Using the in vitro colchicine treatment for the chromosome studies of the rainbow trout (Salmo gairdneri) and the grayling (Thymallus thymallus)

The purpose of this study was to apply the technique of chromosome preparation according to Ida et al. (1978) in the in vitro investigation of gill arches of freshwater fishes, namely rainbow trout (Salmo gairdneri) and grayling (Thymallus thymallus) with colchicine. The optimum time of shaking of gill filaments was found to be 5 hours in the rainbow trout investigated in the nutritive medium (Ringer's solution + + 0.005 colchicine) and 7 hours in the grayling. Hypotenization was performed with 0.005 NKCl within 3-4 hours and fixation lasted 80 minutes in Carnoy's fluid (the ratio being 2:1). Staining was performed with Giemsa stain. Using this technique the diploid chromosome number in 45 counted metaphases of rainbow trout was found to be 60 and in 50 metaphases of grayling 98. The present stage of this technique does not allow a thorough karyological analysis of the species with a high diploid chromosome number.
