IDENTIFICATION OF SOURCES OF RESISTANCE AGAINST CHARCOAL ROT IN COWPEA

ABSTRACT Cowpea [Vigna unguiculata (L.) Walp.] is an important socioeconomic crop in Brazil, mainly in the Northeast and more recently in the Midwest of Brazil. Charcoal rot caused by Macrophomina phaseolina (Tassi) Goid, is an important disease in semiarid regions, where edaphoclimatic conditions are favorable to the development of disease. The aim of this study was to evaluate the response of 100 cowpea lines to two isolates of M. phaseolina. The experiments were conducted in a completely randomized design, with five replications (two plants per pot). The main variables evaluated were lesion length and relative growth compared to control (RGCC). Among the evaluated accessions, 15% of the lines were resistant to isolate 59 and 11% of the lines were resistant to isolate CMM 2106 of M. phaseolina. Therefore, these accessions can be used as a source of resistance to M. phaseolina by farmers directly as new cultivars or in future hybridizations of cowpea genetic breeding programs.

ABSTRACT Iron and zinc deficiency is one of the main problems affecting vulnerable populations in the Colombian Caribbean, thereby generating malnutrition from the consumption of foods with low content of essential minerals. The objective of this study was to evaluate the genotype-environment interaction for iron and zinc accumulation in grains in 10 cowpea bean genotypes by additive main effects and multiplicative interaction (AMMI) model and to select the most stable ones to stimulate their planting or as parents in the genetic improvement program. Nine promising lines and a commercial control were evaluated using the randomized complete block design with 10 treatments and four replications in 10 environments of the northern Colombia in the second semester of 2017 and first of 2018. The adaptability and stability analysis was done using AMMI model. The results showed highly significant differences at the level of environments, genotypes, and genotype-environment interaction for iron and zinc, demostrating a differential adaptability of genotypes in the test environments. Genotypes 2 and 3 expressed greater adaptability and stability for iron contents in the seed; while genotype 1, recorded it for zinc contents. These three genotypes outperformed the commercial control and, therefore, can be recommended for planting or be used as parents in the genetic improvement program.

ABSTRACT Fusarium wilt, caused by soil-borne Fusarium oxysporum f. sp. tracheiphilum (Fot), can reduce cowpea bean yield. Considering that genetic control through resistant genotypes is pivotal for Fusarium wilt control, the aim of the present study was to identify cowpea genotypes that are resistant to Fot from the Germplasm Bank of the Pernambuco Agronomic Institute, Embrapa Middle North, and other producing areas in the North and Northeast regions of Brazil. The cultivar BR-17 Gurguéia was used as a susceptible control and MNC01-649F-2-1 was used as a resistant control. Two experiments were carried out in a completely randomized design in the greenhouse of the Department of Agronomy of the University Federal Rural of Pernambuco. Plants with the first pair of expanded leaves were inoculated with a conidial suspension (106 conidia/mL) using the conidia root-immersion methodology. Fusarium wilt severity in plants was assessed 21 d after inoculation, with the aid of a descriptive scale. In the first experiment, we evaluated the resistance of 38 cowpea genotypes, of which 19 (5 moderately resistant and 14 resistant) were selected for the second experiment. In the first experiment, four genotypes (Canapu PE, Miranda IPA 207, Esperança, and BRS Pujante) did not show any symptoms of Fusarium wilt. In the second, 15 genotypes showed high resistance to Fot, including Canapu PE and Miranda IPA 207, which again had no symptoms of Fusarium wilt. In conclusion, this study identified that the genotypes, Canapu PE, Miranda IPA 207, Esperança, and BRS Pujante have greater resistance to Fusarium wilt.

