Trifolium subterraneum L., sp. yanninicum cv. "Yarloop" is specially adapted to flooded lands during winter, so it is widely used in these types of soils as sheep pasture. Its estrogenic isoflavone content, which is considered to be high, is mainly biochanin A, genistein and formononetin and is of great interest in subclover breeding and selection. The chemical structure was considered in order to carry out the characterization and determination of these compounds using different instrumental techniques. The existence of strongly absorbent conjugated chromophore groups favour the presence of the characteristical maxima in UV-VIS region. Most of the heterocycles are not fluorescent, but when they are condensated with fluorogenic bencenic rings, they can produce potential fluorophores. The position and number of hydroxy and methoxy groups are the bases of chromatographic separation of these compounds in a HPLC system
