Evaluation of Protein Profiles, Bioactivity, Allergenicity and Toxicity of Peptides Generated After in silico Digestion of Common Wheat and Einkorn Wheat

The belief that ancient wheat is more beneficial than common wheat has been growing trend in recent years among the consumers. The present study aims to compare bioactive peptide, allergen peptide and toxic peptide generation after gastrointestinal digestion of modern wheat (Triticum aestivum) and ancient wheat, einkorn (Triticum monococcum var. monococcum), using in silico tools. The primary sequences of both kinds of wheat were obtained from BIOPEP-UWM and UniProtKB/Swiss-Prot database. In silico digestion was applied using BIOPEP-UWM online tool. For the simulation of gastrointestinal digestion pepsin (pH 1.3) (EC 3.4.23.1), trypsin (EC 3.4.21.4), and chymotrypsin (EC 3.4.21.1) were selected and analyzed. Homology analysis was performed for each protein sequences using EMBOSS Needle program. Toxic and allergen peptides were predicted using ToxinPred online tool and Allergen FP v.1.0. The results showed that einkorn and common wheat proteins exhibited similar properties including high similarity rate (58.72-87.40%) indicating the percentage of matches between the two sequences and the identical bioactivities for peptides generated after digestion. Most of the bioactive peptides were dipeptides and the majority of them displayed more than one bioactivities including ACE inhibitory, DPP IV inhibitory or antioxidant activity, etc. Allergen peptides generated after in silico digestion were found to be similar for both kinds of wheat. In silico gastric digestion of einkorn and wheat caused toxic peptides production, but they were disappeared after in silico intestinal digestion. In conclusion, although there is a perception related to the Einkorn that is healthier than common wheat, in silico digestion of common wheat and einkorn did not support this perception.

Development of an efficient plant regeneration protocol is essential for vegetable crop advancement by biotechnological methods. In this study, regeneration protocols of four pepper lines of different pepper types were optimized. Different protocols for organogenesis were investigated in regeneration experiments. Optimum plant regeneration was obtained in different combinations of 6-Benzylaminopurine (BAP) and Indole-3-acetic acid (IAA) in organogenesis assays. In organogenesis experiment, the highest shoot development was determined as 80% from hypocotyl explant of Demre pepper in 4 mg L-1 BA+0.5 mg L-1 IAA, 80% from hypocotyl explant of Charleston pepper in 3 mg L-1 BA+0.5 mg L-1 IAA, 80% from hypocotyl explant of capia pepper in 5 mg L-1 BA+0.5 mg L-1 and 84% from hypocotyl explant of bell pepper in 5 mg L-1 BA+0.5 mg L-1 IAA. Afterward, shoots were rooted and whole plants were obtained.

Bu çalışmanın amacı, Akdeniz Üniversitesi yerleşkesi içerisinde yer alan birimlerde görev yapan akademik ve idari personellerin iç mekân bitki tercihlerinin, sahip oldukları iç mekân bitkilerine uyguladıkları bakım işlemlerinin belirlenmesidir. Bu kapsamda 382 adet akademik ve idari personellere ait ofis ziyaretinde bulunulmuş, ofis büyüklüleri lazer metre ile ölçülmüş, ofislerde bulunan iç mekân bitki türleri teşhis edilmiş ve birebir görüşmeler ile anket formu katılımcılara uygulanmıştır. Elde edilen sonuçlara göre Akdeniz Üniversitesi yerleşkesi içerisindeki örneklenen akademik ve idari personel ofislerinin büyüklüğü ortalama 19 m2 olup, sahip olunan iç mekân bitki ortalaması ise 2,3’dür. Katılımcıların gelir durumu, cinsiyetleri, görev yapmakta oldukları birimler (mesleki eğitimleri) ile ofislerde iç mekân bitkisi bulundurulması arasında istatistiksel açıdan anlamlı bir sonuç bulunmuştur. Orkide ve kaktüs türlerinin ofislerde en çok tercih edilen türler olduğu belirlenmiştir. Katılımcıların çoğu, sahip oldukları türlere rutin bakım işlemlerinden sulama ve kurumuş dal/yaprakların uzaklaştırılmasını gerçekleştirdiklerini belirtmişlerdir. Katılımcılar ofislerinde makul sayıda iç mekân bitkisi varlığından memnuniyet duyduklarını ifade etmiştir. Ofislerde çok sayıda iç mekân bitkisinin varlığı, katılımcılarda gerginlik ve yorgunluk duygularına neden olduğu belirlenmiştir. İşgücü verimliliğini arttırmanın ve kurumsal stratejik hedeflere ulaşmanın en ucuz ve en etkili yöntemlerinden biri olan iç mekân bitkilerini gelecekte bina tasarımına ve yönetimine dâhil etmenin zorunlu hale getirilmesi akılcı bir çözüm olacaktır.