ABSTRACT Given the lack of information on the use of spontaneous plants from the Caatinga biome as green manure to produce green grains, the present work aimed to agronomically and economically optimize cowpea production for green grains and their components when fertilized with equitable biomass amounts of hairy woodrose (Merremia aegyptia L.) and roostertree (Calotropis procera Ait.) in two cropping seasons in a semi-arid environment. The experiment used a randomized complete block design, with five treatments and five replicates. The treatments consisted of equitable amounts of dry M. aegyptia and C. procera biomass of 16, 29, 42, 55 and 68 t ha-1. In each experiment, an additional treatment was planted with cowpea without fertilizer (absolute control), and another was fertilized with mineral fertilizer for the purpose of comparison with the treatment of maximum physical or economic efficiency. The maximum optimized physical (agronomic) efficiencies of cowpea green pods and grain production were reached in the productivity of green pods at 3.90 t ha-1 and green grains at 4.06 t ha-1, with incorporation into the soil of 45.07 and 50.48 t ha-1 of green manure, respectively. The maximum economic efficiency of cowpea green grain production was reached at a net income of R$5826.12 ha-1 and at a rate of return of 1.29 reals for each real invested with application to the soil of 38.74 and 37.85 t ha-1 of the tested green manure biomass.

ABSTRACT Cowpeas (Vigna unguiculata L. Walp) are an economically and socially important legume in northern and north-eastern Brazil and can establish effective symbiosis with nitrogen-fixing bacteria. We evaluated the symbiotic compatibility and efficiency of rhizobial strains from Pernambuco semi-arid soils and determined their symbiotic stability on the IPA-206, BR 17-Gurguéia, and BRS Novaera cultivars, selected for different environments. The experiment was conducted in a greenhouse to evaluate a 3 × 28 factorial arrangement (cultivars selected for different environments × inoculation with the currently recommended strain, uninoculated plants with or without mineral nitrogen, and 25 rhizobial strains from semi-arid soils) in a randomized block design with four replicates. We determined nodule number, shoot and root dry matter, nodule dry matter by nodule number, nitrogen accumulated in the shoot by nodule dry matter, nitrogen content and accumulation in the shoot, relative efficiency of the recommended strain based on nitrogen accumulation, and shoot dry matter. Overall, the cultivars responded differently to different strains and cultivar biological nitrogen fixation potential. Strains G7.85 and BR 3262 showed potential for biological nitrogen fixation. BR 3262 was confirmed to be adequate for inoculation of different cowpea cultivars.

RESUMO São escassas as informações sobre o efeito de herbicidas sobre a qualidade sementes de feijão -caupi. Objetivou-se avaliar o efeito de dessecantes químicos na pré-colheita sobre a antecipação da colheita e qualidade fisiológica das sementes de feijão-caupi Cv BRS Tumucumaque, antes e após o armazenamento. No experimento 1, adotou-se o delineamento de blocos casualizados, com quatro repetições. Os tratamentos foram: sete herbicidas (carfentrazone-ethyl, saflufenacil, glyphosate, paraquat, flumioxazin, amonio -glufosinate e diquat) e um tratamento controle. Avaliou-se a antecipação de colheita, teor de água, germinação e vigor das sementes. As sementes foram armazenadas por seis meses em sala climatizada e ambiente não controlado e avaliadas quanto ao o teor de água, germinação e vigor, compondo o experimento 2, que foi disposto em esquema fatorial 7x2+1, com quatro repetições. O primeiro fator correspondeu a sete herbicidas, o segundo a dois ambientes de armazenamento, mais um tratamento controle. Observou -se um período de 3 a 9 dias de antecipação da colheita em relação a testemunha. O amônio-glufosinate afeta negativamente a germinação e o vigor das sementes. Após o armazenamento, menor germinação e vigor foram observados para o herbicida glyphosate. Os herbicidas carfentrazone-ethyl e flumioxazin não afetaram a qualidade fisiológica das sementes de feijão caupi nos períodos pós-colheita e seis meses após o armazenamento. O paraquat e o diquat foram os herbicidas mais eficientes na antecipação da colheita (nove dias). O glyphosate e o amonio -glufosinate foram os herbicidas mais prejudiciais à qualidade fisiológica das sementes. A sala climatizada é o ambiente mais adequado para conservação das sementes. | ABSTRACT Information about the effect of herbicides on cowpea seed quality is scarce. The objective of this work was to evaluate the effect of using chemical desiccants at pre-harvest on the anticipation of harvest and physiological quality of cowpea seeds of the cultivar BRS-Tumucumaque, before and after storage. Two experiments were conducted: experiment 1 was conducted under a randomized block design, with four replications; the treatments consisted of seven herbicides (carfentrazone-ethyl, saflufenacil, glyphosate, paraquat, flumioxazin, glufosinate-ammonium, and diquat) and a control treatment; anticipation of harvest, and seed water content, germination, and vigor were evaluated. The seeds were stored for six months in controlled environment and not-controlled environment rooms and evaluated for water content, germination, and vigor in Experiment 2, which was conducted in a 7^2+1 factorial arrangement, with four replications; the first factor consisted of the seven herbicides, the second factor was the two storage environments, plus a control treatment. The results showed 3 to 9 days of anticipation of harvest relative to the control. The glufosinate -ammonium herbicide negatively affected seed germination and vigor. The glyphosate herbicide decreased seed germination and vigor after storage. The physiological quality of cowpea seeds at the post -harvest period and after six months of storage was not affected by the carfentrazone-ethyl and flumioxazin herbicides. Paraquat and diquat were the most efficient herbicides for the anticipation of harvest (nine days). Glyphosate and glufosinate -ammonium were the most harmful herbicides to seed physiological quality. The controlled environment room was the most adequate environment for seed conservation.

