Micropropagation of Kalmia latifolia: acclimatation and rooting performance dependent on the preceding steps. Relationship with peroxidase activity

Kalmia latifolia was multiplied comparatively on ANDERSON's (AND) and LLOYD and McCOWN's (WPM) media with their respective or interchanged hormone composition. The highest rates of axillary proliferation were obtained on the AND media but the shoots vitrified. The shoots rooted poorly on full or half strength AND rooting media whatever their origin. Half strength WPM rooting media, with or without charcoal, gave the best results, but superior for shoots raised on WPM multiplication media. The number of roots per shoot was lower in the presence of charcoal but these roots were longer. The passage of the shoots from a multiplication to a rooting medium always resulted in an increase of peroxidase activity up to a more or less precocious, elevated and acute peak, all characteristics which were directly related to the precocity and degree of rooting. The performance of the plantlets at acclimatation (continued rooting, shoot growth and homogeneity) depended on the previously used rooting medium.
