a 3.56% increase in the white blood cell count, a 6.99% increase in the triglyceride level, and 1.70% increase in the total cholesterol level. PAHs may have toxic effects, possibly mediated by inflammation and blood lipids, on the adolescent female liver. Additional confirmatory studies are needed.

Nitrous oxide (N₂O) is a devastating greenhouse gas and acts as an ozone-depleting agent. Earthworms are a potential source of soil N₂O emissions. Application of biochar can mitigate earthworm-induced N₂O emissions. However, the underlying interactive mechanism between earthworms and biochar in soil N₂O emissions is still unclear. A 35-day laboratory experiment was conducted to examine the soil N₂O emission dynamics for four different treatments, earthworm presence with biochar application (EC), earthworm presence without biochar application (E), earthworm absence with biochar application (C) and earthworm absence without biochar application, and the control. Results indicated a negative impact of biochar on earthworm activity, displaying a significantly (p ≤ 0.05) lower survival rate and biomass of earthworms in treatment EC than E. Compared with the control, earthworm presence significantly (p ≤ 0.05) increased cumulative N₂O emissions, while application of biochar in the presence of earthworms significantly (p ≤ 0.05) decreased cumulative N₂O emissions (485 and 690 μg kg⁻¹ for treatments EC and E, respectively). Treatments E and EC significantly (p ≤ 0.05) increased soil microbial biomass carbon (MBC), ammonium (NH₄⁺-N), nitrate (NO₃⁻N), and dissolved organic carbon (DOC) content and soil pH as compared with the control. The gene copy number of 16 S rRNA, AOA, AOB, nirS, and nosZ increased for all treatments when compared with the control; however, a significant (p ≤ 0.05) difference among the studied genes was only observed for the nosZ gene (2.05 and 2.56 × 10⁶ gene copies g⁻¹ soil for treatments E and EC, respectively). Earthworm-induced soil N₂O emissions were significantly (p ≤ 0.05) reduced by biochar addition. The possible underlying mechanisms may include: (1) short-term negative impacts on earthworm activity; (2) a change of functional gene abundance in earthworm casts; and (3) an increase in soil pH due to addition of biochar.

Aquaculture impacts on aquatic organic matter and ecosystem integrity are poorly understood, especially in tropical regions. Here, we investigated the impacts of Nile tilapia net cage farming on the elemental stoichiometry, fluorescence components, and stable isotopes of dissolved organic matter (DOM) of the large, tropical Furnas Reservoir (SE Brazil). Early-stage fish farming, i.e., relatively small and recently implemented farms, had detectable incipient effects on DOM characteristics, and these effects differed between reservoir branches. In the less eutrophic Rio Grande branch of the reservoir, we found a reduction in natural humic-like DOM components and an increase in a protein-like DOM component as far as 100 m away from fish farms. Further, we observed a decrease in δ¹⁵N-TDN due to fish farming. In the more eutrophic Rio Sapucaí branch, there were only local decreases in C:N ratios, as well as rises in C:P and N:P of DOM due to fish farming. These results suggest that early-stage fish farming had local but detectable effects on aquatic DOM that depended on previous eutrophication levels and highlight the need to assess the early impacts of fish farming on tropical reservoirs by combining different monitoring strategies.

Pesticides are heavily applied in rice–vegetable rotations in tropical China, yet publicly available information on the contamination and risk of currently used pesticides (CUPs) and legacy pesticides (LPs) in surface waters of river basins draining these areas is very limited. Therefore, in two tropical river basins (Nandu River and Wanquan River basins) dominated by rice–vegetable rotations in Hainan, China, pesticides were analyzed in 256 surface water samples in wet and dry seasons. Forty-one pesticides were detected, and total concentrations ranged from not detectable to 24.2 μg/L. Carbendazim and imidacloprid were the two most prevalent CUPs, detected in 59.8% and 17.7%, respectively, of surface water samples at concentrations above 0.1 μg/L. Chlorpyrifos was the main LP, detected in 9.0% of samples at a concentration above 0.05 μg/L. The fungicides difenoconazole and emamectin benzoate, the herbicide butachlor, and the insecticide acetamiprid occurred in ≥12.5% samples at concentrations above 0.1 μg/L. Surface waters typically (85.2%) contained 5 to 15 residues, with an average of nine. Seasonally, the concentrations of the 41 pesticides were in the order January > July > November > September. Spatially, the composition of the main CUPs (not LPs) was significantly different depending on position in the drainage, which also changed with seasons. Crop and pest types and wet and dry seasons were the key factors controlling the spatiotemporal distribution of CUPs and LPs in surface waters. On the basis of evaluations of the exposures to individual pesticides and the dominant combinations with ≥8 pesticides, multiple pesticides were likely a significant risk to aquatic organisms, although noncarcinogenic and carcinogenic risks to humans were low. This study provides valuable data to better understand pesticide occurrence and ecological risks in river basins draining areas with rice–vegetable rotation systems in tropical China.