RESUMO Genótipos podem responder aos ambientes de forma diferente; por isso, o estudo de adaptabilidade e estabilidade produtiva é importante para auxiliar melhorista na identificação e recomendação de cultivares. Assim, o objetivo deste trabalho foi inferir acerca da adaptabilidade e estabilidade produtiva de genótipos de feijão-caupi visando subsidiar a recomendação de cultivares para a produção de grãos verdes no estado do Ceará. Para isto, foram conduzidos cinco ensaios de valor de cultivo e uso (VCU) em localidades distintas pertencentes à duas regiões climáticas do estado do Ceará: Tropical quente semiárido brando (Acaraú, Pentecoste e Crato) e Tropical quente semiárido (Mauriti e Madalena). Foram avaliados 20 genótipos de feijão- caupi, sendo 12 linhagens elite e oito cultivares, em delineamento de blocos casualizados, com quatro repetições. Foram realizadas análises de variância e em seguida análises de adaptabilidade e estabilidade da produtividade de grãos verdes por meio da técnica multivariada GGE biplot. Os efeitos de genótipos, ambientes e interação G×E foram significativos, evidenciando respostas diferenciadas dos genótipos nas diferentes localidades. No aprofundamento das análises de adaptabilidade e estabilidade, os resultados do método GGE biplot revelaram que os dois primeiros componentes principais explicaram 72,17% da variação total, permitindo projeções bidimensionais fidedignas. A cidade de Crato foi indicada como local ideal para teste e as linhagens MNC05-847B-123 e MNC00-595F-27 se mostraram produtivas, adaptadas e estáveis, devendo serem recomendadas para produção de grãos verdes no estado do Ceará. | ABSTRACT Genotypes can respond differently to environments; thus, studies on adaptability and production stability are important to assist breeders in the identification and recommendation of cultivars. The objective of this work was to determine the adaptability and production stability of cowpea genotypes focused on subsidize recommendations of cultivars for green grain production in the state of Ceará, Brazil. Five assays were conducted in different locations in two climatic regions of the state of Ceará: a tropical mild hot semiarid region encompassing the municipalities of Acaraú, Pentecoste, and Crato, and a tropical hot semiarid region encompassing the municipalities of Mauriti and Madalena. Twenty cowpea genotypes-12 elite lines and 8 cultivars-were evaluated for cultivation value and use, using a randomized block design, with four replications. The results were subjected to analyses of variance and adaptability and green grain yield stability analyses by GGE biplot multivariate analysis. The effects of genotypes, environments, and G×E were significant, denoting different responses of genotypes in different locations. The results of the adaptability and stability analyses by the GGE biplot method showed that the two principal components explained 72.17% of the total variation, allowing reliable bidimensional projections. The municipality of Crato was the ideal location for tests and the lines MNC05-847B-123 and MNC00-595F-27 showed good production, adaptation, and stability, and can be recommended for green grain production in the state of Ceará, Brazil.

