Toxicity prediction and effect characterization of 90 pharmaceuticals and illicit drugs measured in plasma of fish from a major European river (Sava, Croatia) Texte intégral

Chemical analysis of plasma samples of wild fish from the Sava River (Croatia) revealed the presence of 90 different pharmaceuticals/illicit drugs and their metabolites (PhACs/IDrgs). The concentrations of these PhACs/IDrgs in plasma were 10 to 1000 times higher than their concentrations in river water. Antibiotics, allergy/cold medications and analgesics were categories with the highest plasma concentrations. Fifty PhACs/IDrgs were identified as chemicals of concern based on the fish plasma model (FPM) effect ratios (ER) and their potential to activate evolutionary conserved biological targets. Chemicals of concern were also prioritized by calculating exposure-activity ratios (EARs) where plasma concentrations of chemicals were compared to their bioactivities in comprehensive ToxCast suite of in vitro assays. Overall, the applied prioritization methods indicated stimulants (nicotine, cotinine) and allergy/cold medications (prednisolone, dexamethasone) as having the highest potential biological impact on fish. The FPM model pointed to psychoactive substances (hallucinogens/stimulants and opioids) and psychotropic substances in the cannabinoids category (i.e. CBD and THC). EAR confirmed above and singled out additional chemicals of concern - anticholesteremic simvastatin and antiepileptic haloperidol. Present study demonstrates how the use of a combination of chemical analyses, and bio-effects based risk predictions with multiple criteria can help identify priority contaminants in freshwaters. The results reveal a widespread exposure of fish to complex mixtures of PhACs/IDrgs, which may target common molecular targets. While many of the prioritized chemicals occurred at low concentrations, their adverse effect on aquatic communities, due to continuous chronic exposure and additive effects, should not be neglected.

Disastrous oil spills cause severe environmental issues. The shortcomings of current cleaning methods for remediating oil have prompted the latest research drive to create intelligent nanoparticles that absorb oil. We, therefore, synthesized 197 ± 50 nm floatable photoreactive hybrid nanoparticles with Ag–TiO₂ plasmonic photocatalyst (Eg = 3.08 eV) content to eliminate interfacial water pollutants, especially toluene-based artificial oil spill. We found that the composite particles have non-wetting properties in the aqueous media and float easily on the surface of the water due to the moderate hydrophobic nature (Θ = 113°) of the matrix of polystyrene, and these properties lead to elevated absorption of the interfacial organic pollutants (e.g., mineral oil). We showed that (28.5 mol%) divinylbenzene cross-linker content was required for adequate swelling capacity (2.15 g/g), whereas incorporated 15.8% Ag–TiO₂ content in the swollen particles was enough for efficient photodegradation of the artificial oil spill under 150 min LED light (λₘₐₓ = 405 nm) irradiation. The swollen polymer particles with embedded 32 ± 7 nm Ag–TiO₂ content increase the efficiency of photooxidation by increased the direct contact between both the photocatalysts and the artificial oil spill. Finally, it was also presented that the composite particles destroy themselves: after approximately one and a half months of continuous LED light irradiation, the organic polymer component of the composite was almost completely (88.5%) photodegraded by the incorporated inorganic photocatalyst particles.

Cadmium (Cd) is a heavy metal of concern in contaminated sites because of its high toxicity to soil biota and humans. Typically, Cd exposure is thought to be dominated by dissolved Cd in soil pore water and, thus, dermal uptake. In this study, we investigated the uptake, toxicity, and maternal transfer of Cd in a standard soil invertebrate, the oribatid mite (Oppia nitens), which is common to boreal and temperate ecozones. We found total soil Cd predicted Cd uptake in adult and juvenile O. nitens with no significant uptake from pore water by juvenile mites. Cadmium significantly inhibited juvenile production and recruitment as well as reduced adult fecundity. Adult O. nitens maternally transferred 39–52% of their Cd body burden to juveniles (tritonymphs) while the maternally-acquired Cd accounted for 41% of the juvenile internal Cd load. Our results suggest that dermal adsorption of metal ions is not important for O. nitens and that maternal transfer of Cd in soil invertebrates has ecological and toxicological implications for populations of soil invertebrates. Maternal transfer should be incorporated as a criterion in setting environmental soil quality guidelines (SQGE) for cadmium and other non-essential heavy metals.

