Recovery of rice tungro bacilliform virus (RTBV) from IR52 infected plants [Philippines]

Transmission of rice tungro bacilliform virus (RTBV) was dependent on rice tungro spherical virus (RTSV). Effects of RTSV on the recovery of RTBV from infected IR52 plants were examined. RTBV-infected IR52 plants at 45 days old were used as a source. The leafhoppers previously fed on RTSV-infected plants for 1 day were allowed a 2-day acquisition access while other leafhoppers were directly allowed a 2-day acquisition access to the source. Then the leafhoppers were given a 24-hour inoculation access on 6-day old TN1 or IR52 seedlings at one leafhopper per seedling in test tubes. All inoculated plants were tested for the presence of rice tungro associated virus by latex agglutination test. RTBV was recovered by the leafhoppers which acquired RTSV first and then fed on the RTBV source. However, leafhoppers which fed directly on the RTBV source did not recover RTBV. RTBV recovery was higher from susceptible TN1 than from IR52 indicating that tungro disease can spread in the field with leafhopper-resistant varieties if RTSV is present
