Acute and subacute oral toxicity of propylene glycol enantiomers in mice and the underlying nephrotoxic mechanism

Propylene glycol (PG; 1,2-propanediol) has been commonly used as a food additive and vehicle in pharmaceutical preparations. PG can form rectus (R-) enantiomers and sinister (S-) enantiomers. Herein, Kunming mice were used as the animal model to evaluate the acute and subacute oral toxicity of R-PG, S-PG and RS-PG (1:1 racemic mixture of R-PG and S-PG). The median lethal doses of R-PG, S-PG and RS-PG administered by oral gavage to mice were 22.81 g/kg, 26.62 g/kg and 24.92 g/kg, respectively. In the 28-day oral subacute toxicity study, the body weight, organ weights, serum biochemical, and renal histology were examined. There was no difference in subacute toxicity among R-PG, S-PG and RS-PG. The administration of 1 and 5 g/kg/day PG for 28 days caused nephrotoxicity. The kidney somatic index and levels of blood urea nitrogen exhibited a significant increase. Moreover, the activities of superoxide dismutase, catalase, and glutathione peroxidase significantly decreased after the treatment with PG. The levels of malondialdehyde, tumor necrosis factor α, interleukin 1β, and interleukin 6 significantly increased in the kidney. The results show that the nephrotoxic effects of PG are induced by oxidative stress, and the activation of the inflammatory response is mediated by the NF-κB signaling pathway. Together, these findings provide information on R-PG, S-PG and RS-PG treatments for the risk assessment of toxicity and effects on human health.

House dust is an important medium for exposure to persistent pollutants, such as metals. Detailed characterization of metal composition is needed to identify sources and potential health impacts of exposure. In this study we show that specific metals in dust dominate in different locations within residential homes in a mid-size Canadian city (Fort McMurray, Alberta), up to two years after a major wildfire event in 2016. Dust samples were collected in high-traffic (e.g. bedroom, N = 186), low-traffic (e.g. basement, N = 158), and entranceway areas (N = 171) of residential homes (N = 125), and analyzed for 25 trace metal elements using Inductively Coupled Plasma Mass Spectrometry (ICP-MS). The profile of metal concentrations in the entranceway resembled that of outdoor soils, especially for crustal elements. On the other hand, Cu, Zn, and Pb concentrations in dust sampled in indoor living areas were on average three to six times higher than in other indoor locations indicating indoor sources for these elements. In general, Pb concentrations were similar or lower than in an average Canadian residence, but a substantial fraction showed anomalously high concentrations in the low-traffic areas, particularly on concrete surfaces in basements. Notably, the 2016 wildfires showed limited influence on metal concentrations in indoor dust, despite the widespread concerns in the community about long term exposure. Enrichment factor ratio calculations and principal component analysis showed two classes of sources of metals in dust that represent geogenic-outdoor sources and anthropogenic-indoor sources. Overall, we demonstrate that outdoor and indoor sources of dust impact its composition, and these influences are reflected in the different areas of a home.

Non-ferrous smelting is a primary cause of serious soil pollution. Contamination and health risks of heavy metals in soils around various types of non-ferrous smelteries in China were assessed using data from peer-reviewed papers published between 2000 and 2019. Development in the relevant environmental policy in China was discussed. The studied non-ferrous smelting sites were mainly located in provinces that produced non-ferrous metals on a large scale. The average concentrations of the heavy metals in soils around the non-ferrous smelteries (in mg per kg of soil) were as follows: Cd, 19.8; Cu, 265; Pb, 1536; and Zn, 1371; the concentrations greatly exceeded their corresponding background values. The smelting sites with high soil contamination in terms of metal concentrations, geo-accumulation (Igₑₒ), and pollution index (PI) were mainly distributed in several provinces of China, including Guangxi, Gansu, Hunan, Hubei, Chongqing, and Liaoning. Soils near smelteries that processed copper were the most polluted based on Igₑₒ and PI. The accumulation of Cd and Pb in soils around non-ferrous smelteries would pose potentially high risks to residents. A series of environmental policies have proven successful in lowering the emissions of contaminants from the non-ferrous in China. The findings of the study suggested that the strategies to control soil pollution around non-ferrous smelteries should primarily focus on Cd and Pb.

