Quantitative analysis of abscisic acid by enzyme-linked immunosorbent assay (ELISA) during protoplasts culture

Present study was conducted to evaluate the possible use of abscisic acid (ABA) content as a criterion to measure the degree of senescence of the protoplasts in culture. As an easy method for quantitative analysis of ABA, an enzyme-linked immunosorbent assay (ELISA) with detection range from 0.1 f mol to 10 p mol was developed. From the cross reactivity test with various ABA derivatives and their analogues, the ABA-antibody was found to have a high specificity with ABA. By this assay, the ABA contents in the protoplasts of barley, rye and their fusion products at various stages of culture were measured. From the results, it was found that the ABA contents increased as the senescence of the cells progressed, and decreased in the cells cultured on the media contaning polyamines. Therefore, it was strongly suggested that the ABA-ELISA could be effectiv as a simple method to measure the senescence of the cells in culture
