Variation in callus induction and organ differentiation abilities among sweet potato (Ipomoea batatas L.) genotypes cultured in vitro

The different sweet potato genotypes showed variable response for callus induction, regenerable callus production, growth characteristics, subculture and organ differentiation capacity. Of the 20 genotypes tested, 18 genotypes responded and had 47.91%-100% callus formation among explants. Callus cultures could be maintained for up to 12 months. Tissue cultures of V20-492, BNAs-51 and UPLSp-5 produced roots when regenerable calli from dark-grown callus tissue were subcultured on regeneration medium. Primary explants of 12 genotypes including VSp-4, V20-209, Kaimay, Jewel V20-429 and Kinabakab differentiated roots when inoculated on regeneration medium. Genotypes V20-492, Kabati, UPLSp-5 and BNAS-51 were most responsive to callus induction, regenerable callus formation and root differentiation. Shoot differentiation was not observed in any of the genotypes tested. It is, therefore, apparent that there is a wide range of response by sweet potato genotypes to in vitro culture conditions such that specific physical and chemical combinations have to be worked out for specific genotypes in order to achieve desired results.
