Effect of continuous light and dark treatment on tomato mosaic virus replicating capacity in different plant species [Egypt]

Excised tissue of mottling hosts of tomato mosaic virus (ToMV), Nicotiana tabacum var angustifolia and Lycopersicon esculentum Mill ev. Marmande, and necrotic local-lesion hosts, Nicotiana glutinosa, Datura stramonium and Datura metel had higher virus replicating capacity (VRC) when cultured in continuous light. Protein analysis of excised tissues and RNA determinations indicated a general decline which was more rapid in the dark than in the light. Virus infected leaves of all the above-mentioned hosts had higher ribonuclease (RNase) activity than healthy leaves regardless of light or drak. There was a good correlation between VRC found in plants and the level of RNase activity found after ToMV infection. Virus infection increased the level of peroxidase activity in all plant species tested. Light increased the level of RNase and peroxidase activity for both healthy and infected in all plant species tested. Under continuous light the lesions of N. glutinosa expanded more than in continuous dark. This enlargement in lesion size corresponded with the increase in virus content.
