Isolation and characterization of two crude oil-degrading yeast strains, Yarrowia lipolytica PG-20 and PG-32, from the Persian Gulf

Among six crude oil-degrading yeasts that were isolated from an oil-polluted area in the Persian Gulf, two yeast strains showed high degradation activity of aliphatic hydrocarbons. From an analysis of 18S rRNA sequences and biochemical characteristics, these strains were identified as Yarrowia lipolytica strains PG-20 and PG-32. Gas Chromatography (GC) analysis of the crude oil remaining in the culture medium after 1week at 30°C showed that the strains PG-20 and PG-32 degraded 68% and 58% of crude oil, respectively. The optimal growth condition and biodegradation of hydrocarbons was in ONR medium with an acidic pH (pH5). These two strains may degrade aliphatic hydrocarbons more efficiently than aromatic hydrocarbons, although strain PG-20 had better degradation than strain PG-32. The two Y. lipolytica strains reduce surface tension when cultured on hydrocarbon substrates (1% v/v). These strains showed a cell surface hydrophobicity higher than 70%. These results suggested that Y. lipolytica strains PG-20 and PG-32 have high crude oil degrading activity due to their high emulsifying activity and cell hydrophobicity. In conclusion, these yeast strains can be useful for the bioremediation process in the Persian Gulf and decreasing oil pollution in this marine ecosystem.

This paper describes details of an oil spill model, OILTRANS, developed by the authors. The model is an off-line particle-transport model coupled to the most up to date operational met-ocean model forecasts. Formulations for the dominant oil fate processes of spreading, advection, diffusion, evaporation, emulsification and dispersion have been encoded, providing the model with the ability to accurately predict the horizontal movement of surface oil slick, the vertical entrainment of oil into the water column and the mass balance of spilled oil. The application of the OILTRANS model to an accidental release during a ship-to-ship fuel transfer in the Celtic Sea in February 2009 is presented to validate the system. Comparisons with aerial observations of the oil slick at the time of the incident, and subsequent model simulations, indicate that the OILTRANS model is capable of accurately predicting the transport and fate of the oil slick.

In the present study, persistent organic pollutants (POPs) were determined in 25 livers from Magellanic penguins, Spheniscus magellanicus, found on the state of São Paulo and Rio de Janeiro (Brazil). The following concentrations of POPs (wet weight) were found: ∑ PCBs: 18.9–775.8ngg⁻¹; ∑ DDTs: 2.3–275ngg⁻¹; and ∑ HCHs: 1.0–11.8ngg⁻¹. Among the PCBs, there was a predominance of hexachlorobiphenyls (138 and 153) and heptachlorobiphenyls (180 and 187). Among the organochlorines, DDT predominated. Through histopathologic exams a positive correlation was found between HCB and cardiovascular failure (p=0.012), as HCB values (wet weight) were 6.33–96.49ngg⁻¹ for positive animals and 2.45–19.63ngg⁻¹ for negative animals. Studies on POPs in Magellan penguins contribute to a better understanding of the distribution and trends of these pollutants in the western South Atlantic Ocean.

Solar ultraviolet radiation (UVR, 280–400nm) is known to inhibit the photosynthesis of macroalgae, whereas nitrogen availability may alter the sensitivity of the algae to UVR. Here, we show that UV-B (280–315nm) significantly reduced the net photosynthetic rate of Gracilaria lemaneiformis. This inhibition was alleviated by enrichment with ammonia, which also caused a decrease in dark respiration. The presence of both UV-A (315–400nm) and UV-B stimulated the accumulation of UV-absorbing compounds. However, this stimulation was not affected by enrichment with ammonia. The content of phycoerythrin (PE) was increased by the enrichment of ammonia only in the absence of UVR. Ammonia uptake and the activity of nitrate reductase were repressed by UVR. However, exposure to UVR had an insignificant effect on the rate of nitrate uptake. In conclusion, increased PE content associated with ammonia enrichment played a protective role against UVR in this alga, and UVR differentially affected the uptake of nitrate and ammonia.

Polyethylene pellets provide a convenient means to monitor Persistent Organic Pollutants (POPs) in marine systems. Pellets collected between 1984 and 2008 at three South African beaches were analysed for PCB, HCH and DDT. Concentrations of all three POPs decreased over the last two decades, although this signal was less clear for PCBs, and further monitoring is needed to assess trends in this family of compounds. DDT concentrations at two sites were higher than previous records for southern Africa, but there is no evidence of a link to the ongoing use of DDT for malaria control. HCHs concentrations were lower than in pellets from the east coast of southern Africa, suggesting that this pesticide was mainly used in the eastern part of the region. Our study demonstrates the potential for International Pellet Watch to track temporal as well as geographical patterns in the abundance of POPs in marine environments.

