Effect of sodium chloride and sodium sulfate on growth and metabolite content in shoot tip culture of Chinese cabbage (Brassica campestris L. ssp pekinensis Rupr)

This study was carried out to select salt tolerance lines during long term subcultures of Chinese cabbage (Brassica campestris L. ssp. pekinensis Rupr) shoot cultures on the media with NaCl and Na2SO4, and to investigate the physiological adaptation to salt by analyzing some metabolites of shoots induced during selection. Fresh weight in 0.5% and 1.0% of NaCl and 0.5% of Na2SO4 more increased, but shoot number much decreased, than the control. Protein content of shoots increased by NaCl but was not affected by Na2SO4 regardless of the number of subculture. The starch content of shoot increased in accordance with the number of subculture and increase of salt concentration in the media. When treated with 1.0% NaCl or 0.5% Na2SO4 the shoot content of alpha-amino nitrogen decreased in accordance with the number of subculture. Sucrose content decreased with the number of subculture but reducing sugar was not influenced. The content of protein and phenolics decreased by increasing salt concentration, chlorophyll content decreased in proportion to increasing salt. Selection for salt tolerance did not affect the subsequent growth on media with salt. Salts, especially NaCl, decreased water and osmotic potential of leaf. 0.5% NaCl or Na2SO4 delayed the rooting but increased root fresh weight. 1.0% NaCl decreased percentage of rooting and fresh weight