ABSTRACT Cowpea is a staple food in the Northeast of Brazil, thus, genetic improvement of this species is important. Samples of cowpea landraces were collected in the State of Rio Grande do Norte, Brazil, and the most promising ones were evaluated in the present study. The objective was to identify the most productive varieties and the characteristics that have greater direct effect on grain yield, for breeding purposes. Twelve landraces were evaluated using a randomized block design with five replications, in two experiments-one in the dry season, and other in the rainy season. However, water irrigation was used in both experiments, due to the almost total absence of rainfall in the rainy season. The Baraúna, and Carnaubais cowpea varieties were the most productive in both experiments. Campo Grande was the best variety found in the experiment carried out in the dry season. José da Penha, Lagoa de Pedra, Umarizal, and Upanema were the best varieties in the experiment carried out in the rainy season. The number of pods per plant was the characteristic that had greater direct effect on grain yield. | RESUMO O feijão-caupi é uma das principais culturas alimentícias do Nordeste brasileiro e, por essa razão, existe interesse em melhorá-la geneticamente. Foram coletadas variedades tradicionais dessa cultura no Estado do Rio Grande do Norte e as mais promissoras foram avaliadas no presente trabalho. Objetivou-se identificar as mais produtivas e distinguir, entre as características avaliadas, a que possui o maior efeito direto sobre o rendimento de grãos, para fins de melhoramento. Doze variedades foram avaliadas em blocos ao acaso com cinco repetições em dois experimentos. Um dos experimentos foi irrigado. O outro experimento foi realizado na época das chuvas, mas requereu irrigação devido à ausência quase que total de chuvas. As variedades Baraúna e Carnaubais foram as mais produtivas nos dois experimentos. No experimento realizado com irrigação foi destaque a variedade Campo Grande. Sob condições de sequeiro (com chuvas) destacaram-se também as variedades José da Penha, Lagoa de Pedra, Umarizal e Upanema. O número de vagens por planta foi o principal caráter a influenciar o rendimento de grãos.

ABSTRACT Callosobruchus maculatus, commonly known as cowpea weevil, is the main pest affecting stored cowpea Vigna unguiculata. The damage resulting from the penetration and feeding of cowpea weevil larvae within the grains or seeds of V. unguiculata leads to weight loss, reduction of germination power and nutritional value and commercial depreciation of this plant. The objective of this work was to evaluate the insecticidal activity and repellency of vegetable oils extracted from fennel, citronella, neem and 'moringa' against C. maculatus adults. To evaluate the insecticidal activity, we placed 20 adult insects in plastic containers lined with filter paper impregnated with different doses (0.0 x 106, 5.0 x 106, 1.0 x 105, 1.5 x 105, 2.0 x 105, 3.05, 4.0 x 105 and 5.0 x 105 L) of each vegetable oil. The number of insects was analyzed 24 hours later. To evaluate the repellency activity, we used an arena formed by five circular plastic containers with the central container interconnected symmetrically to the others by plastic tubes arranged diagonally. Samples of 30 g of cowpea were impregnated with different doses (5.0 x 106, 1.0 x 105, 1.5 x 105 and 2.0 x 105 L) of each vegetable oil. We released 50 adults of C. maculatus in the central container and counted the number of insects per container 24 hours later. Citronella and fennel oils at a dose of 3.0 x 105 L demonstrated a higher insecticidal effect on C. maculatus. However, citronella oil presented the best repellent action. | RESUMO Callosobruchus maculatus, conhecido como gorgulho do feijão caupi, é a principal praga que afeta o caupi armazenado Vigna unguiculata. Os danos resultantes da penetração e alimentação das larvas desse gorgulho nos grãos ou sementes do caupi levam à perda de peso, redução do poder de germinação e depreciação do valor nutricional e comercial desta planta. O objetivo deste trabalho foi avaliar a atividade inseticida e a repelência de óleos vegetais de erva-doce, citronela, nim e 'moringa' sobre adultos de C. maculatus. Para avaliar a atividade inseticida, foram colocados 20 insetos adultos em recipientes plásticos revestidos com papel filtro impregnado com doses diferentes (0,0 x 106, 5,0 x 106, 1,0 x 105, 1,5 x 105, 2,0 x 105, 3,0 x 105, 4,0 x 105 e 5,0 x 105 L) de cada óleo vegetal. O número de insetos foi analisado após 24 horas. Para avaliar a atividade repelente, utilizou-se uma arena formada por cinco recipientes circulares plásticos, sendo o recipiente central interligado simetricamente aos demais por tubos plásticos dispostos diagonalmente. Amostras de 30 g do feijão foram impregnadas com diferentes doses (5,0 x 106, 1,0 x 105, 1,5 x 105 e 2,0 x 105 L) de cada óleo vegetal. Liberaram-se 50 adultos de C. maculatus no recipiente central e contados o número de insetos por recipiente 24 horas após a liberação. Os óleos de citronela e erva-doce na dose de 3,0 x 105 L demonstraram maior efeito inseticida sobre C. maculatus. No entanto, o óleo de citronela apresentou a melhor ação repelente.