Freshwater systems serve as important sources and transportation routes for marine microplastic pollution, and inadequate attention has been paid to this situation. Data on microplastic pollution of typical seagoing rivers in northern China are lacking. In the current study, we investigated the distribution and characteristics of microplastics in the main stream of the Haihe River, which flows through a metropolis with a high population density and level of industrialization and then flows into the Bohai Sea. The microplastic samples were collected by manta trawls with pore sizes of 333 μm, and the microplastic concentrations ranged from 0.69 to 74.95 items/m³. Fibers dominated in the surface water of the Haihe River; their shapes that were categorized as fibers, film, foam, fragments, and spheres, and contributed 17.4–86.7% of the total microplastics studied. The size distribution of the microplastics was concentrated in a range of 100–1000 μm, with 54.7% of the total sizes corresponding to the 333-μm trawl. Micro-Fourier transform infrared (μ-FT-IR) spectra showed that the main components were polyethylene, poly(ethylene-propylene) copolymer, and polypropylene. Scanning electron microscopy-energy dispersive spectroscopy (SEM-EDS) measurements revealed scratches, micropores, and cracks on the surfaces of the microplastics due to mechanical friction, chemical oxidation and degradation processes. The results of this study confirmed the high abundance and high diversity of microplastics in an urban river and indicated appreciable impacts from point-source inputs on the microplastic pollution, such as effluents from wastewater treatment plants (WWTPs).

This study presents the results of an integrated model developed to evaluate the environmental and health impacts of Electric Vehicle (EV) deployment in a large metropolitan area. The model combines a high-resolution chemical transport model with an emission inventory established with detailed transportation and power plant information, as well as a framework to characterize and monetize the health impacts. Our study is set in the Greater Toronto and Hamilton Area (GTHA) in Canada with bounding scenarios for 25% and 100% EV penetration rates. Our results indicate that even with the worst-case assumptions for EV electricity supply (100% natural gas), vehicle electrification can deliver substantial health benefits in the GTHA, equivalent to reductions of about 50 and 260 premature deaths per year for 25% and 100% EV penetration, compared to the base case scenario. If EVs are charged with renewable energy sources only, then electrifying all passenger vehicles can prevent 330 premature deaths per year, which is equivalent to $3.8 Billion (2016$CAD) in social benefits. When the benefit of EV deployment is normalized per vehicle, it is higher than most incentives provided by the government, indicating that EV incentives can generate high social benefits.

Toluene is a highly volatile organic solvent present in gasoline. Exposure mainly occurs by absorption via the pulmonary tract and easily reaches the central nervous system, which causes toxic effects. Toluene toxicity has been described but not well established. The present work aimed to evaluate the effects of airborne exposure to toluene, the in vivo model Caenorhabditis elegans was assessed to determine whether nematode could be used to evaluate the effects of exposure to toluene and the possible mechanisms of toxicity of the solvent. Worms at the first or fourth larval stages were exposed to toluene for 48 or 24 h, respectively, in a laboratory-developed vapor chamber at concentrations of 450, 850, 1250 and 1800 ppm. We observed increases in worm mortality and significant developmental delays that occurred in a concentration-dependent manner. An increased incidence of apoptotic events in treated germline cells was shown, which was consistent with observed reductions in reproductive capacity. In addition, toluene promoted significant behavioural changes affecting swimming movements and radial locomotion, which were associated with changes in the fluorescence intensity and morphology of GABAergic and cholinergic neurons. We conclude that toluene exposure was toxic to C. elegans, with effects produced by the induction of apoptosis and neuronal damage.

Acid rain is a serious threat to terrestrial ecosystems. To provide more accurate early warning information for acid rain prevention, urban planning, and travel planning, a novel air pollutant prediction model was proposed in this paper to predict NO₂ and SO₂. First, the data were decomposed into several sub-sequences by a complete ensemble empirical mode decomposition with adaptive noise. Second, the subsequences are reconstructed by variational mode decomposition and sample entropy. Then, the new subsequences are predicted by the extreme learning machine combined with the whale optimization algorithm. The empirical analysis was carried out through 8 data sets. According to the experimental results, three main conclusions can be drawn. First, the proposed model in this paper has excellent prediction performance and robustness. In all the comparison experiments, the R² and RMSE of the proposed model are the best among all the models. Second, data preprocessing is very necessary. After adding the decomposition algorithm, the average improvement levels of R² and RMSE were 897.57% and 50.78%, respectively. Third, the re-decomposition of IMF1 is an effective method to improve prediction accuracy. After the re-decomposition of IMF1, R² can be improved by 13.64% on average on the original basis, and RMSE can be reduced by 31.99% on average. The results of this study can provide a valuable reference for the research of air pollutant prediction. In future work, the application of the proposed model in other air pollutants or other regions can be explored.