During conventional fermentation of milk into yogurt and kefir, changes of antioxidant capacities (DPPH radical scavenging, ferrous chelating, reducing power, hydrogen peroxide/ H2O2 scavenging activity) and total phenolic content were investigated. Both products showed greater phenolic content, DPPH radical scavenging, and reducing power activities than was the case of their unfermented milk forms. But they were unable to scavenge H2O2. Chelating ability of milk increased by yogurt fermentation but decreased by kefir fermentation. In general, antioxidative capacity of milk improved throughout conventional yogurt and kefir fermentations and ripening periods (except chelating ability for kefir and H2O2 scavenging). This study is valuable since it evaluates the antioxidant power of yogurt and kefir produced by traditional fermentation with 5 different antioxidative approaches. In face of rapidly rising consumer demand for safe, healthy, functional but natural foods, revealed results might strengthen importance of conventional fermentation process and highlight the antioxidative contributions of mentioned products

This study aimed to reveal relationship between the extensiveness of trichophytosis on the body and serum zinc levels in cattle. This study was carried out on 92 cattle with trichophytosis and 50 healthy ones. Serum zinc levels of healthy cattle were evaluated as control. The cattle with trichophytosis separated into 5 groups according to diffusiveness of the diseases. Experimental groups of this study were designed as follows; Group 1: up to 1 cm, Group 2: 1-5 cm, Group 3: 5-10 cm, Group 4: 10-20 cm and Group 5: widespread on the body. Serum zinc levels measured by atomic absorption spectrophotometer equipped with flame system. In general, the mean level of Zn in trichophytosis groups (0.613±0.013 mg Zn/L) was statistically lower than healthy ones (0.900±0.017 mg Zn/L). A negative correlation was determined between the diffusiveness degree of trichophytosis and serum zinc levels (r = -0.772). Serum Zinc levels were statistically decreased from the control group to group 3. However, a stable course was observed from 3 to 5. Therefore, the value of group 3 (0.6 mgZn/L) was determined as a critical value for the cattle with trichophytosis. According to regression analysis results, it was observed that the extensiveness of the trichophytosis lesions affected the levels of serum Zn levels significantly (r2=59.6%). Consequently, serum zinc levels of cattle decreased in an inversely proportional manner to the diffusiveness degree of trichophytosis and stabilization was observed at the level of 0.6 mg Zn/L.

Dried apricot farms face many risks and uncertainties in Malatya, which is not only Turkey's but also the largest apricot production region in the world. Despite one of the major factors affecting farm efficiency, it was insufficient study on risk management in Malatya and even Turkey. The main purpose of the study was to measure the risk averse of dried apricot farms and to analyze of the factors affecting risk management strategies use of the dried apricot farms where an extensive production zone in Malatya. In the study, data collected from randomly selected 71 farms using questionnaires were used. The risk averse (attitude) coefficients of farmers were estimated by the “equally likely certainty equivalent” method. The factors affecting the risk management strategies choise were estimated using Multivariate Probit Model.

A survey and field experiment were conducted to evaluate the efficacy of commercial insecticides against tomato leaf miner [Tuta absoluta (Meyrick)(Lepidoptera: Gelechiidae)] under farmer’s field condition in Palpa, Nepal from February 2018 to April 2018. It consists of seven treatments replicated three times. The treatments included: Abamectin 0.15% EC (dose: 0.3ml/liter), Imidachloprid 17.8% SL (dose: 0.3ml/liter), Spinosad 45% SC (0.3ml/liter), Azadirachtin 300 ppm (5ml/liter), Chlorantraniliprole 18.5% SC (0.3ml/liter), Emamectin benzoate 5% SC (2gm/liter) and control. Treatments were applied two times at 12 days interval. From the farmer survey, Tuta absoluta was identified as the major production problem of the study area. The field experiment revealed that there was a significant effect of insecticides in larval mortality and damage reduction. The lowest percentage of leaves damage was obtained in spinosad followed by chlorantraniliprole and emamectin benzoate and the lowest fruits damage was obtained in chlorantraniliprole followed by spinosad and emamectin benzoate. Similarly, the lowest larval population in both leaves and fruits was observed in chlorantraniliprole followed by spinosad and emamectin benzoate. Average mining in infested leaves and fruits was found non-significant at all days of observation. The rate of larval population reduction over control was found highest in chlorantraniliprole followed by spinosad and emamectin benzoate. Thus, chlorantraniliprole, spinosad and emamectin benzoate were superior insecticides for management of Tuta absoluta in the field condition.