ABSTRACT Cowpea is one of the major food crops in Northeast Brazil, where it is commonly cultivated in the semi-arid regions with limited water availability. It is important to study the elicitors associated with cowpea to mitigate any deleterious effects of abiotic stress on the initial establishment of this crop. In this study, we aimed to evaluate the morphophysiological changes in cowpea cultivars under osmotic stress with seeds soaked in salicylic acid. The germination test was conducted in B.O.D germination chambers. The seeds of three cowpea cultivars: BRS Tumucumaque, BRS Aracê, and BRS Guariba, were germinated at five osmotic potentials (0.0; -0.2; -0.4; -0.6, and -0.8 MPa) after three pre-treatments: pre-soaking in deionized water, pre-soaking in salicylic acid, and without pre-soaking. The following parameters were evaluated: germination, germination speed index, seedling height, total phytomass, contents of chlorophyll ‘a’ and ‘b’, carotenoid content, electrolyte leakage, water content, and proline content. Our results indicate that salicylic acid promotes reduction in the harmful effects of abiotic stress, which is reflected in the increase in germination percentage, seedling height, and chlorophyll and carotenoid content as well as in the adjustment of electrolyte leakage and increase in proline content under induced water stress conditions. The cultivar BRS Guariba proved to be more tolerant to water deficit during germination and initial growth stages, when the seeds were treated with salicylic acid (1 mM). | RESUMO O feijão-caupi é uma das principais culturas alimentares no Nordeste do Brasil, onde é geralmente cultivada nas regiões semiáridas com pouca disponibilidade de água. É importante estudar os elicitores associados ao caupi para mitigar os efeitos deletérios do estresse abiótico no estabelecimento inicial desta cultura. Neste estudo, buscaram-se avaliar as alterações morfofisiológicas em cultivares de caupi sob estresse osmótico com sementes embebidas em ácido salicílico. O teste de germinação foi realizado em câmaras de germinação B.O.D. As sementes de três cultivares de caupi: BRS Tumucumaque, BRS Aracê e BRS Guariba foram germinadas em cinco potenciais osmóticos (0,0; -0,2; -0,4; -0,6 e -0,8 MPa) após três pré-tratamentos: pré-imersão em Água desionizada, pré-imersão em ácido salicílico e sem pré-imersão. Foram avaliados os seguintes parâmetros: germinação, índice de velocidade de germinação, altura da plântula, fitometria total, conteúdo de clorofila 'a' e 'b', conteúdo de carotenóides, vazamento eletrolítico, teor de água e conteúdo de prolina. Os resultados indicam que o ácido salicílico promove a redução dos efeitos nocivos do estresse abiótico, o que se reflete no aumento da porcentagem de germinação, altura da plântula e teor de clorofila e carotenóides, bem como no ajuste do vazamento eletrolítico e aumento do teor de prolina sob condições de estresse hídrico. A cultivar BRS Guariba mostrou-se mais tolerante ao déficit de água durante a germinação e estágios de crescimento inicial, quando as sementes foram tratadas com ácido salicílico (1 mM).