Large amounts of antibiotics/disinfectants are used in the farming of ornamental fish so as to prevent and treat bacterial infection. This may exert considerable selection pressures on the prevalence and propagation of antibiotic resistance genes (ARGs). However, the levels of ARGs and their potential prevalence mechanism in the wastewater of the ornamental fish industry remains unclear. In this work, wastewater is collected from a representative ornamental fish market (OFM) that is located at the northern China to study the occurrence and abundance of 21 ARGs and 2 integrase genes. Results indicated that 15 different ARGs and 2 integrase genes are existent and prevalent in the wastewater of OFM, whereby concentrations range from 2.01 to 10.34 copies/L. Proteobacteria, Bacteroidetes, Verrucomicrobia, and Firmicutes are the predominant phyla in the wastewater samples. 17 species of human opportunistic pathogens are present with relative abundance of up to 0.01%, which suggests a considerable risk of pathogens acquiring and disseminating ARGs. Moreover, oxytetracycline, ciprofloxacin, norfloxacin, sulfadiazine, and chloramphenicol are most frequently detected in wastewater, with concentrations of up to 1150, 877, 514, 1970, and 1700 ng/L, respectively. Notably, good correlations have been determined among ARGs and antibiotics, non-antibiotic environmental factors in wastewater of OFM. This current study reveals, for the first time, that OFM is a previously unperceived reservoir for ARG prevalence in aquatic environment and water environmental factors (particularly antibiotics), and their induced shifts in the microbial communities are the key factors for distribution of ARGs in OFM.
Afficher plus [+] Moins [-]Organic-matter decomposition as a bioassessment tool of stream functioning: A comparison of eight decomposition-based indicators exposed to different environmental changes

Organic-matter decomposition has long been proposed as a tool to assess stream functional integrity, but this indicator largely depends on organic-matter selection. We assessed eight decomposition-based indicators along two well-known environmental gradients, a nutrient-enrichment gradient (0.2–1.4 mg DIN/L) in central Portugal and an acidification gradient (pH: 4.69–7.33) in north-eastern France to identify the most effective organic-matter indicator for assessing stream functional integrity. Functional indicators included natural leaf litter (alder and oak) in 10-mm and 0.5-mm mesh bags, commercial tea (Lipton green and rooibos teas in 0.25-mm mesh bags), wood sticks (wood tongue depressors) and cotton strips. Biotic indices based on benthic macroinvertebrates (IPtIN for Portugal and IBGN for France) were calculated to compare the effectiveness of structural and functional indicators in detecting stream impairment and to assess the relationship between both types of indicators. The effectiveness of organic-matter decomposition rates as a functional indicator depended on the stressor considered and the substrate used. Decomposition rates generally identified nutrient enrichment and acidification in the most acidic streams. Decomposition rates of alder and oak leaves in coarse-mesh bags, green and rooibos teas and wood sticks were positively related with pH. Only decomposition rates of rooibos tea and wood sticks were related with DIN concentration; decomposition rates along the nutrient-enrichment gradient were confounded by differences in shredder abundance and temperature among streams. Stream structural integrity was good to excellent across streams; the IPtIN index was unrelated to DIN concentration, while the IBGN index was positively related with pH. The relationships between decomposition rates and biotic indices were loose in most cases, and only decomposition rates of alder leaves in coarse-mesh bags and green tea were positively related with the IBGN. Commercial substrates may be a good alternative to leaf litter to assess stream functional integrity, especially in the case of nutrient enrichment.

Real-time river chloride prediction has received a lot of attention for its importance in chloride control and management. In this study, an artificial neural network model (i.e., multi-layer perceptron, MLP) and a statistical inference model (i.e., stepwise-cluster analysis, SCA) are developed for predicting chloride concentration in stream water. Then, an ensemble learning model based on MLP and SCA is proposed to further improve the modeling accuracy. A case study of hourly river chloride prediction in the Grand River, Canada is presented to demonstrate the model applicability. The results show that the proposed ensemble learning model, MLP-SCA, provides the best overall performance compared with its two ensemble members in terms of RMSE, MAPE, NSE, and R² with values of 11.58 mg/L, 27.55%, 0.90, and 0.90, respectively. Moreover, MLP-SCA is more competent for predicting extremely high chloride concentration. The prediction of observed concentrations above 150 mg/L has RMSE and MAPE values of 9.88 mg/L and 4.40%, respectively. The outstanding performance of the proposed MLP-SCA, particularly in extreme value prediction, indicates that it can provide reliable chloride prediction using commonly available data (i.e., conductivity, water temperature, river flow rate, and rainfall). The high-frequency prediction of chloride concentration in the Grand River can supplement the existing water quality monitoring programs, and further support the real-time control and management of chloride in the watershed. MLP-SCA is the first ensemble learning model for river chloride prediction and can be extended to other river systems for water quality prediction.

