A Morphological and Histological Investigation of the Sinus Interdigitalis in Konya Merino Sheep

In the study, it was aimed to reveal the morphological, morphometric and histological characteristics of sinus interdigitalis found in the fore and hind feet of Konya merino sheep. For this purpose, the fore and hind feet of 10 Konya merino sheep, weighing between 44-79 kg, were used. Sinus interdigitalis were dissected out from the feet, and after morphometric measurements were made, morphological examinations were performed and photographed. The shape of the sinus interdigitalis, present in all the forefeet and hind feet, resembled a pipe consisting of neck and body. It was determined that the neck part was longer than the body at all feet. The inner surface of the gland was covered with hair, and there was more hair on the neck than on the body. The weight of the gland, body length and diameter, flexura, canal length and diameter were measured morphometrically. In the measurements made, it was determined that all the values of the forefeet were higher than the hind feet. In histological examination, it was found that the wall of the sinus interdigitalis consists of three layers: epidermis, dermis and fibrous capsule, and in the dermis, hair follicles, sebaceous glands, m. arrector pili and sweat glands were found.

In this study, electric yellow cichlid were (Labidochromis caeruleus) exposed to acute cadmium (Cd) concentrations (0, 50, 60, 70, 80 mg l-1) for 72 h, and chronic Cd concentrations (0.00, 0.25, 0.50, 1.00, 2.00 mg l-1) for 20 d to examine histological alternation. In addition to the recovery was detected after 10 d. After chronic trial, rest of the fish were exposed to just water not containing cadmium for 10 day for recovery. Gills, liver, muscle, and spleen samples were collected from randomly selected fish. The tissue samples prepared with standard techniques for haematoxylin and eosin (H&E). Cadmium depending on concentrations increase caused severe disorders on fish. After acute exposure, several changes were detected such as edema, hyperplasia, breakdown in epithelial tissues of gills; vacuolation and necrosis in liver; increase of hemosterin clustering and necrosis in spleen. After chronic exposure, some symptoms were also found such as edema, aneurysm, degermation of secondary lamellae in gills; degeneration and necrosis in liver; degeneration and granulomas in spleen. Additionally, after recovery of chronic Cd exposure some of the same symptoms were observed such as edema, hyperplasia and aneurysm in gills; vacuolar degeneration and necrosis in liver; granulomas and degeneration in spleen.

In this study, testicular and ovarian structures of economically important edible Pelophylax ridibundus (Pallas, 1771) were histologically examined before and after reproduction in male and female individuals. Fourty eight (24 ♀, 24 ♂) adult frogs were collected from Gölbaşı Lake in Hatay. The average weight and length values of female frogs collected from nature were found to be 56.61±19.59 g and 79.54±7.07 mm; while, the average weight and length values of male frogs were 36.63±12.84 g and 69.29±9.15 mm, respectively. Frogs were brought to the frog farm established in Aydıncık and placed in breeding ponds with a width of 1m2. Frogs in the ponds were brought to the laboratory of Iskenderun Technical University in different periods, before breeding (March) and after breeding (June). Then, histological samples were taken from ovary and testis. The female frogs were determined ready for reproduction. Moreover, a large number of mature oocytes in the before breeding ovaries in vitellogenic stage, while after reproduction oocytes in primary structure and oocytes which have atresia status observed. Also, increase in the thickness of the theca layer was determined. In the male frog seminiferous tubules containing a large number of spermatogonia, spermatocyte, spermatid and a small number of spermatozoons including sperm bundles and leydig cells were found before reproduction. After the reproduciton, the density of spermatogonia, spermatocyte and spermatids were decreased; while, the density of spermatozoon and sperm bundle were increased in the seminiferous tubules. This study will contribute to the determination of mating and spawning in frog breeding by revealing the histological status of the gonad structure of P. ridibundus in the breeding process.

