Separation of photosystem 1, photosystem 2 and chlorophyll-antenna particles from the chloroplast of peas (Pisum sativum L.)

Chlorophyll-protein complexes of the preserved photosynthetic activity were determined in isolated chloroplasts of peas, cultivated in phytotronic chambers by PAGE - electrophoresis, using digitonin for the solubilization of photosynthetic membranes. By the methodological procedure described, it is possible to separate from peas the clearly characterized protein complexes of thylakoid chloroplast membranes. The obtained 3 main fractions are characterized by the percentage of total chlorophyll, the ratio of "a" and "b" chlorophyll and absorption spectra. Analysis of the percentage of total chlorophyll, the ratio of "a" and "b" chlorophyll and the absorption spectra indicate that fraction 1 represents photosystem 1, fraction 2 - photosystem 2 and fraction 3 - chlorophyllantenna particles.
