Serological Detection of Three Common Tomato Viruses in Tomato Cultivation Areas of Ereğli District, Konya Texte intégral

In this study, it was aimed to reveal some viruses that are a problem in tomato production areas in the Ereğli district of Konya, where tomato production is common. For this reason, in the 2019 production season, leaf samples were taken from 160 tomato plants showing symptoms similar to virus disease symptoms such as mosaic, deformations, yellowing, blight on the leaves and stunted plants. Possible infections of Cucumber mosaic virus (CMV), Tomato spotted-wilt virus (TSWV) and Tomato mosaic virus (ToMV) on these leaves were tested by serological-based DAS-ELISA (Double Antibody Sandwich- Enzyme Linked Immuno Sorbent Assay) method. According to DAS-ELISA results, it was determined that 66.25% of the samples were infected with at least one of these three viruses. In addition, TSWV was detected as the most common virus with a 48.75% relative infection rate in the samples, followed by ToMV (28.75%) and CMV (26.25%). Mixed infections of the viruses were also detected in the leaf samples tested. CMV + TSWV double infection was detected in 22 samples, ToMV + TSWV double infection was detected in 14 samples, and CMV + ToMV + TSWV triple infection was detected in 12 plant samples.

In this study, it was aimed to reveal some viruses that are a problem in Dahlia plants grown for ornamental plants in parks and home gardens in Selçuklu, Karatay and Meram districts of Konya. For this reason, in the 2019 summer, leaf samples were taken from 152 Dahlia plants showing symptoms similar to virus disease symptoms such as mosaic, deformations, yellowing, blight on the leaves and stunted plants. Possible infections of Cucumber mosaic virus (CMV), Tomato spotted-wilt virus (TSWV) and Impatiens necrotic spot virus, (INSV) on these leaves were tested by serological-based DAS-ELISA (Double Antibody Sandwich- Enzyme Linked Immuno Sorbent Assay) method. According to DAS-ELISA results, it was determined that 46.71% of the samples were infected with at least one of these three viruses. In addition, TSWV was detected as the most common virus with a 38.82% relative incidence in the samples, followed by CMV (27.63%). Mixed infections of the viruses were also detected in the leaf samples tested. As a result of the tests, only CMV + TSWV double infection was detected in a total of 30 samples. On the other hand, INSV infection was not detected in any of the leaf samples tested. As a result of survey studies, the average of prevalence rate of virus diseases in Dahlia plants grown in Konya was calculated as 69.56%. Moreover, it was determined that virus diseases were most common in Selçuklu district (75.47%). This is the first report of CMV and TSWV on Dahlia spp. in Türkiye.

Bu çalışma, İstanbul ili Adalar İlçesinde (Büyükada, Heybeliada, Kınalıada, Burgazada) hobi bahçeleri ve peyzaj alanlarında yetiştirilen süs bitkilerinde Tomato spotted wilt virus (TSWV), Iris yellow spot virus (IYSV) ve Impatients necrotic spot virus (INSV) varlığının saptanması amacıyla, 2015 ile 2016 yılları arasında yürütülmüştür. Bölgede yapılan survey çalışmalarında simptomolojik olarak şüpheli süs bitkileri ile simptom göstermeyen süs bitkilerinden doku örnekleri alınmıştır. Toplanan 150 adet süs bitkisi örneği öncelikle Double Antibody Sandwich (DAS) ELISA yöntemi ile testlenmiş ve örneklerin hiçbirinde TSWV ve INSV infeksiyonu saptanmamıştır. ELISA testlerinde IYSV ile bulaşık olduğu saptanan örnekler RT-PCR çalışmalarında kullanılmıştır. IYSV-465c; IYSV-239f primer çifti ile yapılan RT-PCR çalışmalarında Pittosporum tobira ve Hydrangea macrophylla için 240 bp büyüklüğe sahip band gözlenmiştir. Böylece, IYSV’nin Adalar ilçesinde varlığı moleküler olarak da ortaya konulmuştur.

