Chlorophyll content and cell structure of soybean leaf as affected by soybean mosaic virus infection [Egypt]

The first trifoliate leaves of soybean seedlings infected through seeds, that showed severe yellow mosaic, were used in determination of chlorophyll content and in preparing tissue sections for ultrastructural study. For control purposes, similar leaves were obtained from soybean seedlings of healthy seeds. Electron micrographs showed aggregates of soybean mosaic virus (SMV) particles in the cytoplasm of infected cells, close to the plasmalemma, mitochondria or chloroplasts. In addition, few aggregates of virus particles inside some intact chloroplasts were observed. The mature of such particles was discussed. Chloroplasts with ruptured membranes having less grane and chloroplasts with large vesicles in the infected leaf cells, in addition to the significant reduction in chlorophyll content are the main reasons for the severe yellowing observed in the first trifoliate leaf systemically infected with SMV through seeds. Furthermore, the infected leaf cells showed degenerative effect summarized in the presence of myelin-like bodies and large irregular central vacuoles.
