Enzymatic detoxification of plantomycin in Xanthomonas campestris pv. oryzae

Plantomycin, an agrochemical has been used to control bacterial leaf blight (BLB) of rice caused by Xanthomonas campestris pv. oryzae in India. Development of resistance by this bacterium has led to the failure of disease control. Biochemical investigations were made to find out the mechanism of plantomycin resistance in the bacterium. Analysis of the macromolecular content of the resistant strain revealed the presence of an additional protein band in the resistant strain. However, the total protein was less in resistant strain than sensitive strain. Crude enzymes prepared from resistant strain effectively detoxified plantomycin as revealed by the enzyme assays. Plantomycin has the lambda maxima of 272 and 373. The detoxified substance had the ultra violet absorbance of 272 and 373, 340 and 390, 266 and 301 at 0, 6, 24 h, respectively. The effect of plantomycin concentrations, temperature, pH on the detoxifying enzyme was also investigated.