Erwinia amylovora, the causative agent of fire blight disease, threatens a lot of species of the Rosaceae family. Antibiotics and copper compounds in chemical applications are most frequently are applied, but these can be phytotoxic and cause resistant strains of the pathogen. In our experiments, 20 herbal materials were tested for their antimicrobial effectiveness against the fire blight pathogen in vitro and in planta. The air-dried plants ground into fine powder and extraction was performed at room temperature by maceration with 80% (v/v) methanol/distilled water. The minimum inhibitory concentration values were determined by using disc diffusion method and streptomycin was used as control in all experiments. Antimicrobial activity was evaluated by measuring the inhibition zones in reference to the pathogen. Among the tested plants, Szygium aromaticum, Thymus vulgaris and Rhus cararia showed a good antibacterial activity and they inhibited the growth of E. amylovora with inhibition zone diameter ranging from 21 to 27 mm at 20% (w/v) in absolute methanol compared to streptomycin (31 mm) in vitro conditions. In vivo tests were performed by using highly virulent E. amylovora isolate (Eak24b, 91%) grown on TSA medium and inoculation on young shoots of 3-year-old Gala variety of apple and Santa Maria variety of pear seedlings at 107 CFU ml-1 density of the pathogen. Disease severity (%) was assessed by by proportion of blighted shoot length to the whole shoot length and also efficacy of the extracts was determined by using Abbott formula. The highest efficacy was obtained by S. aromaticum and T. vulgaris extracts of reducing shoot blight of cv. Gala and cv. Santa Maria by 67.81% - 64-12% and 51.50% - 51.04% ratios, respectively. Obtaining results showed that some medicinal and aromatic plant extracts might be used against fire blight disease as potential new generation chemicals on pome fruits within integrated and organic control programs.

In this study, the effects of on 20 April, 5 May, 20 May, 5 June, 20 June, 5 July and 20 July sowing times application in Kahramanmaras and Sanlıurfa conditions on the quality criteria of Simal cowpea variety were researched. The experiment was established in 2015 as a randomized trial design with four replications. The pod length, number of grain of each pod, dry hay weight, moisture ratio, oil ratio, protein ratio and starch ratio characteristics of Simal variety (Vigna sinensis L.) were investigated. At the end of the research, it was noted that the number of grain of each pod, dry hay weight, the moisture ratio, the oil ratio, the protein ratio of cowpea, in terms of the locations, sowing times and locationxsowing time interactions were significantly different. It was determined that the length of pod is insignificant in terms of sowing time, and starch ratio was significantly showed differs in terms of location and location x sowing time interactions. The grain number of each pod, the weight of dry hay, the moisture ratio, the fat ratio, the protein ratio, starch ratio of cowpea were reported varieties from 7 to11.37 unit, from 361 to 1631 kg da, from 10.87 to 13.44%, from 1.28 to 2.11%, from 22.81 to 25.45%, from 42.11 to 49.77% respectively. As a result, it was observed that the characteristics of the cowpea investigated were affected by the environmental conditions.

Calendula officinalis (pot marigold), a medicinal ornamental plant belonging to the Asteraceae family, has various medicinal activities such as anticancer, antimicrobial, antioxidant and antitumor. The retention time of quercetin (6.445 min), kaempferol (11.246 min), caffeic acid (2.333 min), beta carotene (9.614 min) standards, which were planned to be analyzed by HPLC, was determined. The HPLC conditions that are most suitable for these metabolites have been determined. The amounts of quercetin, kaempferol, caffeic acid and beta carotene in the seedling leaves of these two Calendula species on the 0., 15., 30., 45., 60. days were compared. Caffeic acid and beta carotene have been detected in the leaves. However, quercetin and kaempferol compounds were not detected. It was determined that the caffeic acid and beta carotene amounts of C. officinalis species were higher than that of C. arvensis. The highest caffeic acid in both C. officinalis and C. arvensis seedlings (0.4042±0.0123 µg/g dry weight for C. officinalis, 0.1918±0.0437 µg / g dry weight for C. arvensis) and beta carotene amounts (0.8520±0.0692 µg/g dry weight for C. officinalis, 0.6389±0.0189 µg/g dry weight for C. arvensis) were determined on 60. day. In addition, it was determined that the amount of these metabolites differed according to the seedling development period.
