Cultural behavior of protoplasts isolated from different organs in the genus Capsicum

The in vitro cultural behavior of protoplast from different organs of 31 genotypes in the genus Capsicum was investigated in order to find a regenerable one. An alginate disc embedding method was developed to facilitate cell division and colony formation by decreasing the production of deleterious phenolic compounds. Cotyledon protoplast of bell pepper varieties and hybrids, and hypocotyl protoplast of inbred lines not belonging to C. annuum responded best end most uniformly. In three genotypes, a chili type C. annuum (No. 40017) and two inbred lines (C. baccatum var. pendulum 25 and C. chacoense 8), rhizogenesis was observed. No shoots were regenerated in any genotype investigated. Thus the question of how to continue the cultivation of calli and how to regenerate entire plants in Capsicum is only partially answered
