This study reports the clean-up procedure of food samples prior to HPLC minicolumns. An experiment was carried out to remove the interfering matrix from samples before injecting to the HPLC column. The peak of matric origin should be eliminated in order to quantify the analyte more accurately. Utilization of SPE in the sample pretreatment is useful not only for separating the solute being analyzed from the matrix compound, but also for concentrating the analyte in the extract. In this study, SPE technique using C18 minicolumns was used as sample preparation step for the deterination of oxytetracycline and tetracycline residues from frozen prawns, carbamate residues from rice and fat-soluble vitamins from yeast. The recoveries from fortified samples ranged from 65.4 to 97.8 percent
