Influence of sediment composition on PAH toxicity using zebrafish (Danio rerio) and Japanese medaka (Oryzias latipes) embryo-larval assays

International audience | Due to hydrophobic and persistent properties, polycyclic aromatic hydrocarbons (PAHs) have a high capacity to accumulate in sediment. Sediment quality criteria, for the assessment of habitat quality and risk for aquatic life, include understanding the fate and effects of PAHs. In the context of European regulation (REACH and Water Framework Directive), the first objective was to assess the influence of sediment composition on the toxicity of two model PAHs, benzo[a]pyrene and fluoranthene using 10 day-zebrafish embryo-larval assay. This procedure was undertaken with an artificial sediment in order to limit natural sediment variability. A suitable sediment composition might be then validated for zebrafish and proposed in a new OECD guideline for chemicals testing. Second, a comparative study of toxicity responses from this exposure protocol was then performed using another OECD species, the Japanese medaka. The potential toxicity of both PAHs was assessed through lethal (e.g. survival, hatching success) and sublethal endpoints (e.g. abnormalities, PMR and EROD) measured at different developmental stages, adapted to the embryonic development time of both species. Regarding effects observed for both species, a suitable artificial sediment composition for PAH toxicity testing was set at 92.5 % dw silica of 0.2-0.5 mm grain size, 5 % dw kaolin clay without organic matter for zebrafish and 2.5 % dw blond peat in more only for Japanese medaka. PAH bioavailability and toxicity were highly dependent on the fraction of organic matter in sediment and of the Kow coefficients of the tested compounds. The biological responses observed were also dependent of the species under consideration. Japanese medaka embryos appeared more robust than zebrafish embryos for understanding the toxicity of PAHs due to the longer exposure duration and it lower sensitivity of sediment physical properties.

International audience | In the last 10 years, behavior assessment has been developed as an indicator of neurotoxicity and an integrated indicator of physiological disruption. Polycyclic aromatic hydrocarbon (PAH) release into the environment has increased in recent decades resulting in high concentrations of these compounds in the sediment of contaminated areas. We evaluated the behavioral consequences of long-term chronic exposure to PAHs, by exposing zebrafish to diets spiked with three PAH fractions at environmentally relevant concentrations. Fish were exposed to these chemicals from their first meal (5 days postfertilization) until they became reproducing adults (at 6 months old). The fractions used were representative of PAHs of pyrolytic (PY) origin and of two oils differing in composition (a heavy fuel oil (HO) and a light crude oil (LO)). Several tests were carried out to evaluate circadian spontaneous swimming activity, responses to a challenge (photomotor response), exploratory tendencies, and anxiety levels. We found that dietary PAH exposure was associated with greater mobility, lower levels of exploratory activity, and higher levels of anxiety, particularly in fish exposed to the HO fraction and, to a lesser extent, the LO fraction. Finally, our results indicate that PAH mixtures of different compositions, representative of situations encountered in the wild, can induce behavioral disruptions resulting in poorer fish performance.

International audience | We expose here a detailed spatially explicit model of aphid population dynamics at the scale of a whole country (Metropolitan France). It is based on convection-diffusionreaction equations, driven by abiotic and biotic factors. The target species is the grain aphid, Sitobion avenae F., considering both its winged and apterousmorphs. In this preliminary work, simulations for year 2004 (an outbreak case) produced realistic aphid densities, and showed that both spatial and temporal S. avenae population dynamics can be represented as an irregular wave of population peak densities from southwest to northeast of the country, driven by gradients or differences in temperature, wheat phenology, and wheat surfaces. This wave pattern fits well to our knowledge of S. avenae phenology. The effects of three insecticide spray regimes were simulated in five different sites and showed that insecticide sprays were ineffective in terms of yield increase after wheat flowering. After suitable validation, which will require some further years of observations, the model will be used to forecast aphid densities in real time at any date or growth stage of the crop anywhere in the country. It will be the backbone of a decision support system, forecasting yield losses at the level of a field. The model intends then to complete the punctual forecasting provided by older models by a comprehensive spatial view on a large area and leads to the diminution of insecticide sprayings in wheat crops.

Studies on remote, uninhabited, near-pristine reefs have revealed surprisingly large populations of large reef fish. Locations such as the northwestern Hawaiian Islands, northern Marianas Islands, Line Islands, U.S. remote Pacific Islands, Cocos-Keeling Atoll and Chagos archipelago have much higher reef fish biomass than islands and reefs near people. Much of the high biomass of most remote reef fish communities lies in the largest species, such as sharks, bumphead parrots, giant trevally, and humphead wrasse. Some, such as sharks and giant trevally, are apex predators, but others such as bumphead parrots and humphead wrasse, are not. At many locations, decreases in large reef fish species have been attributed to fishing. Fishing is well known to remove the largest fish first, and a quantitative measure of vulnerability to fishing indicates that large reef fish species are much more vulnerable to fishing than small fish. The removal of large reef fish by fishing parallels the extinction of terrestrial megafauna by early humans. However large reef fish have great value for various ecological roles and for reef tourism.

