Relationship between incubation period and serial transmission of scrapie in mice

Incubation period of sheep scrapie was studied in mice. The homogenate of 1st passaged mouse brain was inoculated intracerebrally in the 2nd passaged mice. At 3rd passage, the homogenate of 2nd passaged mouse brain was inoculated intracerebrally in one group and intraperitoneally in other group. At 4th passage, there were 6 groups which contained the mice given intracerebral inoculation of the brain, spleen, and salivary gland materials from two groups in 3rd passage, respectively. As the result, the incubation period was 213 days at 1st passage, 180 days at 2nd passage, and 112 days in the intracerebral inoculation group and 231 days in the intraperitoneal inoculation group at 3rd passage. At the 4th passage, the incubation period of the each group which was inoculated with the brain, spleen or salivary gland materials from intracerebral inoculation group in the 3rd passage was 130, 154, or 151 days. On the other hand, the incubation periods of the mice given the brain, spleen or salivary gland materials from intraperitoneal inoculation group in the 3rd passage was 131, 152, or 162 days, respectively. The incubation period of the mouse scrapie was shortened until 3rd passage, and it stabled at 110-130 days. Judging from the experimental transmission of the disease using the spleen and salivary gland materials, it was suggested that the prion protein (PrP) was present in these organs. By western blotting analysis, PrP was detected only in the brain. This might show that the organs other than the brain contained undetectable amount of PrP
