Evaluation of buckwheat populations by RAPD markers

Genetic variability among 84 populations and cultivars of common buckwheat was evaluated using random decamer primers in PCR amplification of bulked DNA samples. 13 primers, previously chosen on the basic of the number and intensity of the fragments they produced, were used for amplification and yielded a total of 141 bands, among which 77 were polymorphic A NTSYS-pc computer program was employed for calculation of a simple matching coefficient of similarity. A moderate level of polymorphism and relatively close relationship among the studied common buckwheat populations were observed. The effectiveness of RAPD analysis of bulked DNA samples for the study of genetic variabilty in common buckwheat was demonstrated and this approach could be very useful for identification of cultivars and phylogenetic studies.