The Water Framework Directive uses the “one-out, all-out” principle in assessing water bodies (i.e., the worst status of the elements used in the assessment determines the final status of the water body). In this study, we assessed the ecological status of two coastal lakes in Italy. Indices for all biological quality elements used in transitional waters from the Italian legislation and other European countries were employed and compared. Based on our analyses, the two lakes require restoration, despite the lush harbor seagrass beds, articulated macrobenthic communities and rich fish fauna. The “one-out, all-out” principle tends to inflate Type I errors, i.e., concludes that a water body is below the “good” status even if the water body actually has a “good” status. This may cause additional restoration costs where they are not necessarily needed. The results from this study strongly support the need for alternative approaches to the “one-out, all-out” principle.

In order to test the relationship between the occurrence of marine debris and the distance from urban areas, nine beaches in the metropolitan area of Salvador, Bahia, Brazil and the adjacent northern coast were studied. Marine debris were collected, sorted in several categories and weighed. It was observed that plastics were numerically the most abundant component of the collected debris. As expected, the beaches closest to Salvador presented the largest density of debris, with the exception of the Porto da Barra beach, which has an efficient public cleaning system and does not have any vegetation, making it difficult toaccumulate solid waste. Linear regression analyses showed significant relationships between the distance from the urban center (Salvador) and the number of marine debris per m2, the total number of debris per beach (abundance), and the diversity of debris types (richness). The results showed that proximity to urban regions was a key factor in the marine debris distribution along the coast.

The BP MC252 well failure in the Gulf of Mexico, April 2010 caused concern for crude oil and polycyclic aromatic hydrocarbon (PAHs) exposure along the sandy beaches of the Florida Panhandle. We began collections of Coquina clams (Donax spp.) from the surf zone of Florida Panhandle beaches to monitor PAH contamination to compliment analysis of surf zone sand samples. These clams had higher levels of PAHs relative to ambient sand, and this allowed us to continue to monitor PAH levels after sand concentrations fell below limits of detection. PAH levels in the Coquina tissues were highly variable, perhaps indicative of the heterogeneous distribution of oil and tar on the beaches and exposure to tar particles. Overall, PAH levels decreased continuously in both sand and Coquina tissues, reaching limits of detection within one and two years respectively after oil landed on Florida Panhandle beaches. Our work suggests these surf zone molluscs may be used to monitor pollutant exposure along high energy sandy beach shorelines.

The Florida Panhandle continental shelf environment was exposed to oil from the BP oil well failure in the Gulf of Mexico during 2010. Floating mats of oil were documented by satellite, but the distribution of dissolved components of the oil in this region was unknown. Shipek® grab samples of sediments were taken during repeated cruises between June 2010 and June 2012 to test for selected polycyclic aromatic hydrocarbons (PAHs) as indicators of this contamination. Sediments were collected as composite samples, extracted using standard techniques, and PAHs were quantified by GC/MS-SIM. PAHs in samples from the continental slope in May 2011 were highest near to the failed well site and were reduced in samples taken one year later. PAHs from continental shelf sediments during the spill (June 2010) ranged from 10 to 165ngg−1. Subsequent cruises yielded variable and reduced amounts of PAHs across the shelf. The data suggest that PAHs were distributed widely across the shelf, and their subsequent loss to background levels suggests these compounds were of oil spill origin. PAH half-life estimates by regression were 70–122days for slope and 201days for shelf stations.

Terrestrial sediments are a significant stressor on coastal ecosystems, with both suspended and deposited sediment having adverse effects on aquatic organisms. However, information on the effect of suspended sediments on fertilization success for urchin species is lacking. Using sediment levels similar to those encountered in situ, a controlled experiment was conducted to test whether suspended sediment affects fertilization success in the urchin Evechinus chloroticus. Analyses used generalized linear mixed models (GLMMs) and hierarchical Bayesian (HB) regression. Both approaches showed a significant decrease in fertilization success with increased suspended sediment levels. Uncertainties in estimates were narrower for HB models, suggesting that this approach has advantages over GLMMs for sparse data problems sometimes encountered in laboratory experiments. Given future global change scenarios, this work is important for predicting the effects of stressors such as sedimentation that may ultimately impact marine populations.

With a coastline of 8500km, Brazil has 34 public ports and various private terminals, which together in 2012 handled 809 million tonnes of goods. The solid wastes produced (from port activities, ships and cargoes) pose a highly relevant problem, both due to the quantity and diversity, requiring a complex and integrated set of practices resulting from legal requirements and proactive initiatives. The main Brazilian law on solid waste management is recent (Law 12,305/2010) and the specific rules on solid waste in ports are badly in need of revision to meet the challenges caused by expansion of the sector and to harmonize them with the best global practices. This paper analyzes the current legal/regulatory framework for solid waste management at Brazilian ports and compares this structure with the practice in Europe. At the end, we suggest initiatives to improve the regulation of solid wastes at Brazilian ports.
