Characterization and identification of yeasts

During the last two years the yeasts present in a vineyard and on its grapes were isolated and characterized by physiological and molecular genetic methods. First the collected yeasts were characterized by 25 different physiological tests. Yeasts with the same characteristics were controlled by PCR and karyotyping. The analyzed yeasts were compared with reference strains to determine the yeast species. The karyotyping results of Hanseniaspora species showed that the chromosomal patterns were comparable. But in the case of Candida the reference strains of the different species showed quite different patterns. Molecular genetical examination of the wine spoiling yeast Brettanomyces confirmed that the Brettanomyces strains analyzed belong to the species Dekkera bruxellensis. However, the reference strains of Dekkera bruxellensis and Dekkera anomala do not differ significantly in their chromosomal patterns. The results described show very clearly that molecular genetic characterizing methods give reproducible information on yeast species and strains. The main problem today is, however, that only a few reference strains are available