A compound system involving three matrices (water, sediment, and paddy soil) was conceived to determine the potential sources of metal(loid)s (Ti, Fe, Co, Ni, Cu, Zn, As, Cd, Pb, and U) and synthetically evaluate their pollution levels in the Le’an River basin. The result indicated that the established background values (BVs) of paddy soil and sediment in the compound system were obviously higher than those of the upper continental crust (UCC) and soils from Jiangxi Province, a difference which was especially marked for sediment. The concentrations of Cu, Zn, As, Cd in the system had high coefficients of variation (CVs), and metal(loid)s in sediment showed higher levels than those in paddy soil, except for Pb. Cd and Cu in the system had the highest Ef levels, which probably pose a high risk to organisms and the health of local residents. There were significantly linear relationships between the site rank index (SRI) for water and that for sediment or paddy soil, revealing that matrices in the system interacted with each other. Principal component analysis (PCA) and absolute principal component scores and multiple linear regression model (APCS-MLR) results demonstrated that Cu, Zn, As, Cu, Pb, and U enrichments in the system were mainly affected by mining activities and were predominately deposited in sediment. Point pollution sources rather than non-point pollution sources such as mining activities, contributed most of the anthropogenic metal(loid)s to sediment. Both SRI and Hierarchical cluster analysis (HCA) results visually showed that S5, S8, S9, S10, S11, and S12 severe pollution grouped together and scattered through areas with extensive mining activities, while other sites with moderate pollution were spread along the main stream of the Le’an River.

Information on the occurrence and trophodynamics of per- and polyfluoroalkyl substances (PFASs) in the Antarctic region is limited. We investigated the occurrence of PFASs in an ecosystem in the Fildes Peninsula at King George Island and Ardley Island, Antarctica. The profiles, spatial distribution, and trophic transfer behavior of PFASs were further studied. ∑PFASs ranged from 0.50 ± 38.0 ng/g dw (dry weight) in algae to 4.97 ± 1.17 ng/g dw in Neogastropoda (Ngas), which was lower than those in the low- and mid-latitude regions and even Arctic regions. Perfluorobutyric acid (PFBA) was predominant with detection frequencies above 50% in all types of samples, and the relative contribution of PFBA ranged from 22% to 57% in the biota samples. The biomagnification factors of PFBA, perfluoroheptanoate (PFHpA), perfluorohexane sulfonate (PFHxS), and perfluorooctane sulfonate (PFOS) between Archaeogastropoda (Agas) and Ngas were 0.67 ± 0.54, 0.77 ± 0.38, 1.04 ± 1.56, 3.30 ± 4.07, and 1.61 ± 0.89, respectively. The trophic magnification factors of PFHxS and PFOS were 2.09 and 2.92, respectively, which indicated that they could be biomagnified through the food chain. Considering the increasing production and uncertain toxicological risks of emerging PFASs and the sensitive ecosystems in Antarctic regions, more attention should be paid, especially for the short-chain ones in the Antarctic region.

Phoxim, a broad-spectrum organophosphate pesticide, is widely used in agriculture to control insect pests in vegetable crops as well as in farm mammals. However, the indiscriminate use of phoxim has increased its release into the environment, leading to the contamination of plant-based foods such as vegetables. In this study, we investigated the effect of Trichoderma asperellum (TM, an opportunistic fungus) on phoxim residue in tomato roots and explored the mechanisms of phoxim metabolism through analysis of detoxification enzymes and gene expression. Degradation kinetics of phoxim showed that TM inoculation rapidly and significantly reduced phoxim residues in tomato roots. Phoxim concentrations at 5d, 10d and 15d post treatment were 75.12, 65.71 and 77.45% lower in TM + phoxim than only phoxim treatment, respectively. The TM inoculation significantly increased the glutathione (GSH) content, the activity of glutathione S-transferase (GST) and the transcript levels of GSH, GST1, GST2 and GST3 in phoxim-treated roots. In addition, the activity of peroxidase and polyphenol peroxidase involved in the xenobiotic conversion also increased in TM + phoxim treatment. The expression of detoxification genes, such as CYP724B2, GR, ABC2 and GPX increased by 3.82, 3.08, 7.89 and 2.46 fold, respectively in TM + phoxim compared with only phoxim. Similarly, the content of ascorbate (AsA) and the ratio of AsA to dehydroascorbate increased by 45.16% and 57.34%, respectively in TM + phoxim-treated roots. Our results suggest that TM stimulates plant detoxification potential in all three phases (conversion, conjugation and sequestration) of xenobiotc metabolism, leading to a reduced phoxim residue in tomato roots.
