Preliminary report on optimal incubation temperature for the isolation of Plesiomonas shigelloides using IBB and PDA medium

Overgrowth of Aeromonas spp. on the culture plates from the environmental sample for the isolation of plesiomonas has been a problem. Environmental strains of plesiomonas shigelloides and Aeromonas were used to test the optimal laboratory condition they required for growth. Two kinds of selective media such as Inositol bile-salt brilliant green (IBB) and plesiomonas differential agar (PDA) were tested against 37C, 42C and 44C as incubation temperature. It was observed that colony forming unit count of Plesiomonas sp. was not affected in all three temperatures tested while Aeromonas spp. considerably inhibited at 42C and 44C. Among the two media used, PDA was found more inhibitory in action to Aeromonas spp. at 42 and 44C. From our observation we found PDA medium if used and incubated at 44C is the optimal condition for Plesiomonas isolation and enumeration from the environmental samples. This will inhibit several other organisms present in the sample especially Aeromonas spp.
