Photosynthesis, chloroplast pigments, and antioxidants in Pinus canariensis under free-air ozone fumigation

High O3 levels, driving uptake and challenging defense, prevail on the Canary Islands, being associated with the hot and dry summers of the Mediterranean-type climate. Pinus canariensis is an endemic conifer species that forms forests across these islands. We investigated the effects of ozone on photosynthesis and biochemical parameters of P. canariensis seedlings exposed to free-air O3 fumigation at Kranzberg Forest, Germany, where ambient O3 levels were similar to those at forest sites in the Canary Islands. The twice-ambient O3 regime (2xO3) neither caused visible injury-like chlorotic or necrotic spots in the needles nor significantly affected violaxanthin, antheraxanthin and zeaxanthin levels and the de-epoxidation state of the xanthophyll cycle. In parallel, stomatal conductance for water vapour, net photosynthesis, intercellular CO2 concentration, chlorophyll fluorescence parameters, as well as antioxidant levels were hardly affected. It is concluded that presently prevailing O3 levels do not impose severe stress on P. canariensis seedlings.

Previous studies provided no unequivocal evidence demonstrating that field populations of Lumbricus rubellus Hoffmeister (1843), exhibit genetically inherited resistance to As-toxicity. In this study F1, F2 and F3 generation offspring derived from adults inhabiting As-contaminated field soil were resistant when exposed to 2000 mg kg⁻¹ sodium arsenate. The offspring of uncontaminated adults were not As-resistant. Cocoon viability was 80% for F1 and 82% for F2 offspring from As-contaminated adults and 59% in the F1 control population. High energy synchrotron analysis was used to determine whether ligand complexation of As differed in samples of: resistant mine-site adults, the resistant F1 and F2 offspring of the mine-site earthworms exposed to the LC₂₅ sodium arsenate (700 mg kg⁻¹) of the F1 parental generation; and adult L. rubellus from an uncontaminated site exposed to LC₂₅ concentrations of sodium arsenate (50 mg kg⁻¹). XANES and EXAFS indicated that As was present as a sulfur-coordinated species. As-resistance in F1, F2 and F3 offspring of the earthworm Lumbricus rubellus.

We compared heavy metal levels, calcium levels, breeding parameters and condition of nestling and adult Cyanistes caeruleus and Parus major along a heavy metal pollution gradient. Both species started laying earlier and showed inferior nestling growth and smaller fledging probability in the polluted areas, which are phenologically advanced in spring due to sparse forests. The major inter-specific difference in the responses was that the clutch size and hatching success were decreased in the polluted area in P. major, but not in C. caeruleus. Heavy metal profiles in nestling feces were relatively similar in the two species, though Ni and Pb levels were higher in C. caeruleus than in P. major. However, the latter species showed markedly higher fecal calcium concentrations. Lower calcium levels and higher levels of some heavy metals in C. caeruleus suggest that in Ca-deficient environments this species might be more susceptible to negative pollution effects than P. major. The effect of Pb + Zn on coelomocyte riboflavin content in the epigeic earthworm Dendrodrilus rubidus inhabiting three metalliferous soils and one reference soil was measured by flow cytometry and spectrofluorimetry. A reciprocal polluted <-> unpolluted worm transfer experiment (4-week exposure) was also performed. High proportions of autofluorescent eleocytes were counted in worms from all localities, but intense riboflavin-derived autofluorescence was detectable only in reference worm eleocytes. Other findings were: (i) fluorophore(s) other than riboflavin is/are responsible for eleocyte autofluorescence in residents of metalliferous soils; (ii) riboflavin content was reduced in the eleocytes of worms transferred from unpolluted to metal-polluted soil; (iii) the riboflavin content of D. rubidus eleocytes is a promising biomarker of exposure; (iv) COII mitochondrial genotyping revealed that the reference population is genetically distinct from the three mine populations; (v) metal exposure rather than genotype is probably the main determinant of inter-population differences in eleocyte riboflavin status. Soil metal pollution reduces riboflavin content of earthworm eleocytes.

