Tissue distribution, isozyme abundance and sensitivity to chlorpyrifos-oxon of carboxylesterases in the earthworm Lumbricus terrestris
2009
Sanchez-Hernandez, Juan C. | Wheelock, Craig E.
A laboratory-based study was conducted to determine the basal carboxylesterase (CbE) activity in different tissues of the earthworm Lumbricus terrestris, and its sensitivity to the organophosphate (OP) pesticide chlorpyrifos-oxon (CPx). Carboxylesterase activity was found in the pharynx, crop, gizzard, anterior intestine, wall muscle and reproductive tissues of L. terrestris, and multiple tissue-specific isozymes were observed by native gel electrophoresis. Esterase activity and sensitivity to CPx inhibition varied on a tissue- and substrate-specific basis, suggesting isoforms-specific selectivity to OP-mediated inhibition. Three practical issues are recommended for the use of earthworm CbE activity as a biomarker of pesticide exposure: (i) CbE should be measured using several routine substrates, (ii) it should be determined in selected tissues instead of whole organism homogenate, and (iii) earthworm CbE activity should be used in conjuncture with other common biomarkers (e.g., ChE) within a multibiomarker approach to assess field exposure of OPs, and potentially other agrochemicals. The measurement of carboxylesterase inhibition in earthworm is a sensitive and complementary biomarker of pesticide exposure.

Polychlorinated biphenyls (PCBs) were measured in sediment cores from ponds located near a large seabird colony at Cape Vera, Devon Island, Arctic Canada. Surface sediment PCB concentrations were approximately 5x greater in seabird-affected sites relative to a nearby control pond and were correlated with independent indicators of seabird activity including, sedimentary δ¹⁵N and lakewater chlorophyll a and cadmium concentrations. PCB fluxes were amongst the highest recorded from the High Arctic, ranging from 290 to 2400 ng m⁻² yr⁻¹. Despite a widespread ban of PCBs in the mid-1970s, PCB accumulation rates in our cores increased, with the highest values recorded in the most recent sediments. Possible mechanisms for the recent PCB increases include a vertical flux step driven by seabird-delivered nutrients and/or delayed loading of PCBs from the catchment into the ponds. The high PCB levels recorded in the seabird-affected sites suggest that seabird colonies are exposing coastal ecosystems to elevated levels of contaminants.

2009
Rubal, Marcos | Guilhermino, Lúcia M. | Medina, Matías H.
The study presented here searched for the level of taxonomic resolution required to detect the effects of low-level chronic pollution on estuarine meiobenthic communities. Meiofauna from two sites, with special attention to harpacticoid copepods, was analysed at different taxonomic levels of aggregation using uni- and multivariate methods. Adaptation processes that could buffer biodiversity disruptions were also considered through the analysis of fitness-related and tolerance traits in the harpacticoid copepod Paronychocamptus nanus. Results showed that uni- and multivariate analyses could be inadequate when assessing subtle anthropogenic contamination. Instead, the assessment of inter-population differences in tolerance to the main source of stress rises as a required procedure if potential effects of this type of contamination are being investigated. Specifically, a 96 h acute toxicity test performed with populations from the affected site appears as a faster and reliable general tool to assess impacts of low-level chronic pollution in estuaries.

Stomatal closure and biosynthesis of antioxidant molecules are two fundamental components of the physiological machinery that lead to stress adaptation during plant's exposure to salinity. Since high stomatal resistance may also contribute in counteracting O3 damages, we hypothesized that soil salinization may increase O3 tolerance of crops. An experiment was performed with alfalfa grown in filtered (AOT40 = 0 in both years) and non-filtered (AOT40 = 9.7 in 2005 and 6.9 ppm h in 2006) open-top chambers. Alfalfa yield was reduced by O3 (-33%) only in plants irrigated with salt-free water, while the increasing levels of soil salinity until 1.06 dS m-¹ reduced both stomatal conductance and plant O3 uptake, thus linearly reducing O3 effects on yield. Therefore a reliable flux-based model for assessing the effects of O3 on crop yield should take into account soil salinity. Moderate saline stress can reduce ozone uptake and yield losses in alfalfa plants.

2009
Cieslik, S.
Among air pollutants, ozone is the most important stressor to vegetation, which undergoes damage and biomass reduction after penetration of ozone molecules into the leaf tissues through the stomata. Stomatal ozone fluxes are considered the governing factor needed to assess risk to plant health due to ozone. Although this parameter may be calculated by modeling, direct measurements are scarce. Moreover, southern European situations, especially regarding Italy, require special attention due to the decoupling between ozone concentrations and fluxes. This work reviews ozone flux measurements made during the last 15 years through Italy. The dependence of ozone fluxes on environmental factors such as water supply is stronger than the dependence on the surface covering species.

