Effect of varying growth conditions on the cellulase production of a new isolate of Penicillium funiculosum Thom

A new isolate of Penicillium funiculosum Thom, Isolate No. 171, isolated for high Beta-glucosidase activity was grown in corn cobs and rice straw with different organic additives such as polypeptone, rice bran, casein, Avicel and cellobiose at initial pH, 4.88-5.96 and at 28 degrees Celsius and 30 degrees Celsius. The effect of these growth factors on the production of the (CMCase, FPase and Beta-glucosidase) component enzymes of the cellulase complex varied. CMCase production was most sensitive to temperature, FPase to substrate and Beta-glucosidase to temperature, substrate and initial pH. Nevertheless, rice straw was a better substrate than corn cob, at initial pH of 6.0 and at 30 degrees Celsius. Yeast extract or polypeptone served as good organic N-sources compared with casein or rice bran. Rice bran decreased cellulase production. The concentration of Avicel used did not affect CMCase production of a more efficient FPase species and depressed Beta-glucosidase production. This suggests a level of regulation separate for each of the enzyme components. Nevertheless correlation analysis showed a strong correlation between FPase and CMCase production suggesting that at another level a common regulatory mechanism for the two components exists.