Guidelines provided by the OECD and EPPO allow the use of single-species tests performed in greenhouses to assess the risk of herbicides to non-target terrestrial plant communities in the field. The present study was undertaken to investigate the use of greenhouse data to determine effects of herbicides with a different mode of action on the biomass, seed production and emergence of field-grown plants. In addition, a single species approach was compared with a mixed species approach. Effects on the biomass of greenhouse and field-grown plants were found to be related at different effect levels, indicating that it might be possible to translate results from greenhouse studies to field situations. However, the use of single-species tests may not be valid. The response of a single plant species to sublethal herbicide dosages differed to the response of the same species grown in a mixture with other species. The use of single-species greenhouse tests in the ecological risk assessment of crop protection products may only be valid for single species in the field, not for vegetations. Particle-size-fractioned transfer of dioxins from sediments to water columns by resuspension process was investigated, using supernatant samples obtained from shaking experiments of sediment-water pairs simulating natural disturbances. The concentrations (dry-matter mass basis) of individual compounds (Cfraction) in two particle size fractions (0.1-1 and 1-10 μm) in the supernatants were generally slightly higher than those in the original sediment (Csed). Cfraction/Csed ratios ranged from 0.45 to 5.9 (median 1.5) without consistent differences among congener groups or consistent correlations against the number of chlorine atoms. The dioxin concentrations in the water column associated with the remaining sediment particles can therefore be estimated by those in the original sediment and by the concentration of suspended sediment particles in the water. The concentration of each compound in the remaining sediment particles (mostly 0.1-10 μm in size) can be roughly estimated by multiplying the concentration in the original sediment by 1.5. Dioxin concentrations (dry-matter mass basis) in sediment particles resuspended in the water column were slightly higher than or comparable to those in the bottom sediment.

The amount of toxic metal accumulated by an organism is often taken as an indicator of potential toxicity. We investigated this relationship in the freshwater snail, Lymnaea stagnalis, exposed to 500 μg l⁻¹ Al over 30 days, either alone or in the presence of phosphate (500 μg l⁻¹ P) or a fulvic acid surrogate (FAS; 10 mg l⁻¹ C). Behavioural activity was assessed and tissue accumulation of Al quantified. Lability of Al within the water column was a good predictor of toxicity. FAS increased both Al lability and behavioural dysfunction, whereas phosphate reduced Al lability, and completely abolished Al-induced behavioural toxicity. Tissue accumulation of Al was not linked to toxicity. Higher levels of Al were accumulated in snails exposed to Al + P, compared to those exposed to Al alone, whereas FAS reduced Al accumulation. These findings demonstrate that the degree of tissue accumulation of a metal can be independent of toxicity. Total Al accumulation in the tissues of Lymnaea stagnalis does not provide a direct indication of its toxicity.

Three ex situ collections of poplar clones from natural populations of Populus alba and P. nigra growing in northern Italy were assessed for their genetic dissimilarity (GD) by means of amplified fragment length polymorphism (AFLP). The high GD evidenced within populations was exploited for screening 168 clones in a field trial on heavy metal-polluted soil. After one growth season, clonal differences in plant survival and growth were observed. On the basis of performance, six clones were singled out, and used to evaluate copper and zinc accumulation in different organs. Clonal differences in metal concentrations were most evident for leaves and stems; one clone of P. alba (AL35) had a distinctly higher concentration of both metals in the roots. Leaf polyamine (putrescine, spermidine, spermine) profiles correlated with tissue metal concentrations, depending on the clone, plant organ and metal. In particular, the high metal-accumulating clone AL35 exhibited a dramatically higher concentration of free and conjugated putrescine. Overall, the results indicate that, given the high GD of Populus even within populations, it is possible to identify genotypes best suited for soil clean-up, and useful also for investigating physiological markers associated with high metal accumulation/tolerance High survival rate and heavy metal accumulation are associated with high polyamine concentration in an elite poplar clone.

Among the numerous PCB congeners, most of the dioxin-like PCBs (DL-PCBs) need to be characterized by hyphenated techniques. It has been shown in several instances that these congeners are well related to the total PCB content in fish. We examined datasets collected mainly in France, on freshwater and marine fish and sediments. A statistical model linking DL- and indicator PCBs was developed for a dataset composed of freshwater fishes, and proved to predict well DL-PCBs from indicator PCBs in all other fish sets, including marine ones. Type II error rates remained low in almost all fish sets. A similar correlation was observed in sediments. Non-dioxin-like PCBs elicit various adverse effects and represent 95% of the total PCBs. A European guideline for them is needed; the correlation between DL- and indicator PCBs could help develop this standard in the future. (C) 2009 Elsevier Ltd. All rights reserved.

Whereas concentrations and loads of traditional pollutants (suspended solids, BOD, COD, nutriments) in stormwater discharges are well documented, very few information is available on the presence, the concentrations and the loads of priority substances. The ESPRIT project aims to identify, evaluate, characterise and later on model fluxes of priority pollutants in urban stormwater discharges. Two sites were chosen for measurement campaigns: Ecully (residential site, combined sewer) and Chassieu (industrial site, separate sewer). In total, 26 metals and 36 organic pollutants (in particulate and dissolved phases) are measured in stormwater discharges at the catchment outlets. Some results obtained for a set of rainfall events monitored since the beginning of 2008 are presented. Significant inter-site and inter-event variability in concentrations and specific fluxes (g/active ha) are observed for metals and organics pollutants, particularly for pesticides.
