Regeneration of adventitious shoots and embryos from root segments of in vitro cultured colt cherry (Prunus pseudocerasus x Prunus avium L.) plants

In order to develop an adventitious system for Colt cherry root segments several experiments were conducted to study the role of important parameters of culrure conditions, regeneration media and explant sources. One centimeter long root segments of in vitro produced roots were used as explants on Murashige and Skoog(MS) basic medium supplemented with either thidiazuron(TDZ) or benzyiaminopurine(BAP) at different concentrations. The two different cytokinins promoted the formation of completely different adventitious structures. Optimal concentrations for BAP and TDZ were 6.6M and 15M respectively. A dark incubation period of two weeks prior to exposing the cultures to a 16h photoperiod affected BAP and TDZ effect in different ways.