The present study was carried out to understand the current contamination status of organochlorine compounds (OCs) in human breast milk from three metropolitan cities in India (New Delhi, Mumbai and Kolkata). Among the OCs analyzed, DDTs were predominant followed by HCHs and PCBs. CHLs and HCB levels were much lower. Contamination patterns were different in human milk found in our previous study in Chennai, a metropolitan city in southern India, indicating region specific exposure routes and variable sources. In comparison with previous data, levels of DDTs and HCHs generally declined with time, implying the effect of various bans and restrictions on their usage. No association between concentrations of OCs and demographic characteristics such as parity and age of mothers was observed which might be due to narrow range of mother's age. Estimated daily intake shows that some infants are exposed to OCs to a greater extent, particularly HCHs than the guideline standard. Indian infants may be exposed to relatively high levels of DDTs and HCHs through mothers' milk.

Sustained cultivation of Bacillus thuringiensis (Bt) transgenic crops requires stable transgene expression under variable abiotic conditions. We studied the interactions of Bt toxin production and chronic ozone exposure in Bt cry1Ac-transgenic oilseed rape and found that the insect resistance trait is robust under ozone elevations. Bt Cry1Ac concentrations were higher in the leaves of Bt oilseed rape grown under elevated ozone compared to control treatment, measured either per leaf fresh weight or per total soluble protein of leaves. The mean relative growth rate of a Bt target herbivore, Plutella xylostella L. larvae was negative on Bt plants in all ozone treatments. On the non-transgenic plants, larval feeding damage was reduced under elevated ozone. Our results indicate the need for monitoring fluctuations in Bt toxin concentrations to reveal the potential of ozone exposure for altering dosing of Bt proteins to target and non-target herbivores in field environments experiencing increasing ozone pollution. Elevated atmospheric ozone can induce fluctuations in insecticidal protein concentrations in transgenic plants.

A study of a large number of samples of flaking old paint from various buildings in Bergen, Norway (N = 68) suggests that paint may be the most important contemporary source of PCBs in this urban environment with concentrations of PCB7 up to 3.39 g/kg. Twenty-three of the samples were collected from a single building, and the concentrations were found to vary over 3 orders of magnitude. In addition, 16 concrete samples from a large bridge previously coated with PCB-containing paint were collected and separated into outer- and inner samples indicating that PCBs are still present in high concentrations subsequent to renovation. PCBs were found in several categories of paint from wooden and concrete buildings, potentially introduced to the environment by natural weathering, renovation, and volatilization. Consequently, this dispersion may lead to increased levels of PCBs in urban atmospheres, soils, and harbor sediments where high concentrations have resulted in Governmental advice against consumption of certain seafood. Paint from structures built during the period 1950-1970 may be the most important source of PCBs in an urban environment.

2009
André, A. | Antunes, S.C. | Gonçalves, F. | Pereira, R.
As part of the tier 2 of a site-specific risk assessment, this study was the first reporting an intensive in situ application of the bait-lamina assay; two exposure periods (7 and 14 days) were tested during four seasons in ten different sites, within a uranium mine area and at two different depths. The most contaminated sites (by deposition of sludge from the effluent treatment pond) were discriminated after 14 days of exposure because extremely low percentages of feeding activity were recorded. Previous sub-lethal ecotoxicological assays, already had demonstrated that the habitat function of these soils is compromised. Nevertheless, seasonality has proved to have a significant influence on responses. Thus to strength conclusions about the impact of contaminants, the in situ bait-lamina assay should be performed on different annual seasons, at least for temperate regions. It was also found that some environmental parameters (e.g. soil moisture and litter) can act as confounding factors in the bait-lamina assay. For phytoremediation of organic contaminants, plants have to host an efficiently degrading microflora. To assess the role of endophytes in alkane degradation, Italian ryegrass was grown in sterile soil with 0, 1 or 2% diesel and inoculated either with an alkane degrading bacterial strain originally derived from the rhizosphere of Italian ryegrass or with an endophyte. We studied plant colonization of these strains as well as the abundance and expression of alkane monooxygenase (alkB) genes in the rhizosphere, shoot and root interior. Results showed that the endophyte strain better colonized the plant, particularly the plant interior, and also showed higher expression of alkB genes suggesting a more efficient degradation of the pollutant. Furthermore, plants inoculated with the endophyte were better able to grow in the presence of diesel. The rhizosphere strain colonized primarily the rhizosphere and showed low alkB gene expression in the plant interior. Bacterial alkane degradation genes are expressed in the rhizosphere and in the plant interior.