In humans and animal models, the kidneys and cardiovascular systems are negatively affected by BPA from the environment. It is considered that BPA have some potential estrogen-like and non-hormone-like properties. In this study, RNA-sequencing and its-related bioinformatics was used as the basic strategy to clarify the characteristic mechanisms of kidney–heart axis remodeling and dysfunction in diabetic male rats under BPA exposure. We found that continuous BPA exposure in diabetic rats aggravated renal impairment, and caused hemodynamic disorders and dysfunctions. There were 655 and 125 differentially expressed genes in the kidney and heart, respectively. For the kidneys, functional annotation and enrichment, and gene set enrichment analyses identified bile acid secretion related to lipid synthesis and transport, and MAPK cascade pathways. For the heart, these bioinformatics analyses clearly pointed to MAPKs pathways. A total of 12 genes and another total of 6 genes were identified from the kidney tissue and heart tissue, respectively. Western blotting showed that exposure to BPA activated MAPK cascades in both organs. In this study, the exacerbated remodeling of diabetic kidney–heart axis under BPA exposure and diabetes might occur through hemodynamics, metabolism disorders, and the immune-inflammatory response, as well as continuous estrogen-like stimulation, with focus on the MAPK cascades.

Insulation materials are essential components in construction, and their main objective is to increase the efficiency of thermal energy by minimizing internal and external thermal exchange. Accordingly, research and development studies are being actively conducted to increase the thermal resistance of insulation materials, and high-performance insulation materials that use organic chemicals have been developed after industrialization. However, thermal insulation comprising chemicals poses a potential risk of pollutant emissions and can cause health problems. In this study, five types of insulation materials and the contaminants generated from the building materials used in insulation construction were quantitatively analyzed. In addition, an empirical study on the discharge of pollutants was conducted using a test bed, and the effects of the pollutants discharged from the insulation material on the indoor environment were examined by analyzing the pollutant concentration for 90 days. In addition, we analyzed the effect of an insulation material on an indoor environment through the standard specifications. Moreover, the necessity of legal management of the emission of contaminants from insulation materials was proposed based on the empirical research results.

The European Noise Directive proposes using strategic noise maps as tools to assess populations affected by environmental noise. It recommends using computational methods instead of in situ measurements when possible. A sound source’s emission power is an important factor in the calculation of noise indicators. For traffic noise, this parameter is usually defined based on vehicle flow considering an emission spectrum that depends on the type of vehicle and its speed. This study analysed the possibility of using the categorisation method to propose an alternative method of defining a sound source’s emission power to develop noise maps. This was accomplished using previously published values of the emission power per unit length. Another method is also proposed that estimates traffic flows. To verify their estimation capacity, the results of both methods were compared with the values obtained from in situ measurements. The results demonstrated similar uncertainties in both methods and were in the range of expected average uncertainties compared to the results obtained by calculating a noise map with the measured experimental values. In particular, for the differences between calculations and measurements, in absolute values, the mean uncertainties were approximately 2 dBA in estimating different long-term noise indicators. For the differences, the mean of the uncertainties obtained via the categorisation method did not present significant differences for the null value for all the analysed noise indicators. Street stratification is a rapid and low-cost approach for road traffic noise mapping without increasing uncertainties.

Bisphenol A (BPA) is an endocrine disrupting chemical able to promote hormone-responsive tumors. The major route of BPA contamination being oral, the aim of the present study was to investigate BPA effects on oral cells. Here, we evaluated the impact of sub-chronic in vivo exposure to BPA and its in vitro effects on neoplastic and non-neoplastic oral cells. We evaluated the oral mucosa of mice chronically exposed to BPA (200 mg/L). The response of keratinocytes (NOK–SI) and Head and Neck (HN) Squamous Cell Carcinoma (SCC), HN12 and HN13 cell lines to BPA was examined. In vivo, BPA accumulated in oral tissues and caused an increase in epithelial proliferative activity. BPA disrupted the function of keratinocytes by altering pro-survival and proliferative pathways and the secretion of cytokines and growth factors. In tumor cells, BPA induced proliferative, invasive, pro-angiogenic, and epigenetic paths. Our data highlight the harmful effects of BPA on oral mucosa and, tumorigenic and non-tumorigenic cells. Additionally, BPA may be a modifier of oral cancer cell behavior by prompting a functional shift to a more aggressive phenotype.