This study evaluated the toxicity of dispersant application which is, in nearshore area, a controversial response technique to oil spill. Through an experimental approach with juveniles of Liza aurata, the toxicity of five exposure conditions was evaluated: (i) a chemically dispersed oil simulating dispersant application; (ii) a single dispersant as an internal control of chemically dispersed oil; (iii) a mechanically dispersed oil simulating natural dispersion of oil; (iv) a water soluble fraction of oil simulating an undispersed and untreated oil slick and (v) uncontaminated seawater as a control exposure condition. The relative concentration of PAHs (polycyclic aromatic hydrocarbons) biliary metabolites showed that the incorporation of these toxic compounds was increased if the oil was dispersed, whether mechanically or chemically. However, toxicity was not observed at the organism level since the aerobic metabolic scope and the critical swimming speed of exposed fish were not impaired.

Scroby Sands offshore wind farm was built close to a haul-out and breeding site for harbour seal, a species of conservation concern. An aerial survey programme conducted during a five-year period spanning wind farm construction, revealed a significant post-construction decline in haul-out counts. Multivariate model selection suggested that the decline was not related to the environmental factors considered, nor did it mirror wider population trends. Although cause and effect could not be unequivocally established, the theoretical basis of hearing in pinnipeds and previous studies suggested that extreme noise (to 257dB re 1μ Paₚₚ @ 1m) generated by pile-driving of turbine bases led to displacement of seals. A lack of full recovery of harbour seal during the study was also linked to their sensitivity to vessel activity and/or rapid colonisation of competing grey seal. Any impact of offshore wind farm development upon pinnipeds would be much reduced without pile-driving.

This study employed three chemometric data mining techniques (factor analysis (FA), cluster analysis (CA), and discriminant analysis (DA)) to identify the latent structure of a water quality (WQ) dataset pertaining to Kinta River (Malaysia) and to classify eight WQ monitoring stations along the river into groups of similar WQ characteristics. FA identified the WQ parameters responsible for variations in Kinta River’s WQ and accentuated the roles of weathering and surface runoff in determining the river’s WQ. CA grouped the monitoring locations into a cluster of low levels of water pollution (the two uppermost monitoring stations) and another of relatively high levels of river pollution (the mid-, and down-stream stations). DA confirmed these clusters and produced a discriminant function which can predict the cluster membership of new and/or unknown samples. These chemometric techniques highlight the potential for reasonably reducing the number of WQVs and monitoring stations for long-term monitoring purposes.

The impact of typical pollutants upon the fish-farming was assessed by use of a battery of biomarkers in two typical marine aquaculture regions in South China. Biotransformation parameters including 7-ethoxyresorufin-O-deethylase (EROD), aminopyrine N-demethylase (APND), erythromycin N-demethylase (ERND), glutathione-S-transferase (GST) and Malondialdehyde (MDA) were measured in five cultured fish species. Pollutants such as polycyclic aromatic hydrocarbon (PAHs), organochlorinated compounds (OCs), heavy metals and antibiotics (quinolones and sulfonamides) in sediments were characterized. Higher pollutant residue levels were observed in Dapeng Cove. EROD, APND and ERND activities were lower in fish from Dapeng Cove compared with fish from Hailing Island, while it is just on the contrary for GST and MDA. ERND, APND and GST showed sensitivity corresponding to different pollutants. Small fish species seemed to exhibit more sensitive to pollutants. The study further supports usefulness of multi-biomarker approach considering multiple species to define the effects of anthropogenic inputs in marine aquaculture systems.

Shipping is almost certainly the most prevalent human-mediated transport vector for non-indigenous species (NIS) within the marine environment. The Royal Australian Navy (RAN) has long acknowledged the importance of sound environmental management and in recent years has taken a proactive approach to addressing risks associated with marine biosecurity. primarily as a result of biofouling on Navy vessel returning from overseas operations. This paper describes two case studies that highlight the effectiveness of the RAN marine biosecurity management framework in identifying an unwanted marine species on Navy vessels, and the successful biosecurity management program that ensued. In particular, the early detection and identification of a suspect NIS, the quick response to the discovery and the collaborative approach adopted between the RAN and the Government regulatory agency (Western Australian Department of Fisheries) charged with coordinating the incursion response serves as a model for how future incursion responses should be reported and managed.
