Protoplast fusion as a means of producing new nucleo-cytoplasmic combinations in rice.

Results showed that higher protoplast yields, were obtained when carried out under light pre-isolation condition. Effective enzyme osmoticum concentration was found to be relative to the cultivar and species used. Viability of protoplasts isolated under light using 4% cellulase in mannitol was higher for both cultivars and species. Neutral Red stain was used to assess protoplast viability. PEG produced no significant effect on the viability of fused products but significantly increased the frequency of homoplasmic fusions. Detection of fusion products (homoplasmic and heteroplasmic) were facilitated using Neutral Red stain as marker. From the data gathered, it can be seen that non-conventional hybridization techniques to produce new nucleo-cytoplasmic combinations through protoplast fusion is feasible.
