Study on the association between Anabaena azollae Strasburger and Azolla microphylla Kaulfuss during germination of megasporocarps

Experiments aimed to reunite the anabaena and anabaena-free azolla to reconstitute the symbiosis had been successfully conducted. The frequency of reconstitution was 9.6 +- 3.1% which was obtained from statistical analysis of 16 successful experiments. Artificially reconstituted symbiotic azolla was confirmed by direct observation under SEM and by monoclonal antibody test (McAB-C16). The key points to reestablished symbiosis between Anabaena and azolla were outlined. "Incomplete symbiosis" in some azolla was found to exist. Under the circumstance, only some anabaena filaments entered the leaf cavity while others stayed outside the leaf cavity. Incompletely symbiotic azolla had low nitrogen-fixing capacity (ARA) and failed to survive in N-free culture medium. It appears that the observation of this "incomplete symbiosis" opens a new vista to study further the azolla-anabaena symbiotic relationship. Present study improved the knowledge of the initial association between anabaena and azolla, particularly on the infection of Anabaena azollae onto emerging azolla sporeling. The results were useful for the control of anabaena infection in azolla breeding study.