Stress parameter changes (cortisol, glucose, sodium and chloride) and histological alterations (gills, liver, kidney, and spleen) in Asian Catfish (Pangasius hypophthalmus) in transport were investigated. In this study, three separated trials were fulfilled. These were without tricaine, 40 and 60 mg L-1 of tricaine, respectively. Each trial consisted of 4 stages, control (C), transfer (AT), transport (TP) and recovery (24 h). After each stage, blood samples were taken and then, gills, liver, kidney, and spleen samples were collected for histological examination. In the transport without tricaine, the fish glucose increased from 50-60 mg dL-1 to 70-80 mg dL-1 and cortisol increased from 30 ng dL-1 to 90 ng dL-1 in. Sodium was 127-132 meq L-1 in the AT and TP, while 138 meq L-1 in the C and recovery. Cloride changed 103 meq L-1 in the TP, while approximately 107 meq L-1 in C, AT and recovery in the transport without tricaine. In the second trial, the fish glucose increased from 50-65 mg dL-1 to 80-90 mg dL-1 cortisol decreased from 60-70 ng dL-1 to 35-45 ng dL-1 in the transport with 40 mg L-1 of tricaine. Sodium was 130-140 meq L-1 in the C and recovery, while 125 meq L-1 in the AT and TP. Chloride was about 110 meq L-1 in the C, AT and recovery, while 102 meq L-1 in the TP in the transport with 40 mg L-1 of tricaine. In the third trial, the fish glucose was 40-55 mg dL-1 in the C, AT and TP, while 25-30 mg dL-1 in the recovery. Cortisol was 35-40 ng dL-1 in the C and AT, while 25 ng dL-1 in the TP and recovery. Sodium was 126-128 meq L-1 in the C, AT and recovery, while about 119-122 meq L-1 in the TP. Chloride was 110 meq L-1 in the C, AT and recovery, while 104-106 meq L-1 in the TP in the transport with 60 mg L-1 of tricaine. The end of the experiment, it is realized that the usage of the tricaine had positive effects on Asian catfish transport.

The objective of this study was to test the effects of dietary natural zeolite (clinoptilolite) on performance, skin and liver histology in rats. In this study, 24 10-week-old, weaned, adult male Spraque-Dawley rats with approximately 306 +- 18.93 g initial live weight were used. The rats were randomly divided into four groups with three replicates, including a control group and groups with one of three doses of clinoptilolite (2%, 4%, and 6%) in their diets. All the rats were fed these concentrates throughout the experimental period of 56 days. The animals were reared individually in stainless steel cages. There were no significant differences in the primary and secondary follicle numbers among groups, but the diameters of each follicle were found to be significant. The primary and secondary follicle numbers and diameters ranged from lowest to highest as follows: 2.00-2.33, 4.50-7.17; and 11.53-20.42, 57.63-102.12um, respectively. The differences occurred between the control group and group IV (containing 6% zeolite). In addition, the skin and liver histology results showed that there were no differences among the groups.

Tissue monitoring generally includes the stages of fixation, dehydration, clearing, hardening (infiltration), paraffin blocking/paraffin(emmeding), sectioning, water removal, routine staining, and mounting. Fixation is the basic and first step in the microscopic examination of tissues. The histotechnical process, which includes components such as detection, tissue tracking and staining, basically aims to capture and visualize the state of the relationships between tissue parts inside and outside the cell and various cells at a certain time as close as possible to the living state. Maintaining the natural structure of the tissue is important for the follow-up phase. The main feature of a good fixative should protect the sample and make the macromolecules insoluble without changing the chemistry of the sample studied and allowing it to be examined as closely as possible to its living state. In histological tissue analysis including light microscope and electron microscope techniques, an appropriate fixation method is selected for each study. Detection solutions are classified in terms of content. The most commonly used fixative in light microscopic follow-up procedures is 10% formaldehyde. For the electron microscope, the gluteraldehyde-osmium tetraoxide binary is widely used for fixation purposes. Gluteraldehyde acts more slowly and is more expensive than formaldehyde. Formalin is obtained by dissolving formaldehyde in water. In addition, the fixed samples can be stored in the solution for months. With a successful fixation process, the structural properties of the tissue are preserved and thus it is possible to examine the tissue as closely as possible. Thus, better quality sections are obtained from the tissue samples taken. For this reason, it will be more efficient to interpret well-fixed samples by photographing them. In this review, which was created by using various sources, the elements to be considered for an ideal fixation were determined and it was aimed to provide an overview of successful fixation for light microscope and electron microscope.