Lettuce (Lactuca sativa L.) is a member of the family Compositae (Asteraceae). This plant, which is grown for its leaves, is grown in open field and greenhouse conditions almost everywhere in the world and in Turkey. With the present study virus diseases of lettuce and their prevalence in Konya province was revealed for the first time. For this purpose, leaf samples were taken from lettuce plants showing virus diseases symptoms with surveys carried out in Konya province lettuce planting areas from May to August in 2020. Then the collected lettuce leaf samples were tested in laboratory conditions by Double Antibody Sandwich Enzyme-linked Immunosorbent Assay (DAS-ELISA) method to reveal infections of Cucumber mosaic virus (CMV), Miraflori lettuce big vein virus (MiLBVV), Lettuce mosaic virus (LMV), and Tomato spotted wilt virus (TSWV). According to the information obtained from the results of the study, it was determined that 40 out of 97 (41.23%) lettuce plant samples and all (6) weed samples were infected with at least one of the viruses. In lettuce leaf samples; TSWV (27.83%), LMV (12.37%), CMV (10.31%) and MiLBVV (5.15%) infections have been detected. In weed samples; infections of CMV (83.33%), LMV (66.66%), and TSWV (50%) have been revealed. The infections of TSWV, LMV, CMV, and MiLBVV on lettuce plants in Konya province were reported firstly with the study.

There are many viruses that infect pepper and limit its production. Among these viruses, Tomato spotted wilt tospovirus (TSWV) infects crops in 35 plant families that are economically important, including pepper. In the present study; totaly 156 leaf samples were collected, including 57 from pepper plants showing virus-like symptoms and 99 from weeds and/or plants other than peppers in and around the greenhouse, through surveys carried out in pepper greenhouses in Kumluca district of Antalya province, from September to December 2020. Then, the plant leaf samples were tested to determine TSWV infections by the Double Antibody Sandwich Enzyme-linked Immunosorbent Assay (DAS-ELISA) method. According to result of the tests, it was determined that 55.76% of the tested leaf samples were infected with TSWV, while this rate was determined as 96.49% for pepper samples and 32.32% for other plant samples. During the survey studies, it was revealed that the leaf samples of 13 out of 31 weed and different plant species except pepper were infected with at least one of the viruses. In addition, pepper plants showing symptoms TSWV-like symptoms in pepper greenhouses were counted during the survey, and the prevalence of this virus disease was calculated on the basis of Kumluca district and neighborhoods. As a result of these calculations, the prevalences of TSWV; for Kumluca, Mavikent, Beykonak, Salur, Hacıveliler, Adrasan, Merkez, and Kavakköy were determined as 26.93%, 26.92%, 32.27%, 20.66%, 21.13%, 17.66%, 13%, and 25%, respectively.

Al finalizar el ensayo, AV en T1 aportó cerca de 4 t MS/ha, 1668.3 kg C/ha, 78.7 kg N/ha y 11.0 kg P/ha, con beneficio económico-social de 9.2 ton choclo/ha. | This study provided knowledge about the agro-ecosystem N dynamics mediated by the use of agroecological practices such as GM. GM is established as legume its symbiotic action with soil rhizobia and arbuscular mycorrhiza formation, allows the cycling of nitrogen and phosphorus, among others. This study aimed at evaluating the influence of GM in the nitrogen dynamics of a Typic Haplustert located in the municipality of Candelaria (Colombia). In completely randomized blocks design with six replications, the GM coming from the intercropping Mucuna pruriens var utilis - Zea mays L. var. ICA 305 was established as T1 treatment and the native arvense Rottboellia cochinchinensis L. as T2, during the second half of year 2011. During the stage of preflowering of M. pruriens the content of organic C (OC) was evaluated as well as total N (TN), nitrate, ammonium, number of copies of amoA gene of ammoniaoxidizing bacteria, total porosity filled with water (TPW), temperature, flow of greenhouse gases: methane (CH4), carbon dioxide (CO2) and nitrous oxide (N2O), as well as the dry matter (DM) and the contents of C, N and P in plant tissues. Significantly higher concentrations (p <0.05) of CO, NT, ammonium and nitrate, were recorded in T2. The number of oxidizing bacteria of ammonium was significantly higher in T1 which coincided with the higher TPW and the lower soil temperature. The emission of atmospheric CO2 was significantly lower in T1, in contrast to the CH4 and N2O which scored the highest values. At the end of the trial, the GM in T1 provided about 4 t MS / ha, 1668.3 kg C / ha, 78.7 kg N / ha and 11.0 kg P / ha, with social economic benefit of 9.2 t corn/ha.
