Serological test studies of fungal disease with reference to Basidiobolomycosis

Serological tests are useful adjuncts to other means of diagnosing infections. There is little information on serological methods for the diagnosis of animal mycosis. An attempt was made to develop an agar gel precipitation test (AGPT) and enzyme-linked immunosorbent assay (ELISA) to investigate the immunoreactivity of antigens prepared from three isolates of Basidiobolus against antisera from immunized rabbits. In the AGPT, culture filtrate antigens (CFA) from pathogenic and non-pathogenic isolates and rabbit anti Basidiobolus sera were used. There were common antigens present in all the isolates tested and antigenic differences between pathogenic and saprophytic (non-pathogenic) strains were not detected. A CFA was used as the solid phase antigen to determine the optimal concentration of conjugate, serum and antigen in an ELISA for Basidiobolus. The ELISA and AGPT could be used as the methods for the diagnosis of basiodiobolomycosis in horses. This would necessitate the use of convalescent sera to determine the sensitivty and specificity of the test more accurately.