Forest fires significantly affect the wildlife, vegetation, composition and structure of the forests. This study explores the potential of partially burnt wood recovered in the aftermath of a recent Canadian forest fire incident as a feedstock for generating hydrogen-rich syngas through hydrothermal gasification. Partially burnt wood was gasified in hydrothermal conditions to study the influence of process temperature (300–500 °C), residence time (15–45 min), feed concentration (10–20 wt%) and biomass particle size (0.13 mm and 0.8 mm) using the statistical Taguchi method. Maximum hydrogen yield and total gas yield of 5.26 mmol/g and 11.88 mmol/g, respectively were obtained under optimized process conditions at 500 °C in 45 min with 10 wt% feed concentration using biomass particle size of 0.13 mm. The results from the mean of hydrogen yield show that the contribution of each experimental factors was in the order of temperature > feed concentration > residence time > biomass particle size. Other gaseous products obtained at optimum conditions include CO₂ (3.43 mmol/g), CH₄ (3.13 mmol/g) and C₂–C₄ hydrocarbons (0.06 mmol/g).

Owing to the high nonlinearity and noise in the air quality index (AQI), tackling the uncertainties and fuzziness in the forecasting process is still a prevalent problem. Therefore, this study developed an intelligent hybrid air-quality forecasting system based on feature selection and a modified evolving interval type-2 quantum fuzzy neural network (eIT2QFNN), which provides accurate air-quality forecasting information by considering climate influencing factors. The main contributions of this study are as follows. The optimal input structure of the model is determined by the proposed second-stage feature-selection model, which can better extract the influencing variables and remove redundant information. Moreover, a novel multi-objective chaotic Bonobo optimizer algorithm is proposed to improve the eIT2QFNN. The modified eIT2QFNN implements AQI prediction by considering the importance of influencing variables that can cope with the uncertainties and fuzziness in the forecasting process. Finally, the Diebold-Mariano and modified Diebold-Mariano tests are employed to evaluate the performance of the proposed system. The experimental results demonstrate that our proposed system significantly improves the modeling performance in terms of high accuracy and compact structure, and can thus serve as an effective tool for air-quality management.

Understanding the impact of increases in pCO₂ (OA) and extreme changes in temperature on marine organisms is critical to predicting how they will cope with climate change. We evaluated the effects of OA as well as warming and cooling trend temperature on early reproductive traits of Lessonia trabeculata, a bio-engineer kelp species. Sori discs were maintained for an exposure time (ET) of 3 (T3) and 7 (T7) days to one of two contrasting pCO₂ levels (450 and 1100 μatm). In addition, at each pCO₂ level, they were subjected to three temperature treatments: 15 °C (control), 10 °C (cool) and 19 °C (warm). Subsequently, we compared sorus photosynthetic performance (Fv/Fm), the number of meiospores released (MR) and their germination rate (GR) after 48 h of settlement, with values obtained from sori discs not exposed (DNE) to the treatments. The Fv/Fm measured for DNE was lower than at T3 and T7 at 10 and 15 °C but not at 19 °C. Regardless of temperature, we found no significant differences between MR measured at T0 and T3. MR at T7 was significantly lower at 19 °C than at 10 and 15 °C. We found only a significant reduction in MR in response to elevated pCO₂ at T3. The GR of meiospores released by DNE and then maintained for 48 h to 19 °C decreased significantly by ~33% when compared with those maintained for the same time at 10 and 15 °C. A similar, but more drastic reduction (~54%) in the GR was found in meiospores released by sori discs exposed for T3 and maintained for 48 h to 19 °C. We suggest that OA and warming trend will threaten the early establishment of this species with further consequences for the functioning of the associated ecosystem.

Lead (Pb) is a heavy metal that has been proven to be toxic to both animals and humans. Genom-wide DNA methylation in domestic dogs exposed to high levels of Pb in Kabwe, Zambia was analyzed in this study. Using next-generation sequencing on samples from 20 domestic dogs (mean blood Pb concentration: 43.6 μg/dL and 7.2 μg/dL in the high and low exposure groups), a digital restriction enzyme analysis of methylation was performed to identify the genomic locations of differentially methylated CpG sites. A validation study on an additional 20 dogs followed (blood Pb concentration: 4.9–29.7 μg/dL). The cluster analysis resolved two broad clusters indicating high and low Pb exposure. The study identified 827 (1.2%) CpG sites with differences in methylation (101 CpG sites were hypermethylated in the low exposure group and 726 were hypermethylated in the high exposure group). The sites corresponded to 26 genes with differentially methylated CpG sites at their promoter regions, including the NGF gene. The methylation of four CpG sites was validated using bisulfite pyrosequencing. The results indicate that aberrant hypermethylation is prevalent in dogs exposed to Pb. The altered DNA methylation of the genes identified in this study contributes to a greater understanding of the epigenetic changes caused by Pb exposure and highlights novel biomarker discoveries across species.
